环孢素A体外诱导HL—60细胞凋亡的研究 总被引:3,自引:1,他引:3
目的:观察环孢素A是否有诱导白血病细胞凋亡的作用。方法:应用细胞形态学检查、二苯胺法DNA片段化定量,DNA凝胶电泳及流式细胞术等方法观察细胞凋亡。结果:CsA50mg/L作用HL-60细胞4h,DNA片段率显著增高,达(28.2±5.8)%,而对照组仅为(12.5±1.7)%(P<0.01,n=10)。光镜及电镜检查见细胞核固缩,碎裂,有凋亡小体形成。DNA电泳显示典型的DNAladder。流式细胞术检测发现CsA50mg/L作用HL-60细胞6h凋亡细胞率为49.7%,而空白组仅为9.1%。CsA诱导HL-60细胞DNA片段化呈剂量和时间依赖性。结论:CsA在体外能诱导HL-60细胞凋亡。 相似文献

目的:探讨小檗胺(BER)诱导K562细胞凋亡及其可能的机制。方法:以流式细胞仪(FCM)分析、电镜观察细胞微结构变化及基因组DNA电泳等方法检测细胞凋亡。以RT-PCR及Westernblot方法检测K562细胞BCR/ABLmRNA和蛋白质水平表达。结果:FCM检测见到典型的凋亡峰,8.0mg/LBER作用24-72h,随药物作用时间延长,细胞凋亡率由(29.20±3.82)%上升至(61.77±4.35)%(P<0.01);以2.0-8.0mg/LBER作用24h,随药物浓度增加,K562细胞的凋亡率也增加。电镜下可见明确的细胞凋亡的形态学改变。DNA电泳呈现典型的梯形条带。16.0mg/LBER处理24h,K562细胞bcr/ablmRNA相对表达量及BCR/ABL融合蛋白质P210水平均明显低于对照组,分别为0.73±0.02vs1.19±0.02(P<0.01)和0.63±0.01vs1.04±0.02(P<0.01),呈时间-浓度依赖效应。经BER作用后,K562细胞bcr/ablmRNA表达强度与P210水平呈正相关(r=0.928,P<0.05),且细胞凋亡率与bcr/ablmRNA表达呈负相关(r=-0.997,P<0.01)。结论:在体外BER对K562细胞有明显的促凋亡作用,抑制bcr/ablmRNA表达及其蛋白质水平可能是其作用的重要机制之一。 相似文献

Summary A rate on autopsy of up to 30% systemic fungal infections and difficulties in diagnosing systemic mycosis antemortem have led to the empiric use of amphotericin B in patients with hematological malignancies, prolonged fever, and neutropenia. Routine empiric antifungal treatment was initiated in our institution in 1982. Amphotericin B was given to granulocytopenic patients with hematological malignancies with (a) unremitting fever after 48–72 h of antibiotic treatment, (b) recurrent fever during antibiotic treatment, or (c) with newly detected pulmonary infiltrates, sinusitis, skin and retinal lesions suggestive of a fungal infection. With this approach the rate of systemic fungal infections decreased significantly from 10% (27 of 270 patients; 1973–1981) to 4% (6 of 153 patients; 1982–1986,P<0.02). The reduction of systemic fungal infections was most prominent in patients with acute myelogenous leukemia, where its proportion decreased from 16% (16 of 98 patients; 1973–1981) to 4% (2 of 50 patients; 1982–1986,P<0.023). Our data support the hypothesis that the incidence of systemic fungal infections in patients with hematological malignancies and especially in acute myelogenous leukemia can be reduced significantly by empirical treatment with amphotericin B. 相似文献

Background
We conducted a monocentric retrospective study of patients with myelodysplastic syndromes (MDS) and autoimmune or inflammatory disorders (AIMs) and a literature review. We analyzed the association with subgroups of the WHO 2016 MDS classification and patient's survival in a case control study. Risk factors associated with survival were analyzed by uni- and multivariate analysis.Results
From all MDS patients 11% presented with AIMs. These were heterogeneous and the most frequent where polyarthritis (25%) and autoimmune cytopenias (17%). No difference for frequency and type of AIMs was observed for the WHO 2016 MDS subgroups (p?=?.3). In the case control study WHO classification, karyotype abnormalities, IPSS-R and IPSS were similar in both groups. The overall survival from MDS diagnosis was better in the group with AIMs [10.3?±?0.6 (IC95% 6.2–12.9) versus 4.8?±?1.1?years (IC95% 4.2–8.7), p?=?.04]. The better survival was restricted to MDS with low or intermediate-1 IPSS [11.1?±?1.5 (IC95% 9.9-NR) versus 8.7?±?1.3?years (IC95% 4.8–10.3), p?=?.006]. The better survival was only observed when AIMs diagnosis was timely associated or appeared after MDS diagnosis (p?=?.04). Factors associated with a better overall survival and survival without AML were steroid dependence [respectively HR?=?0.042, p?=?.003, (IC95% 0.005–0.33) and HR?=?0.07, p?=?.002, (IC95% 0.013–0.39)], a diagnosis of AIMs and MDS timely associated [respectively HR?=?0.05, p?=?.009, (IC95% 0.006–0.478) and HR?=?0.1, p?=?.008, (IC95% 0.018–0.54)] or a diagnosis of AIMs after MDS [respectively HR?=?0.024, p?=?.009, (IC95% 0.001–0.39) and HR?=?0.04, p?=?.008, (IC95% 0.003–0.43)].Conclusion
Autoimmune and inflammatory diseases associated to MDS are heterogeneous. AIMs diagnosed after or concomitantly to MDS seems associated with a better survival. Prospective studies are necessary to demonstrate that autoimmunity is associated to a better control of the MDS clone. 相似文献35.

Summary The question as to whether the serum concentration of erythropoietin is relatively high for the degree of anemia in patients with erythrocytic hypoplasia has regained interest, since recombinant human-like erythropoietin has become available as a drug for replacement therapy. We have compared the concentration of serum immunoreactive erythropoietin in nonrenal anemic patients with erythrocytic hypoplasia (22 cases) or active erythropoiesis (82 cases). In both groups a negative correlation was determined between the blood hemoglobin concentration and the logarithm of the erythropoietin concentration. However, the two regression lines were not identical, and the serum erythropoietin concentration was significantly higher for the degree of anemia in the patients with erythrocytic hypoplasia. Additional measurements in four patients suffering from acute leukemia with marrow failure showed that the erythropoietin concentration decreased towards the values observed in anemic patients with active erythropoiesis when the erythron recovered in the early phase of complete remission. These data support the idea that, independent of the O2 offer, the proliferating erythrocytic progenitors by negative feedback lower the blood level of erythropoietin.Abbreviations Epo

目的:试图用计算机设计并筛选出血管内皮生长因子(VEGF)的高效反义核酸;用实验方法研究VEGF反义核酸对K562细胞增殖的影响。方法:用RNAstructure(version3.7)软件,选择总自由能(overall△G37)相对低的反义核酸,共计7条,长度18-20核苷酸,全硫代修饰;细胞培养72h,采用台盼蓝拒染法观察存活细胞,用ELISA法检测培养液中VEGF蛋白水平,分析反义核酸对K562细胞的作用。结果:筛选出6条反义药物对K562细胞生长有明显抑制作用,均优于阳性对照组(X2)。与随机对照组(X1)相比,最优序列X7组细胞生长抑制率达31.9%、培养液中VEGF蛋白表达抑制率达51.4%,overall△G37与反义药物活性密切相关(r=0.887,P<0.01)。结论:计算机辅助设计有助于获得更好反义药物,VEGF反义药物可抑制K562细胞生长及VEGF蛋白表达,内源性VEGF蛋白具有促进K562细胞增殖的功能。 相似文献
