目的 建立两足动物鸸鹋的新股骨头坏死塌陷动物模型,提高塌陷的发生率和一致性,以期更好地进行骨坏死机制及治疗评价的研究.方法 采用成年鸸鹋20只,通过定制的治疗探针插入股骨头负重区,采用局部液氮冷冻和射频加热交替的方法,进行闭合回路的液氮冷冻损伤和射频加热损伤的交替,术后2、4、8、12、16周进行髋关节MRI检查,并在动物处死前进行下肢骨内微血管造影,标本进行Micro-CT扫描及组织学检查.结果 术后无明显感染征象,未发生实验动物异常死亡.髋关节MRI表现为:(1)术后4周广泛水肿;(2)术后8周异常信号逐渐局限;(3)术后12周开始出现股骨头塌陷.术后16周股骨头标本Micro-CT和组织学检查显示股骨头内血运受损,骨小梁骨折,空骨陷窝;塌陷侧股骨头的骨矿物量和骨小梁间距分别为(380.31±28.12) mg/cm3和(0.86±0.32) mm,明显低于对照侧的(415.75±41.28) mg/cm3和(1.17±0.17) mm(P均<0.05);骨体积分数和骨小梁厚度分别为(47.28±17.14)%和(506.17±220.58)μm,明显高于对照侧的(30.92±4.01)%和(325.50±44.53) μm(P均<0.05).术后16周骨内微血管造影显示,对照侧血管体积分数为(0.315±0.055)%,明显大于塌陷侧的(0.142±0.059)% (P <0.05).结论 采用局部冷热交替的方法成功建立鸸鹋塌陷性股骨头骨坏死动物模型,该模型具有较高的塌陷率,是股骨头骨坏死研究的良好模型,可用于骨坏死的塌陷机制研究和治疗评价.  相似文献

We investigated the effect of eel calcitonin (elcatonin) on the process of fracture repair in the osteotomized femur of cynomolgus monkeys, since they possess a Haversian remodeling system similar to that of humans. Alendronate was used for comparison. Twenty female cynomolgus monkeys (Macaca fascicularis), aged 18–22 years, were allocated into five groups: control (CNT, n = 4), low-dose elcatonin group (0.5 U/kg; ELL, n = 4), high-dose elcatonin group (5 U/kg; ELH, n = 4), low-dose alendronate group (10 μg/kg; ALL, n = 4) and high-dose alendronate group (100 μg/kg; ALH, n = 4). All animals were given subcutaneous injections twice a week for 3 weeks. Then fracture was produced surgically by transversely cutting the midshaft of the right femur and fixing with stainless steel plate. After fracture, treatments were continued until sacrifice at 26 weeks after surgery. The femora were assessed by micro CT, contact microradiograph, three-point bending mechanical test and histomorphometry. Micro CT showed that callus sizes in elcatonin-treated groups were similar to CNT, whereas alendronate-treated groups had larger calluses than those in the CNT and elcatonin-treated groups. Fracture lines almost disappeared in the CNT and elcatonin-treated groups but remained clear in the alendronate-treated groups. Total area did not differ significantly between the elcatonin-treated groups and the CNT but was significantly greater in the ALH compared to the CNT and elcatonin-treated groups, due to increased callus area in the ALH group. Callus remodeling was less suppressed in the elcatonin-treated groups than in the alendronate-treated groups when compared with callus remodeling in the CNT. Although no significant differences in structural mechanical properties such as ultimate load, stiffness and work to failure were found among all groups, ultimate stress was significantly reduced in the ALH group compared with CNT and ELL groups. In conclusion, mild suppression of callus remodeling by elcatonin did not impair overall fracture healing process. In contrast, alendronate delayed structural fracture healing process by strongly suppressing callus remodeling.  相似文献

Acute spinal cord injury (SCI) always leads to severe destruction of the microvascular networks. To investigate the three-dimensional (3D) alterations of microvasculature following SCI, we utilized an established rat SCI model. Based on the hypothesis that the spinal cord would undergo reorganization and postinjury modification of the vascular networks after SCI, we reconstructed the normal and injured angioarchitecture using micro-CT images of silicone rubber microsphere-perfused specimens. Several morphometric parameters were used to study the 3D vascular alterations in the SCI rat model, including the casting-based vessel volume fraction, connectivity density, separation, thickness and thickness distribution. Our results indicated that the microvascular spatial conformations were significantly different between the normal and injured spinal cord segments. The morphometric changes showed an increase of the vessel volume fraction and separation and a decrease of vessel connectivity density during the vascular healing process after SCI. Our results may contribute to elucidation of the mechanisms of compensatory vascular reconstitution in traumatized spinal cord. The method used here has the potential to improve our understanding of changes in the spatial architecture of vascular networks after SCI compared to the conventional histomorphology techniques. In summary, we developed a new methodology to analyze neurovascular pathology based on 3D vascular network patterns and features in an experimental rat SCI model. This technique could be used as a complementary tool to investigate the efficacy and side effects of therapeutic drugs or rehabilitation regimens.  相似文献

The study aimed to assess the accuracy and reproducibility of occlusal caries depth measurements obtained from different imaging modalities. The study comprised 21 human mandibular molar teeth with occlusal caries. Teeth were imaged using film, CCD, two different cone-beam computerized tomography (CBCT) units and a microcomputer tomography (micro-CT). Thereafter, each tooth was serially sectioned, and the section with the deepest carious lesion was scanned using a high-resolution scanner. Each image set was separately viewed by three oral radiologists. Images were viewed randomly, and each set was viewed twice. Lesion depth was measured on film images using a digital caliper, on CCD and CBCT images using built-in measurement tools, on micro-CT images using the Mimics software program, and on histological images using AxioVision Rel. 4.7. Intra- and inter-rater reliabilities were assessed according to the Bland/Altman method by calculating Intraclass Correlation Coefficients (ICCs). Mean/median values obtained with intraoral systems were lower than those obtained with 3-D and histological images for all observers and both readings. Intra-observer ICC values for all observers were highest for histology and micro-CT. In addition, intra-observer ICC values were higher for histology and CBCT than for histology and intra-oral methods. Inter-observer ICC values for first and second readings were high for all observers. No differences in repeatability were found between Accuitomo and Iluma CBCT images or between intra-oral film and CCD images. Micro-CT was found to be the best imaging method for the ex vivo measurement of occlusal caries depth. In addition, both CBCT units performed similarly and better than intra-oral modalities.  相似文献

Objectives

This study aims to assess the viability of using the acoustic emission (AE) measurement technique to detect and monitor in situ the interfacial debonding in resin composite restorations due to build-up of shrinkage stresses during polymerization of the composite.

Materials and methods

The non-destructive testing technique that measures acoustic emission (AE) was used to detect and monitor the interfacial debonding in resin composite during curing of the composite. Four groups of specimens, n = 4 each, were tested: (1) intact human molars with Class-I cavities restored with the composite Z100 (3M ESPE, USA); (2) intact human molars with Class-I cavities restored with the composite Filtek™ P90 (3M ESPE, USA); (3) ring samples prepared from the root of a single bovine tooth and ‘restored’ with Z100; (4) freestanding pea-size specimens of Z100 directly placed on the AE sensor. The restorations in Groups (1)-(3) were bonded to the tooth tissues with the adhesive Adper™ Scotchbond™ SE Self-Etch (3M ESPE, USA). The composites in all the specimens were cured with a blue light (3M ESPE, USA) for 40 s. The AE signals were recorded continuously for 10 min from the start of curing. Non-destructive 3D imaging was performed using X-ray micro-computed tomography (micro-CT) to examine the bonding condition at the tooth-restoration interface.

Results

The development of AE events followed roughly that of the shrinkage stress, which was determined separately by the cantilever beam method. The number of AE events in the real human tooth samples was more than that in the ring samples, and no AE events were detected in the pea-size specimens placed directly on the AE sensor. The number of AE events recorded in the specimens restored using Z100 was more than that found in specimens restored with Filtek P90. The micro-CT imaging results showed clear interfacial debondings in the tooth specimens restored with Z100 after curing, but no clear debonding was found in the P90 specimens.

Conclusions

The AE technique is an effective tool for detecting and monitoring in situ the interfacial debonding of composite restorations during curing. It can potentially be employed to evaluate the development of shrinkage stress and the quality of interfacial bonds in teeth restored with different composite materials, cavity geometries, and restorative techniques.  相似文献

Objectives

The present study evaluated the effects of casein phosphopeptide-amorphous calcium phosphate (CPP-ACP) and CPP-ACP with 900 ppm fluoride (CPP-ACPF) pastes on inhibition of enamel demineralization over time, using polychromatic micro-computed tomography (micro-CT).

Methods

Enamel blocks were prepared from bovine teeth. The specimens were each treated by one of the following agents, 30 min daily for 7 days: deionized water (negative control); CPP-ACP paste; CPP-ACPF paste; and NaF solutions (positive controls) (90, 900, and 9000 ppm F). After treatment, the specimens were immersed in a demineralizing solution (pH 4.5) for 24, 72, and 120 h. Mean mineral loss (ML) and lesion depth (LD) after each period were determined from mineral density profiles obtained using micro-CT.

Results

ML values in all the treatment groups were significantly smaller than those in the control group after 72 and 120 h of demineralization (p < 0.05, two-way ANOVA and t-test with Bonferroni correction). ML values in CPP-ACPF and NaF solution groups were significantly smaller compared to CPP-ACP group after 72 h (p < 0.05). LD values in the CPP-ACPF and all the NaF solutions groups were significantly smaller compared to the control group after 120 h (p < 0.05). The 9000 ppm F group showed the lowest nominal ML and LD values.

Conclusions

The application of CPP-ACP or CPP-ACPF pastes to sound enamel surfaces resulted in inhibition of enamel demineralization, and a better effect was noted for the latter paste. Quantitative assessment using polychromatic micro-CT demonstrated to be useful for detecting mineral density changes.  相似文献

Objective

The purpose of this study was to evaluate the different patterns of bone loss between the lumbar spine and the femur after ovariectomy in rats.

Methods

Twenty-four female Sprague-Dawley rats underwent a sham operation (the sham group) or bilateral ovariectomy (the ovariectomized group). Four and eight weeks after operation, six rats from each of the two groups were euthanized. Serum biochemical markers of bone turnover including osteocalcin and alkaline phosphatase (ALP), which are sensitive biochemical markers of bone formation, and the telopeptide fragment of type I collagen C-terminus (CTX), which is a sensitive biochemical marker of bone resorption, were analyzed. Bone histomorphometric parameters of the 4th lumbar vertebrae and femur were determined by micro-computed tomography.

Results

Ovariectomized rats were found to have higher osteocalcin, ALP and CTX levels than sham controls. Additionally, 8 weeks after ovariectomy in the OVX group, serum levels of osteocalcin, ALP and CTX were significantly higher than those of 4 weeks after ovariectomy. Bone loss after ovariectomy was more extensive in the 4th lumbar spine compared to the femur. Bone loss in the 4th lumbar spine was mainly caused by trabecular thinning, but in the femur, it was mainly caused by trabecular elimination.

Conclusion

The present study demonstrates different patterns of bone loss between the 4th lumbar spine and the femur in ovariectomized rats. Therefore, when considering animal models of osteoporosis, it is important that bone sites should be taken into account.  相似文献

High resolution tomographic images acquired with a digital X-ray detector are often degraded by the so called ring artifacts. In this paper, a detail analysis including the classification, detection and correction of these ring artifacts is presented. At first, a novel idea for classifying rings into two categories, namely type I and type II rings, is proposed based on their statistical characteristics. The defective detector elements and the dusty scintillator screens result in type I ring and the mis-calibrated detector elements lead to type II ring. Unlike conventional approaches, we emphasize here on the separate detection and correction schemes for each type of rings for their effective removal. For the detection of type I ring, the histogram of the responses of the detector elements is used and a modified fast image inpainting algorithm is adopted to correct the responses of the defective pixels. On the other hand, to detect the type II ring, first a simple filtering scheme is presented based on the fast Fourier transform (FFT) to smooth the sum curve derived form the type I ring corrected projection data. The difference between the sum curve and its smoothed version is then used to detect their positions. Then, to remove the constant bias suffered by the responses of the mis-calibrated detector elements with view angle, an estimated dc shift is subtracted from them. The performance of the proposed algorithm is evaluated using real micro-CT images and is compared with three recently reported algorithms. Simulation results demonstrate superior performance of the proposed technique as compared to the techniques reported in the literature.  相似文献

目的探讨去势法联合皮质激素快速诱导骨质疏松(OP)模型兔的可行性。材料与方法 40只8月龄雌性大白兔随机分为假手术组(A组,n=20)及去势+肌注甲强龙组(B组,n=20)。术后第2、4、8、10周行血清骨代谢标志物检测及离体L5椎体显微CT(Micro-CT)扫描。结果两组骨密度(BMD)在OVX术后第8周开始差异有统计学意义,而骨体积分数(BV/TV)、骨小梁连接密度(Conn.D)、骨小梁数目(Tb.N)、结构模型指数(SMI)、骨小梁间隙(Tb.Sp)及骨小梁厚度(Tb.Th)分别在术后4、8周开始差异有统计学意义。结论去势联合肌注甲强龙可在8周内快速建立OP兔模型,骨微结构参数变化早于BMD降低。  相似文献

X-ray computed tomography (CT) is a cardinal tool in clinical practice. It provides cross-sectional images within seconds. The recent introduction of clinical photon-counting CT allowed for an increase in spatial resolution by more than a factor of two resulting in a pixel size in the center of rotation of about 150 µm. This level of spatial resolution is in the order of dedicated preclinical micro-CT systems. However so far, the need for different dedicated clinical and preclinical systems often hinders the rapid translation of early research results to applications in men. This drawback might be overcome by ultra-high resolution (UHR) clinical photon-counting CT unifying preclinical and clinical research capabilities in a single machine. Herein, the prototype of a clinical UHR PCD CT (SOMATOM CounT, Siemens Healthineers, Forchheim, Germany) was used. The system comprises a conventional energy-integrating detector (EID) and a novel photon-counting detector (PCD). While the EID provides a pixel size of 0.6 mm in the centre of rotation, the PCD provides a pixel size of 0.25 mm. Additionally, it provides a quantification of photon energies by sorting them into up to four distinct energy bins. This acquisition of multi-energy data allows for a multitude of applications, e.g. pseudo-monochromatic imaging. In particular, we examine the relation between spatial resolution, image noise and administered radiation dose for a multitude of use-cases. These cases include ultra-high resolution and multi-energy acquisitions of mice administered with a prototype bismuth-based contrast agent (nanoPET Pharma, Berlin, Germany) as well as larger animals and actual patients. The clinical EID provides a spatial resolution of about 9 lp/cm (modulation transfer function at 10%, MTF10%) while UHR allows for the acquisition of images with up to 16 lp/cm allowing for the visualization of all relevant anatomical structures in preclinical and clinical specimen. The spectral capabilities of the system enable a variety of applications previously not available in preclinical research such as pseudo-monochromatic images. Clinical ultra-high resolution photon-counting CT has the potential to unify preclinical and clinical research on a single system enabling versatile imaging of specimens and individuals ranging from mice to man.  相似文献
