The geometry of the somata and dendritic trees of motoneurons innervating neck and shoulder muscles was investigated by using intracellular injections of HRP. In general, these motoneurons did not belong to a homogeneous population of motoneurons. Differences in average primary dendritic diameter, number of primary dendrites, and other measures of dendritic tree size were found between different neck and shoulder motoneuron groups. Several indices of proximal dendritic tree size (number of primary dendrites, sum of dendritic diameters, Rall's dendritic trunk parameter, and the sum of dendritic holes) were weakly correlated with the diameter or surface area of the soma. Some of these correlations depended on the muscle supplied by the motoneuron. The total combined dendritic length ranged from 66,660 to 95,390 microns. There was a weak, but positive, correlation between the diameter of primary dendrites and combined dendritic length. This relationship varied from motoneuron to motoneuron. The diameters of all dendrites of three trapezius motoneurons were examined in detail. The total dendritic surface area examined ranged from 415,000 to 488,100 microns 2 and represented approximately 99% of the total neuronal surface area. Last-order dendrites showed a high degree (39.9%) of taper. Dendritic tapering, by itself, was a major factor in the decrease of the (sum of dendritic diameters)3/2 measured at progressively distal sites from the soma. Although few parent and daughter dendrites obeyed the "three-halves law," the average exponent was 1.57. The diameters of primary dendrites and dendritic surface area were weakly correlated. The correlation between dendritic diameter and combined dendritic length or surface area improved if the weighted average of the diameter of second-order dendrites was used as a measure of dendrite size. Second-order dendrites, whose branches terminated in different regions of the spinal cord, showed different relationships between dendritic diameter and combined dendritic length or surface area. Comparisons between the motoneurons examined in the present study and motoneurons innervating other muscles indicate that, although all spinal motoneurons share several common features (e.g., long dendrites, dendritic tapering), each motoneuron group has a set of unique features (e.g., soma shape, relationship between primary dendrite diameter and dendritic surface area). Thus, the rules governing motoneuron dendritic geometry are not fixed but depend on the species of the motoneuron.  相似文献

We used the rapid Golgi and horseradish peroxidase (HRP) techniques to study the dendritic spread of relay neurons in functionally distinct laminae of the tree shrew dorsal lateral geniculate nucleus (LGNd). On the basis of their dendritic spread in relation to laminar and interlaminar zones, we describe three classes of relay neurons. Unilaminar neurons with multipolar radiate, bitufted, and intermediate types of dendrites. Dendrites of these neurons are confined to one lamina only, but also can have some of their segments in adjacent interlaminar zones. Multilaminar neurons with multipolar radiate, bitufted, and intermediate types of dendrites. Independent of the site of their cell bodies in a laminar or interlaminar zone, these neurons spread their dendrites over two or more laminae. Interlaminar neurons whose cell bodies and dendrites are confined to a single interlaminar zone. Unilaminar neurons are found in all the laminae. In the medial three laminae, they are more of the radiate type, whereas in laminae 4 and 5 their dendrites tend to be more of a tufted nature. Lamina 6 shows a preponderance of the elongated bitufted type. Multilaminar neurons, although less common as compared to the unilaminar, are also observed in all the laminae. Some neurons have their dendrites confined to an interlaminar zone. By retrograde transport of HRP injected into the visual cortex, we have shown that these neurons are, in fact, relay neurons. In addition to relay neurons, there are small interneurons with "axoniform" dendrites and an unmyelinated axon whose arborization is confined within the limits of the neuron's dendritic spread. Neurons of this type are not labeled with HRP injected into the visual cortex. We conclude that although each lamina is functionally specialized by input from ipsilateral or contralateral retina and by segregation of neurons responding to on or off stimuli, some multilaminar neurons can be found in each lamina. Thus, laminar as well as interlaminar zones contain a class of neurons that could provide a cross-talk between the functionally specialized laminae. Most relay neurons in all the laminae, however, confine their dendrites to their home lamina. Thus, the dendritic architecture of relay neurons allows for processing of information both within channels and between channels.  相似文献

Worldwide, the prevalence of allergies in young children, but also vitamin D deficiency during pregnancy and in newborns is rising. Vitamin D modulates the development and activity of the immune system and a low vitamin D status during pregnancy and in early life might be associated with an increased risk to develop an allergy during early childhood. This review studies the effects of vitamin D during gestation and early life, on allergy susceptibility in infants. The bioactive form of vitamin D, 1,25(OH)2D, inhibits maturation and results in immature dendritic cells that cause a decreased differentiation of naive T cells into effector T cells. Nevertheless, the development of regulatory T cells and the production of interleukin-10 was increased. Consequently, a more tolerogenic immune response developed against antigens. Secondly, binding of 1,25(OH)2D to epithelial cells induces the expression of tight junction proteins resulting in enhanced epithelial barrier function. Thirdly, 1,25(OH)2D increased the expression of anti-microbial peptides by epithelial cells that also promoted the defense mechanism against pathogens, by preventing an invasive penetration of pathogens. Immune intervention by vitamin D supplementation can mitigate the disease burden from asthma and allergy. In conclusion, our review indicates that a sufficient vitamin D status during gestation and early life can lower the susceptibility to develop an allergy in infants although there remains a need for more causal evidence.  相似文献

Interdigitating dendritic cell tumor (IDCT) is an extremely rare malignancy. It occurs primarily in lymph nodes, but extranodal involvement has also been reported. A 38-year-old woman with IDCT with breast and cervical lymph-node involvement is reported in this paper. To our knowledge, this is the first case of IDCT originating from the breast. In the breast and lymph node, the tumor displayed diffuse sheets, fascicles and storiform growth pattern. It was composed of oval to spindle cells with pale to eosinophilic cytoplasm, ill-defined cell outlines, oval nuclei with vesicular chromatin and prominent eosinophilic nucleoli. Mitotic activity was three per ten high-power fields. The neoplastic cells were intermingled with small mature lymphocytes and plasma cells. Immunohistochemical studies showed that the tumor cells were strongly and diffusely positive for vimentin, CD68, S-100 protein, CD45/leukocyte common antigen and fascin and focally positive for lysozyme, alpha-1 antitrypsin and CD4. Ki-67 labeling index was 10%. The patient was treated with combined therapeutic approaches, including surgery, radiotherapy and chemotherapy. IDCT has the potential for an aggressive clinical course. However, 32 months after the initial diagnosis, the patient is still alive and being followed with a stable tumor burden.  相似文献

Dendritic cells (DC) are key components of innate and adaptive immune responses. Plasmacytoid DC (PDC) are a specialized DC subset that produce high amounts of type I interferons in response to microbes. High mobility group box 1 protein (HMGB1) is an abundant nuclear protein, which acts as a potent pro-inflammatory factor when released extracellularly. We show that HMGB1 leaves the nucleus of maturing PDC following TLR9 activation, and that PDC express on the plasma membrane the best-characterized receptor for HMGB1, RAGE. Maturation and type I IFN secretion of PDC is hindered when the HMGB1/RAGE pathway is disrupted. These results reveal HMGB1 and RAGE as the first known autocrine loop modulating the maturation of PDC, and suggest that antagonists of HMGB1/RAGE might have therapeutic potential for the treatment of systemic human diseases.  相似文献

The migration and ultimate domain invasion of postmitotic lateral reticular nucleus (LRN) neurons were followed in embryonic day 15-20 (E15-E20) rat embryos, by using a horseradish peroxidase (HRP) in vitro axonal tracing method. All of the LRN axons elongate and neuronal somata migrate via the subpial or marginal migratory stream (mms), circumnavigating the ventrolateral aspect of the medulla at the glial endfeet level. They reach the ventral midline at E16, bypass it, and begin to settle in their final territory at E17. At E18 the LRN anlage is fully formed, and at E19-E20 its cells have finished their migration and are rapidly differentiating. Comparison of these sequential steps with their counterparts in the development of the inferior olive (ION) and external cuneatus (ECN) brings to light the essential role of the neuroepithelial cells of the interolivary commissure (the "floor plate"). This zone is likely to act as 1) a chemoattractant for the growth cones of the LRN, ION, and ECN, and 2) a decision-making center, which instructs the somata of these neurons to cross the midline or not, ultimately governing the crossed or uncrossed pattern of their projection to their common target, the cerebellum. Finally, the ontogeny of the LRN and ECN provides a most surprising example, even unique in the central nervous system, of long-distance, neurophilic migration that conveys neuronal cell bodies contralaterally to the side on which they proliferate.  相似文献

目的 :探讨细菌脂多糖 (LPS)对正常成人外周血单核细胞来源的树突状细胞 (Dendritic cell,DC)表面分子表达及免疫功能的影响。方法 :分离正常人 (n=5 )外周血单个核细胞 (PBMC)以 GM-CSF、IL-4联合诱生 DC,实验组在培养的第 2天加入LPS,正常对照组不加。于第 4、8天 FACS检测 DC表面分子 CD1a、CD14、CD83、CD80、HL A-DR的表达。同期检测 DC的增殖和刺激同种异体 T淋巴细胞的增殖情况。结果 :经 LPS刺激后 ,正常人 DC早期、晚期表面分子 CD1a、CD83、CD80表达增高 ,CD14表达降低 (P<0 .0 5 )。DC数量的增加及刺激同种异体 T细胞能力明显增强。结论 :GM-SF、IL-4和 L PS联合刺激能促进正常人 DC的成熟和免疫功能的提高  相似文献
