Neutrophil oxidative metabolism, C3d and beta 2 microglobulin levels, were assessed in nine consecutive patients undergoing cardiopulmonary bypass surgery with polypropylene hollow fiber oxygenators for open cardiac operations. Generation of oxygen free radicals by neutrophils was measured as luminol-enhanced chemiluminescence after stimulation with opsonized Zymosan and phorbol myristate acetate. A significant increase in light emission was detected by using both of the chemiluminescence stimulators. Moreover, a remarkable and significant increase in C3d levels was found already at 10 min. Conversely minimal changes in levels of beta 2 microglobulin were detected during cardiopulmonary bypass surgery. These data suggest that the impact of the patient blood with the foreign surface of cardiopulmonary bypass results in activation of phagocyte cells with increased potential in oxygen consumption. Previous work has reported that the 5-hydroxytryptamine (5-HT)1A agonist, 8-hydroxy 2-(di-n-propylamino)tetralin (8-OH DPAT), reduces ethanol intake by rats. However, as 8-OH DPAT reduces 5-HT neurotransmission, these findings are inconsistent with the proposed inhibitory role of central 5-HT neurons on ethanol intake. We examined the effect of 8-OH DPAT on ethanol, water and food intake in rats maintained on a limited access schedule using a lower dose range (6–250 µg/kg) and by assessing concomitant changes in behaviour. Low doses of 8-OH DPAT enhanced ethanol intake even when food and water were offered as alternatives. Suppression in ethanol intake was observed at higher doses where elements of the 5-HT syndrome were apparent. Similar observations were made in both fluid and non-fluid deprived water drinking rats, suggesting the latter effect is non-selective. Therefore 8-OH DPAT may both increase or decrease ethanol consumption in the rat depending on the dose used. 相似文献

Intestinal intraepithelial lymphocytes (iIEL) are primarily CD8 cells and most of them have a CD28? phenotype, the phenotype of effector cytotoxic T cells. We asked whether the predominance of CD8+ CD28? T cells in the gut may result from peripheral blood T cells preferentially migrating to the iIEL compartment and adhering to iEC. Compared with CD4 cells, adhesion of resting CD8+ T cells to iEC cell lines was significantly higher. Adhesion could be blocked with a MoAb to gp180, a molecule expressed on iEC which is known to interact with CD8/lck. No significant difference in the level of adhesion was observed between CD8+ CD28+ and CD8+ CD28? T cells. Thus CD8 cells may preferentially migrate to the iIEL compartment, but loss of CD28 expression could occur in situ after migration. Consistent with this hypothesis, the CD8+ CD28? cells became enriched after co-culturing T cells with iEC cell lines and primary iEC. Induction of the CD8+ CD28? phenotype in cord blood and adult T cells was observed in co-cultures with iEC and also with mitogens and superantigens. In the latter case, CD28 down-modulation was seen specifically in the Vβ subset targeted by the superantigen, indicating that loss of CD28 expression is a direct result of T cell receptor (TCR)-mediated stimulation. The combined results suggest that CD8+ CD28? T cells are antigen experienced T cells, and that they may have a survival advantage in the presence of gut epithelial cells in vitro. Background Exposure of the skin or respiratory tract to proteinases is frequently associated with allergic sensitization. This is of particular significance in the domestic indoor environment where the proteolytic activity of Der p I, the group I allergen of the house dust mite Dermatophagoides pteronyssinus, may influence the allergenicity of mites. Using class-specific proteinase inhibitors and active-site affinity chromatography, we have previously shown that Der p I exhibits a mixed cysteine-serine proteinase activity. Measurement of the amount of cleavage, however, did not determine whether the inhibitors used were targeting exactly the same proteolytic mechanism. Objective To resolve this issue, we have examined whether the cleavage specificity of the cysteine and serine proteinase activities of Der p I was the same. Methods HPLC and mass spectrometry were used to analyse and identify the products of a Der p I-digested peptide substrate and thus identify the peptide bonds cleaved. Results Der p I cleaves different peptide bonds, depending upon the class of proteolytic mechanism used. In the model peptide substrate insulin B chain, the cysteine and serine proteinase activities of Der p I showed preference for glutamic acid and arginine respectively in the P1 position. Conclusion These data suggest the existence of more than one mechanistic form of the allergen immunologically identified as Der p I. If proteolytic activity is indeed a function of allergenicity, this information may have important implications for the pathogenicity of Der p I and the ability of innate antiproteinase defences in the respiratory tract to prevent immune sensitization. 相似文献

Background: Fluctuations in the level of mite allergens in domestic house dust are the result of changes in the balance between synthesis, removal and decay. Purely physical forces as well as enzymatic degradation, mediated by house dust inhabiting microbes, may contribute to the decay of allergens in domestic dust. Knowledge about the speed of decay is essential for an understanding of the dynamics of allergen levels. Objective: The present study is a quantitative assessment of the speed of decay at nine combinations of temperature (15°C, 20°C and 25°C) and relative humidity (33%, 55% and 75%). Methods: Samples of mite infested material of an old rug were stored at these temperature/relative humidity-combinations for 6, 12 or 18 months, after the mites were killed by cither a freezing treatment or an acaricide (lindane). The microbes living in the rug presumably survive these treatments. Concentrations of Der p I and Der p II + Der f II. in extracts of the rug material, were measured by a radio immunoassay. Results: No significant changes in the levels of Der p I and Der p II +Der f II, could be detected even after 11/2 year at a high temperature and humidity. Conclusion: These findings incidate that mite allergens can be extremely stable under normal domestic circumstances. 相似文献

Gingival crevicular fluid (GCF) IL-8 and IL-1,1β levels were determined by sandwich enzyme-linked immunosorbent assays. Associations between IL-8 and IL-1β GCF levels, and between these cytokines and patient estrogen status were evaluated. IL-8 and IL-1β were detected more frequently and in higher amounts/30 s GCF sample in estrogen-deficient patients than in estrogensufficient patients. IL-8 and IL-1β GCF levels were significantly correlated. These lindings suggest that GCF IL-8 levels are associated with patient estrogen status and local IL-1β concentrations. 相似文献

目的 探讨毛喉萜 (Forskolin)的抗肝癌作用。方法 采用克隆形成、MTT比色法观察毛喉萜对SMMC 772 1细胞的作用 ,并利用免疫细胞化学方法检测其对rasp2 1、p5 3蛋白表达的影响。结果 毛喉萜可明显抑制SMMC 772 1细胞的增殖 ,其抑制率与剂量呈正相关 (P <0 .0 1 ) ,经毛喉萜处理后 ,rasp2 1、p5 3蛋白表达均有明显下降。结论 毛喉萜能明显抑制肝癌SMMC 772 1细胞增殖 ,其作用可能与降低rasp2 1、p5 3蛋白表达有关。 相似文献

Introduction. – L'objectif de cette étude est d'observer l'influence des paramètres anthropométriques sur la performance en aviron, chez des rameurs de niveau national et international.Synthèse des faits. – Les mesures anthropométriques ont été réalisées sur 88 rameurs (dont 11 évoluant au niveau international) lors des championnats de France de 2002. Ces mesures ont été corrélées aux performances réalisées sur ergomètre et en bateau. Des différences significatives sont mises en évidence entre les caractéristiques anthropométriques des rameurs de niveau national et international. Ces caractéristiques semblent être reliées directement aux performances ergométriques et non à celles réalisées en bateau.Conclusion. – L'influence des caractéristiques anthropométriques sur la performance en aviron pourrait être modifiée en fonction des niveaux de compétition.Aim. – Observe the influence of anthropometrics characteristics on rowing performance, at national and international level.Results. – Anthropometrics measurements of 88 rowers (included 11 international rowers) were realized during the national championships, data were correlated to ergometer and boat performances. Significant differences appear between anthropometrics characteristics of national and international rowers. Anthropometrics parameters seem related to ergometer performance but not to boat performances.Conclusion. – The influence of anthropometrics characteristics on rowing performance could be modified by the competitive level. 相似文献
