SC142-reactive antigen are highly glycosylated glycoproteins expressed on tissues of gastric and colon cancers but not on normal tissues. Murine SC142 antibody specific for the SC142-reactive antigen has been produced by immunisation with SNU16 stomach cancer cells. However, SC142 antibody has several potential problems such as high immunogenicity and poor tumour penetration owing to their large size. To improve tumour penetration potential in vivo, recombinant single-chain fragments have been produced using the original hybridoma cells as a source of variable heavy- and variable light-chain-encoding antibody genes. The use of the polymerase chain reaction, expression cloning technology and gene expression systems in E. coli has led to the production of SC142 single-chain fragments, which was similar in activity to the SC142 parent antibody confirmed by immunohistochemistry. Analysis by DNA sequencing, SDS-PAGE and Western blotting has demonstrated the integrity of the single-chain fragments. Competitive ELISA showed that SC142 single-chain fragments originated from parent SC142 antibody. BIAcore biosensor binding experiments showed that the SC142 single-chain fragments had an ideal dissociation rate constant as a tumour imaging reagent. SC1 is an extracellular matrix glycoprotein that is related to the multifunctional protein SPARC. These matricellular members play regulatory roles in modulating cellular interactions. SC1 expression is enriched in the central nervous system during embryonic and postnatal development as well as in the adult brain. In the rat cerebellum, SC1 is expressed at high levels in Bergmann glial cells and their radial fibers which project into the synaptic-rich molecular layer. At specific stages of development and in the adult, SC1 mRNA is selectively transported into cellular processes of these cells. In the present study, we have examined the effect of whole-body hyperthermia on the transport of SC1 mRNA in Bergmann glial cells of the rat cerebellum. Our results show that SC1 mRNA transport is diminished at 10 and 15 h post-hyperthermia, but returns to control levels by 24 h after heat shock. One of the characteristics of a heat shock on cells grown in tissue culture is a collapse of the cytoskeletal network. Intact components of the cytoskeleton are necessary for the transport of mRNA into peripheral processes of cells. However, in vivo hyperthermia does not appear to affect the morphology of the intermediate filament proteins GFAP, vimentin, or the beta-tubulin component of microtubules in Bergmann glial cell processes. During the hyperthermic time course, levels of vimentin protein increase, which is reflected by immunoreactivity of activated astrocytes and microvasculature in cerebellar white matter. 相似文献

目的探讨海鞘醇类似物002抗肿瘤活性。方法采用MTS法检测不同浓度的海鞘醇类似物对HepG2、A549和NIH3T3等细胞增殖的抑制作用;流式细胞、Annexin V-FITS/7-AAD等技术检测海鞘醇类似物对HepG2、A549和NIH3T3等细胞凋亡的影响。结果海鞘醇类似物对HepG2和A549的增殖有明显抑制作用,抑制率分别为73.67%和62.4%;海鞘醇类似物在浓度为2.0μg/ml时,引起HepG2和A549细胞的早期凋亡率分别为25.72%和25.46%。而对NIH3T3细胞影响不大。结论海鞘醇类似物对人肝癌和肺癌细胞的增殖有较强抑制作用,且能促进人肝癌和肺癌细胞的早期凋亡。 相似文献

Low density lipoproteins (LDL) isolated from poorly controlled diabetic patients are known to be taken up and degraded by fibroblasts at a lower rate than LDL isolated from normal subjects. This aberrant metabolic behavior has been attributed to a diabetic-related abnormality in LDL composition yet to be characterized. The studies reported in this article show that the decrease in uptake and intracellular degradation of LDL from diabetic patients is further enhanced when the cells are exposed to lipoprotein deficient serum (LPDS) isolated from the same poorly controlled diabetic patients. Comparative studies of the composition of LPDS obtained from normal donors and poorly controlled diabetic patients showed an increase in saturated and total unesterified fatty acids (UFA), lecithin, apolipoprotein A1, and immunoreactive insulin in the LPDS from diabetic patients. We postulate that exposure of cells to LPDS obtained from poorly controlled diabetic patients may induce changes in the composition of the fibroblast membrane and alter its fluidity, leading to further decrease in the uptake and degradation of LDL. During poor diabetic control, cell membrane changes, and modification of LDL composition are likely to act either additively or synergistically to induce an abnormal LDL-cell interaction. This abnormal interaction may be a relevant factor to explain the greater incidence of arteriosclerosis in diabetes mellitus. 相似文献

In rats anesthetized with urethane, effects of intravenously administered haloperidol (HAL; 0.1 mg/kg) and bicuculline (BCL; 0.2 mg/kg) were tested on inhibition of neuronal discharges of the substantia nigra, pars compacta (SNC), induced by single-shock stimulation of the sciatic nerve (SC). In two-thirds of the SNC neurons tested these two drugs antagonized the SC-induced inhibition, independent of the side of stimulation. In the remaining cases the effects of HAL and BCL were asymmetric. With HAL the inhibition from the contralateral SC was antagonized, whereas that from the ipsilateral SC was not antagonized or sometimes slightly potentiated. We have used the superior approach through the dome of the left atrium to repair cardiac lesions in the left atrium in 15 children ranging from 3 months to 17 years old. The single hospital death occurred in a 16-month-old infant with levo (L) transposition of the great arteries, Ebstein's malformation of the left atrioventricular valve, and ventricular septal defect. Exposure of the intraatrial structures was excellent, injury to adjacent cardiac structures did not occur, and hemorrhage from the left atrial suture line was not a problem. Though a few transient arrhythmias were seen, all survivors are in sinus rhythm without evidence of sick sinus syndrome, except 1 patient in whom atrial flutter associated with cardiomyopathy developed 1 year after operation. The excellent surgical exposure obtained of the interior of the left atrium and the absence of important complications related to the incision cause us to recommend this approach in children. 相似文献
