目的了解急性脑梗死患者血清胃泌素水平的变化及影响因素。方法应用RIA法测定95例急性脑梗死患者入院24h及1周时血清胃泌素水平,并与51名健康对照组比较。结果急性脑梗死组血清胃泌素水平明显升高,且胃泌素水平男女间无明显差异。继发胃肠症状组患者较无胃肠症状组患者胃泌素水平明显偏高,1周时无胃肠症状组胃泌素水平较入院24h有升高趋势。且脑干与脑叶梗死患者血清胃泌素水平升高显著。结论脑梗死组急性期血清胃泌素处于高水平,可基本排除单纯应激影响的结果,且胃泌素水平与梗死部位相关。此外脑梗死急性期并发的胃肠功能障碍与高胃泌素血症有一定的相关性。  相似文献

目的对时间分辨荧光免疫分析技术(TRFIA)检测乙型肝炎病毒(HBV)进行临床应用分析。方法用时间分辨荧光免疫分析技术TRFIA和放射免疫分析(RIA)同时检测261例乙型肝炎病毒感染者和150例正常人血清的HBV标志物,并对两种方法的检测结果HBsAg、HBsAb、HbeAg、HbeAb、HBcAb进行比较分析。同时对TRFIA法HBsg阳性而RIA阴性及TRFIA法HBcAb阴性而RIA阳性用化学发光免疫分析法(ECLIA)进行确诊检验。结果TRFIA法同RIA法比较,检测结果示二者之间差异无统计学意义(P>0.05)。结论TRFIA技术检测HBV标志物较RIA法的敏感度高,特异性强,可作为HBV感染的常规检测方法。  相似文献

This paper aimed to assess a method to measure eight thyroid-related compounds in serum by liquid chromatography-mass spectrometry (LC-MS/MS), to verify the correlation with radioimmunoassay (RIA), to evaluate the possible cross-reactivity, and to observe differences between athletes declaring the consumption of sodium levothyroxine and nonathletes serum samples. Validation was carried out to assess carryover, working range and linearity, limit of detection and limit of quantification, precision, matrix influence, recovery, accuracy, and uncertainty. Comparison between RIA and LC-MS/MS results was done. The assay was applied to serum samples, and comparison with RIA was done for T3 and T4 levels supported by RIA Thyroid-stimulating hormone (TSH) measurements. Validation parameters showed satisfactory results. Correlation between RIA and LC-MS/MS for T3 and T4 showed good results, but a cross-reactivity between T3 and T3AA was observed. Although no significant differences were proved, preliminary comparison between athletes and nonathletes serum samples showed a shift towards high values of TSH and lower for T4 values in the athletes' group. Differences between thyronine and T4AA concentrations and ratios were observed. The trend of T4 values supported by TSH measures might indicate subclinical hypothyroidism in athletes. This represents one of the most controversial thyroid statuses as different criteria about its treatment are described, especially since one of the exogenous causes is inadequate levothyroxine therapy. Because the variation of thyroid hormones and TSH has been extensively studied in high-performance sports, it is worth considering the need to set an adequate reference interval to accurately assess the thyroid status in athletes.  相似文献

Objective. The aim of this study was to develop and validate a high‐pressure liquid chromatography (HPLC) method for assessing vitamin D status as 25‐hydroxyvitamin D2 (S‐25OHD2) and 25‐hydroxyvitamin D3 (S‐25OHD3) in serum. Material and methods. We assessed the within‐ and between‐subject variation of vitamin D status in serum samples from four different dietary intervention studies in which subjects (n = 92) were supplemented with different doses of vitamin D3 (5–12?µg/day) and for different durations (4–20 months). Results. The HPLC method was applicable for 4.0–200?nmol S‐25OHD/L, while the within‐day and between‐days variations were 3.8?% and 5.7?%, respectively. There was a concentration‐dependent difference between results obtained by a commercial radioimmunoassay and results from the HPLC method of ?5 to 20?nmol 25OHD/L in the range 10–100?nmol 25OHD/L. The between‐subject variation estimated in each of the four human intervention studies did not differ significantly (p = 0.55). Hence, the pooled standard deviation was 15.3?nmol 25OHD3/L. In the studies with 6–8 samplings during 7–20 months of supplementation, the within‐subject variation was 3.9–7.2?nmol 25OHD3/L, while vitamin D status was in the range 47–120?nmol/L. Conclusions. The validated HPLC method was applied in samples from human intervention studies in which subjects were supplemented with vitamin D3. The estimated standard deviation between and within subjects is useful in the forthcoming decision on setting limits for optimal vitamin D status.  相似文献

Diethyl hexyl phthalate (DEHP) is an endocrine disruptor, it influences various organ systems in human beings and experimental animals. DEHP reduced the serum testosterone and increased the blood glucose, estradiol, T3 and T4 in rats. However, the effect of DEHP on insulin signaling and glucose oxidation in skeletal muscle is not known. Adult male albino rats were divided into four groups: Group I: Control; Groups II and III: DEHP treated (dissolved in olive oil at a dose of 10 and 100 mg/kg body weight, respectively, once daily through gastric intubation for 30 days); and Group IV: DEHP (100 mg/kg body weight) plus vitamins E (50 mg/kg body weight) and C (100 mg/kg body weight) dissolved in olive oil and distilled water, respectively, once daily through gastric intubation for 30 days. On completion of treatment, animals were euthanized and perfused (whole body); gastrocnemius muscle was dissected out and subjected to assessment of various parameters. DEHP treatment increased the H2O2, hydroxyl radical levels and lipid peroxidation which disrupt the membrane integrity and insulin receptor. DEHP impaired the insulin signal transduction, glucose uptake and oxidation through decreased expression of plasma membrane GLUT4, which may partly be responsible for the elevation of fasting blood glucose level. The present study suggests that DEHP exposure affects glucose oxidation in skeletal muscle and is mediated through enhanced lipid peroxidation, impaired insulin signaling and GLUT4 expression in plasma membrane. Antioxidant vitamins (C and E) have a protective role against the adverse effect of DEHP.  相似文献

目的:建立高特异性和高灵敏度的人胰岛素样生长因子Ⅱ(IGF-Ⅱ)放射免疫分析方法,用于测定人血清以及培养液中IGF-Ⅱ水平。方法:用人工合成的IGF-Ⅱ多肽连接甲状腺球蛋白,多次免疫兔子获取高效价、高特异的人IGF-Ⅱ抗体。用氯氨T法制备125I标记的IGF-Ⅱ,经交联葡聚糖凝胶(Sephadex G-25)纯化。抗原抗体反应采用平衡一步法,4℃温浴24h后经PR试剂分离结合和游离的标记抗原。用该法测定正常献血员150例和肝癌患者180例,并比较了肝癌患者介入治疗前后IGF-Ⅱ水平的变化。结果:该法测定范围为(0.1~24.3)ng/ml,最低检出值<0.1ng/ml,批内和批间误差<6.3%和10.0%。正常献血员血清IGF-Ⅱ水平(1.266±0.298)ng/ml明显低于肝癌各组患者(P<0.05);肝癌患者进行介入治疗3d后IGF-Ⅱ水平(4.263±1.411)ng/ml明显低于治疗前(4.916±1.24)ng/ml(P<0.05);介入治疗后3d与治疗后1个月(4.796±1.647)ng/ml水平相比无明显差异。结论:该法稳定、简便、特异、灵敏,适于检出人血清中的IGF-Ⅱ水平。  相似文献

目的:测定慢性心力衰竭(CHF)患者血清醛固酮(ALD)、睾酮(T)和雌二醇(E2)的水平,探讨其临床意义。方法:采用放射免疫分析186例CHF患者血清ALD、T和E2水平,并进行对照统计分析。结果:CHF组患者血清ALD显著地高于正常对照组(P<0.01),T和E2均显著地低于正常对照组(P<0.01,P<0.05),Ⅱ、Ⅲ、Ⅳ级心力衰竭组间血清ALD依次递增,T和E2依次递减,方差检验具有显著性差异(P<0.01);ALD与T和E2呈正相关(r=0.433、0.461,P<0.05),T与E2间无明显相关性(r=-0.131,P>0.05)。住院期间死亡组血清ALD显著地高于好转出院者(P<0.01),T和E2水平显著地低于好转出院者(P均<0.01)。结论:CHF患者血清ALD显著升高,T和E2显著下降,且与心功能分级和住院死亡有关。  相似文献

目的:探讨慢性阻塞性肺疾病(COPD)患者血清心钠素(ANP)、脑钠肽(BNP)、C型钠尿肽(CNP)水平的变化及其临床意义。方法:采用放射免疫分析79例COPD患者和36例健康对照组血清ANP、BNP和CNP水平,并进行统计分析。结果:COPD组血清ANP、BNP和CNP水平显著地高于健康对照组(t=3.6841,P〈0.01;t=11.70,P〈0.01;t=2.177,P〈0.05),但Ⅰ、Ⅱ、Ⅲ和Ⅳ级组间血清ANP、BNP和CNP水平方差检验无显著性意义(F=2.123、F=1.515、F=0.165,P均〉0.05)。相互间相关性分析揭示:ANP、BNP和CNP三者间均呈显著正相关(r=0.369,P〈0.01;r=0.354,P〈0.01;r=0.426,P〈0.01)。住院期间死亡的患者血清ANP、BNP和CNP水平显著地高于好转出院的患者(t=5.149,P〈0.01;t=4.875,P〈0.01;t=2.830,P〈0.01)。结论:COPD患者血清ANP、BNP和CNP显著升高,且与病人的稳定情况、肺动脉压力及预后相关。  相似文献
