The nonstructural protein 3 (NS3) of Dengue virus (DV) is a multifunctional enzyme carrying activities involved in viral RNA replication and capping: helicase, nucleoside 5'-triphosphatase (NTPase), and RNA 5'-triphosphatase (RTPase). Here, a 54-kDa C-terminal domain of NS3 (DeltaNS3) bearing all three activities was expressed as a recombinant protein. Structure-based sequence analysis in comparison with Hepatitis C virus (HCV) helicase indicates the presence of a HCV-helicase-like catalytic core domain in the N-terminal part of DeltaNS3, whereas the C-terminal part seems to be different. In this report, we show that the RTPase activity of DeltaNS3 is Mg2+-dependent as are both helicase and NTPase activities. Mutational analysis shows that the RTPase activity requires an intact NTPase/helicase Walker B motif in the helicase core, consistent with the fact that such motifs are involved in the coordination of Mg2+. The R513A substitution in the C-terminal domain of DeltaNS3 abrogates helicase activity and strongly diminishes RTPase activity, indicating that both activities are functionally coupled. DV RTPase seems to belong to a new class of Mg2+-dependent RTPases, which use the active center of the helicase/NTPase catalytic core in conjunction with elements in the C-terminal domain. 相似文献

目的:在原核表达系统中表达对凋亡神经元具有保护作用的重要蛋白Bcl-XL与蛋白质转导序列(PTD)的融合蛋白,并检测重组蛋白对重金属离子所诱导的细胞凋亡的保护作用。方法:通过RT-PCR的方法,用Bcl-XL特异引物从乳腺癌细胞系MCF-7细胞的总RNA中扩增出Bcl-XL基因,构建相应的原核表达载体,体外表达的TAT-Bcl-XL融合蛋白经镍亲和层析介质纯化后,通过免疫荧光方法检测其转导293T细胞的能力;并用流式细胞仪检测融合蛋白抑制细胞凋亡的能力。结果:经RT-PCR从MCF-7细胞总RNA中得到相应的TAT-Bcl-XL基因片段,并将其克隆入pCRT7/CT-TOPO载体中,重组质粒pTBTOPO转化大肠杆菌后,在SDS-PAGE和Western blot的结果中出现了与预期分子量相同的蛋白条带和阳性信号,纯化的TAT-Bcl-XL重组蛋白经免疫荧光检测,主要分布于细胞的细胞质中。流式细胞仪的检测结果显示融合表达蛋白可以有效地抑制Zn2+离子所诱导的细胞凋亡,使细胞的存活率提高40%。结论:TAT-Bcl-XL融合蛋白在大肠杆菌中获得高效表达,初步的功能性检测表明融合蛋白具有抑制细胞凋亡的功能。 相似文献

The major histocompatibility complex (MHC), a region of high gene density, contains a large number of genes relevant to the immune response, belonging to different multigenic families. We studied the genomic organization and polymorphism of a set of genes in the MHC class I region containing the tripartite motif (TRIM), consisting of a RING domain, B-box and coiled coil region, and a B30.2-like domain. A cluster of seven genes at 6p21.33 and two related family members telomeric of the cluster were characterized. All MHC-encoded TRIM-B30.2 genes showed moderate levels of polymorphism, affecting predominantly the RING and B-box domains. In terms of structure, the genes varied by the loss of partial and, in some cases, complete domains. They were strongly conserved in exons 2, 3 and 4, which form the coiled-coil region. The last exon, encoding the B30.2-like domain, is shared with the otherwise unrelated butyrophilin-like (BTN) genes, located 4.3 Mb telomeric of the TRIM-B30.2 cluster. Refractoriness to Gal N toxicity occurs especially in fetal rats, newborn rats, and in rats after partial hepatectomy. An injury however (laparotomy, incision on the back or ip BaSO4 suspension), prior to Gal N administration, also inhibits Gal N toxicity. In all these circumstances high levels of rat α2-macrofetoprotein (αMFP) occur. This protein is an acute phase reactant and is identical to rat α2-macroglobulin. αMFP isolated from the serum of injured rats and then administered to normal rats strongly inhibits Gal N toxicity. When time interval between the preceding injury, provoking αMFP production and Gal N administration shortens, the inhibiting effects are less and αMFP production remains low.During resistance to Gal N, the primary and secondary biochemical lesions of Gal N persist and the protecting effect of αMFP must be due to another mechanism, operating in later phases of cell injury. Very probably this is attributable to the stabilizing effect on membranes of hepatocytic organelles and the plasma membranes. In a study of 70 patients with asthma, rhinitis, and eczema, those giving a definite history of allergic reactions to egg more frequently showed positive skin tests to egg extracts (p = less than 0.003), the wheal diameters of which were significantly larger (p = less than 0.01) than in patients with only a possible or no such history. Patients with a definite history of egg allergy had significantly higher levels of specific IgE antibody against egg yolk, egg white, and allantoic fluid than patients in the other two groups (p = less than 0.005). Seven patients, all of whom had given a definite history of allergy to egg, were found to have positive skin prick tests to influenza vaccine, at the concentration used in medical practice. Two of these patients had previously been given influenza vaccine and both had developed adverse reactions. Of the 22 patients giving a definite history of allergy to egg, the 7 (35 per cent) with positive skin tests to influenza vaccine had significantly larger skin tests and higher levels of specific IgE antibody to the egg extracts than the group as a whole (p = less than 0.001). Allergic reactions to influenza vaccine are likely to occur in patients who have a definite history of allergy to egg and large skin prick test reactions or high levels of specific IgE antibody to egg extracts. Recent studies have identified genes that influence the length of the circadian period maintained by mice housed under constant lighting conditions. However, a less studied circadian activity variable is the amplitude of daily oscillations in locomotor activity. This parameter reflects spontaneous activity exhibited under standard lighting and housing conditions and, therefore, differs conceptually from assessments of exploratory or open-field activity, voluntary wheel-running, or circadian period during exposure to constant light or constant darkness conditions. We recently observed a greater daily amplitude of oscillation in spontaneous locomotor activity in C57BL/6 mice compared to BALB/cBy mice. To identify genetic loci with potential linkage to circadian variation in the amount of locomotor activity, we measured the spontaneous activity of 13 CXB recombinant inbred (RI) strains of mice. The probability density distributions of locomotor activity phenotypes for the 13 CXB RI strains were consistent with the presence of a low number of major quantitative trait loci affecting this trait. Regions of chromosomes 3, 8, 12, 13, and 19 showed provisional linkage to strain variation in locomotor activity. Probabilities of linkage were not sufficient for declaration of an activity-related quantitative trait locus but were sufficient to warrant further analysis either with additional RI strains or with F2 panels. 相似文献

Hepatitis B virions (Dane particles) were purified from the sera of chronic HBsAg carriers by consecutive rate-zonal and isopycnic centrifugations in sucrose gradients using HBsAg, HBcAg and endogenous DNA polymerase activities as specific markers. Purified Dane particles, radiolabelled with Na 125I by the chloramine-T procedure, had a higher buoyant density in CsCl (1.28 g/cm3) than unlabelled particles (1.26 g/cm3) and an estimated sedimentation coefficient of 280 s. 125I-Dane particles were fully precipitated by anti-HBs and not by anti-HBc sera. Heavy and light density core particles were purified from heavy and light density populations of Dane particles and radioiodinated. The iodinated polypeptides of Dane particles and HBcAg were compared with those of the iodinated 22-nm form of HBsAg by SDS-PAGE. Iodinated Dane particles contained seven polypeptides with molecular weights of 18,000, 23,000, 26,000, 34,000, 43,000, 48,000 and 115,000.
