AbstractProliferation is an important assessment in both clinical and research applications. Ki-67 has been shown to be a reliable immunohistochemical marker for actively dividing cells, but challenges exist for performing the staining on delicate or difficult tissues. Bone is one of these difficult tissues because the retrieval protocol required to detect the Ki-67 antigen often results in loss of tissue from the slide. We propose a new method for clear immunohistochemical labeling of rat tibial growth plate with Ki-67. This new method includes using thinner sections, decreasing sectioning artifact and increasing primary antibody incubation time. These changes, paired with optimal baking time of slides before staining, result in quantifiable nuclear labeling within the rat tibial and femoral growth plates. 相似文献

AbstractEighty-five subjects, recorded soon after eruption of second permanent molars, were followed longitudinally for 3 or 4 years.Correlation and multiple regression analyses were used to examine the relationship between the change in lower arch space condition and (1) the horizontal component of the change in position of various points on or near the anterior borders of the maxilla and mandible, (2) the differential horizontal-vertical change in these points and (3) differences in horizontal change between maxillary and mandibular, and between intramandibular points.No significant differences in amount or direction of growth were found between the maxillary and mandibular teeth or jaws. Pogonion changed more in a horizontal direction than B point. Increased lower arch crowding was weakly associated with differential horizontal growth of the teeth and jaws. ObjectiveThe present study was performed to standardize an effective protocol for micropropagation of Tigridia pavonia using tissue culture.MethodsThe explants were cultured on Murashige and Skoog (MS) medium supplemented with cytokinins like thidiazuron (TDZ), zeatin, kinetin and auxins such as indole-3-acetic acid (IAA), 1-naphthalene acetic acid (NAA) and indole-3-butyric acid (IBA), individually at different concentrations.ResultsMultiple shoots were obtained on MS medium containing either 2.0 mg/L TDZ or 2.0 mg/L IAA or 0.5 mg/L IBA and in the same medium for a long period (120 d) produced tiny bulbs at the base of the senescent leaves. TDZ favored only multiple shoots without roots, whereas IAA or IBA individually or in combination with TDZ produced rooted shoots. Shoots developed on MS medium supplemented with TDZ were rooted on MS medium containing either IBA or NAA at 0.5 mg/L. The plantlets were acclimatized in pots containing garden soil. Regenerated plantlets developed into normal plants. The plants showed 99% survival.ConclusionsThe highest number of bulblets obtained in the present study represents an effective alternative to the conventional method. 相似文献

Inflammation or pain during neonatal development can result in long-term structural and functional alterations of nociceptive pathways, ultimately altering pain perception in adulthood. We have developed a mouse model of neonatal colon irritation (NCI) to investigate the plasticity of pain processing within the viscerosensory system. Mouse pups received an intracolonic administration of 2% mustard oil (MO) on postnatal days 8 and 10. Distal colons were processed at subsequent timepoints for myeloperoxidase (MPO) activity and growth factor expression. Adult mice were assessed for visceral hypersensitivity by measuring the visceromotor response during colorectal distension. Dorsal root ganglion (DRG) neurons from adult mice were retrogradely labeled from the distal colon and calcium imaging was used to measure transient receptor potential vanilloid 1 (TRPV1) and ankyrin 1 (TRPA1) responses to acute application of capsaicin and MO, respectively. Despite the absence of inflammation (as indicated by MPO activity), neonatal exposure to intracolonic MO transiently maintained a higher expression level of growth factor messenger RNA (mRNA). Adult NCI mice displayed significant visceral hypersensitivity, as well as increased sensitivity to mechanical stimulation of the hindpaw, compared to control mice. The percentage of TRPA1-expressing colon afferents was significantly increased in NCI mice, however they displayed no increase in the percentage of TRPV1-immunopositive or capsaicin-sensitive colon DRG neurons. These results suggest that early neonatal colon injury results in a long-lasting visceral hypersensitivity, possibly driven by an early increase in growth factor expression and maintained by permanent changes in TRPA1 function. 相似文献

The purpose of this study was to investigate the feasibility of microvascular transplantation, based on the anterior tibial artery, of the epiphysis and the proximal half of the diaphysis of the fibula. India ink injections into the anterior tibial artery were performed in 13 unembalmed adult cadavers, following which transverse frozen sections were taken. Dye was observed in the fibula, both within the intramedullary cavity and at the level of the periosteum, in all specimens. The average distal limit reached by the dye, measured from the tip of the epiphysis, was 10.4cm in the intramedullary cavity and 11.8cm in the periosteum. Based on these anatomical findings one case of humeral reconstruction has been performed with a fibular transplant, including both the epiphysis and a segment of diaphysis, vascularized solely by the anterior tibial artery. 相似文献
