This study investigated the in vivo formation of engineering cartilage within living or devitalized cartilage discs using reflectance mode confocal microscopy and conventional light microscopy. Pig articular chondrocytes were suspended in fibrin glue and placed between two cartilage discs. Four experimental groups were prepared: in groups 1 and 2, the cell-hydrogel composite was placed between two live or between two devitalized cartilage discs, respectively; in groups 3 and 4, acellular fibrin glue was placed between two live or between two devitalized cartilage discs, respectively. Samples were implanted in the back of nude mice and analyzed after 2, 5, and 8 weeks. Results showed that engineered cartilage seems to grow more homogenously when the cell-seeded gel was placed between devitalized cartilages than when it was placed between live cartilage matrices. Confocal microscopy provides valuable information on the integration of tissue-engineered cartilage with native tissue and could be useful for nondestructive imaging in vivo. 相似文献

背景:生物医药产业是21世纪全球公认的最具有发展前景的国际化高技术产业之一,也是世界贸易增长最快的朝阳产业之一。目的:分析北京生物医药产业的发展现状,指出北京生物医药产业跨越式发展中存在的问题及相应的解决对策。方法:应用计算机检索百度文库、中国知网及手工检索学校图书馆内书籍2005至2014年间的文献,分析了北京生物医药产业目前的发展现状和存在的问题,并针对这些问题提出对北京生物医药企业实现跨越式发展有指导意义的观点与结论。结果与结论:近年来,北京生物医药产业发展迅速并取得了良好成果,但在实施跨越式发展过程中也存在着诸多问题,如产业基础较薄弱、企业创新主体地位还未显现、缺乏足够的资金投入等。针对这些问题,提出了不断优化调整产业结构、进一步优化所有制结构、加强技术创新、建设以企业为主体的研发体系、实施品牌扩展战略5方面对策,实现北京生物医药产业由战略产业向主导产业的跨越式发展,成为世界重要的研发中心、高端制造基地和市场中心。 相似文献

Small instestinal submucosa (SIS) is an easily produced material that has been used experimentally for tissue engineering. To evaluate the ability of SIS to facilitate bone growth within a long-bone defect, a segment of the radius was surgically removed in adult, female Sprague-Dawley rats. The defect was either left unfilled or implanted with SIS, demineralized cortical bone (DMCB), or ovalbumin. The defect was evaluated radiographically and histologically after 3, 6, 12, and 24 weeks. Tissue remodeling within the defect was evident by week 3 in SIS- and DMCB-treated rats. Filling was characterized initially by infiltration of mononuclear cells and extracellular material in SIS-implanted rats and multifocal remodeling bone particles and cartilage formation in DMCB implanted rats. Cartilage was observed as early as 3 weeks and bone as early as 6 weeks in SIS-implanted rats. Filling of the defect arose from multiple foci in DMCB-implanted rats, but was contiguous with and parallel to the ulnar shaft in SIS-implanted rats, suggesting that defect repair by SIS may be conductive rather than inductive. Rats in which the defect was left unfilled demonstrated slow but progressive filling of the defect, characterized by mononuclear cell infiltrates and fibrous extracellular material. In summary, SIS facilitated rapid filling of a longbone defect. These results suggest that SIS may be useful as a bone repair material. 相似文献

BackgroundBetter midsole materials and comfort have been incorporated into more expensive shoes and are popular with runners. Consequently, knockoff running shoes are currently widely distributed in the Chinese market and and cost only 30%–50% of the total price of genuine branded products.Research questionUncertainty exists concerning the beneficial effects of advanced shoe material application in decreasing foot loading or impact force during running. Additionally, using comfort as a criterion to identify genuine branded running shoes may exclude brand factor.MethodsFifteen healthy male volunteers were asked to perform two different tests, including running and a comfort evaluation. Each participant was asked to identify which footwear was the Adidas brand shoe based on their perception of comfort.ResultsTime to the first peak of the vertical ground reaction force occurred significantly later when subjects wore the genuine branded shoe compared to knockoff shoe 1 (p = 0.003) and knockoff shoe 2 (p = 0.015) footwea. The genuine branded shoe (p = 0.005) and knockoff shoe 1 (p = 0.029) were significantly more comfortable compared to the knockoff shoe 2. Only four subjects selected the genuine branded shoe, whereas six subjects selected both the genuine branded shoe and knockoff shoe 1.SignificanceKnockoff running footwear significantly increases impact loading compared to the genuine branded product, thereby posing greater risk of running injury. 相似文献

BackgroundDengue fever (DF) is the most rapidly spreading mosquito-borne viral disease. Practical vaccines or specific therapeutics are still expected. Environmental factors and genetic factors affect the susceptibility of Dengue virus (DV) infection. Asthma is a common allergic disease, with house dust mites (HDMs) being the most important allergens. Asthmatic patients are susceptible to several microorganism infections.MethodsA nationwide population-based cohort analysis was designed to assess whether to determine whether asthma can be a risk factor for DF.ResultsUnexpectedly, our data from a nationwide population-based cohort revealed asthmatic patients are at a decreased risk of DF. Compared to patients without asthma, the hazard ratio (HR) for DF in patients with asthma was 0.166 (95% CI: 0.118–0.233) after adjustment for possible confounding factors. In the age stratification, the adjusted HR for DF in young adult patients with asthma was 0.063. Dendritic cell-specific intercellular adhesion molecule 3-grabbing non-integrin (DC-SIGN) of dendritic cells (DCs) is an important entry for DV. Through another in vitro experiment, we found that HDM can diminish surface expression of DC-SIGN in monocyte-derived DCs and further decrease the cellular entry of DV.ConclusionsDecreased DC-SIGN expression in DCs of allergic asthmatic patient may be one of many factors for them to be protected against DF. This could implicate the potential for DC-SIGN modulation as a candidate target for designing therapeutic strategies for DF. 相似文献

BACKGROUND:Mouse pluripotent stem cells are induced to differentiate into insulin-secreting cells that can effectively improve blood glucose levels in diabetic mice.
OBJECTIVE:To detect mRNA and protein levels of insulin-like cell clusters from induced pluripotent stem cells and to investigate the function of insulin-secreting cells in vitro and in vivo.
METHODS:Mouse induced pluripotent stem cells cultured in vitro were induced to differentiate into insulin-secreting cells using combined inducers through three stages. The morphology of endodermal cells, islet-derived progenitor cells and mature islet cells in each stage was observed and relative gene expression levels were detected by PCR. Mature insulin-like cell clusters underwent dithizone staining and functions of insulin released in vitro were observed by ELISA assay. Finally, the insulin-secreting cells were transplanted into the subrenal capsule of diabetic mice, and then blood glucose levels were observed.
RESULTS AND CONCLUSION:The mature spherical insulin-like cell clusters were successfully obtained in vitro, which were in iron red by dithizone staining, and expression of insulin mRNA was determined by PCR. The insulin-like cell clusters could secrete insulin in response to various blood glucose levels by ELISA assay. In addition, after the cells clusters were transplanted into the subrenal capsule of mice with type 1 diabetes, the blood glucose levels were marbedly improved. 相似文献

目的 研究抗Pgp 抗CD3双特异双链抗体介导的特异性靶向杀伤活性。方法 采用亲和层析法分离纯化本室构建的抗Pgp 抗CD3双特异双链抗体可溶性表达产物 ,并用SDS PAGE、Westernblot及抗活细胞间接免疫荧光法鉴定纯化产物 ;采用51 Cr释放试验测定其介导的体外靶向杀伤活性。结果 纯化的抗Pgp 抗CD3双特异双链抗体具有与Jurkat细胞 (CD3 )和K5 6 2 A0 2 (Pgp )细胞结合的活性 ;抗Pgp 抗CD3双特异双链抗体具有介导激活的T淋巴细胞杀伤Pgp表达阳性的耐药肿瘤细胞的作用 ,杀伤作用的强弱显示出效靶比和剂量依赖关系 ,且可被CD3ScFv或PgpScFv特异性的阻断。结论 抗Pgp 抗CD3微型双功能抗体有望成为治疗耐药肿瘤 ,特别是肿瘤残留灶和微小转移灶的治疗的一种新策略。 相似文献
