目的:观察中波紫外线(UVB)辐射后人原代角质形成细胞中环丁烷嘧啶二聚体(cycl obut ane pyrimidine dimmer s,CPDs)的生成和清除情况,以及没食子儿茶素没食子酸酯(epigall ocatechingall ate,EGCG)干预对上述过程的影响。方法:以不同剂量UVB照射原代角质形成细胞后并加入EGCG干预处理,采用免疫组织化学法检测UVB照射后不同时段细胞中CPDs的产生和清除情况。结果:UVB照射后细胞中CPDs开始产生,1h左右达到高峰;CPDs在照光后4h内清除速率较快,4h后清除速率逐渐降低,至24hCPDs被基本清除。照光前后加入EGCG干预减少UVB引起的35.7%~42.9%CPDs产生(P<0.05)。结论:UVB可明显引起原代人角质形成细胞损伤,受损程度随辐射剂量增大而加重;EGCG可以减少UVB辐射所致的光产物的产生,加速光产物的清除。  相似文献

Oxytocin (OT), traditionally associated with reproductive functions, was revisited recently, which revealed several new functions of the hormone in cardiovascular regulation. To support this contention, we have demonstrated the presence and synthesis of OT receptors in all heart compartments and the vasculature. The functionality of these receptors has been established by the ability of OT to induce atrial natriuretic peptide and nitric oxide (NO) release from the perfused heart and atrial slices. OT's cardiovascular actions include natriuresis, blood pressure reduction, negative inotropic and chronotropic effects, parasympathetic neuromodulation, as well as vasodilatation triggered by the NO pathway that is also involved in endothelial cell growth and anti-inflammatory activity. In addition, we have reported the abundance of the OT system in the early developing heart and OT's capacity to generate cardiomyocytes from mouse embryonic stem cells. The most potent inducer of cardiac differentiation, OT-Gly-Lys-Arg, is an extended form of OT that is abundantly expressed in the fetal heart. Therefore, in pathological conditions, OT plays an anti-inflammatory and cardioprotective role, improving vascular and metabolic functions; it has potential for therapeutic use.  相似文献

目的 筛选影响纳武单抗和派姆单抗治疗非小细胞肺癌(non-small cell lung cancer, NSCLC)疗效的差异基因,为免疫治疗药物的选择及治疗预后提供参考。方法 通过GEO数据库搜索“Nivolumab”、“Pembrolizumab”找到目的芯片,下载免疫治疗相关表达芯片“GSE93157”,筛选NSCLC相关样本共35个,利用R语言数据包对样本进行表达差异基因进行聚类分析。对差异基因进行基因功能注释GO分析和KEGG通路分析,构建蛋白相互作用网络,筛选枢纽基因进行生存分析,确定影响不同抗程序性细胞死亡蛋白1药物治疗的关键基因。结果 筛选出影响纳武单抗治疗疗效差异基因共58个,其中免疫相关基因25个;影响派姆单抗治疗疗效差异基因231个,免疫相关基因82个。基于两种药物免疫相关差异基因的蛋白互作网络提示纳武单抗共得到2个子网络,主要模块共11个节点,51个边;派姆单抗共得到4个子网络,主要模块共24个节点,231个边。影响两种药物治疗疗效的前10位主要免疫相关基因生存分析,显示生存差异具有统计学意义(P<0.05)的基因,与纳武单抗相关的免疫差异基因为CD5、...  相似文献

ObjectiveThe aim was to determine the role of visfatin (VF) and human fetuin A (AHSG) in the development of gestational diabetes mellitus (GDM) and to explore the association between these variables and adverse outcomes.Materials and methodsWe carried out our study on 68 cases of GDM pregnant women and 42 cases of healthy pregnant women, including 56 cases with diet control and 12 cases with insulin treatment. Enzyme-linked immunoassay (ELISA) was used to test the expression levels of VF and AHSG in maternal and umbilical cord serum. Immunohistochemistry (ICH) was used to test the expression level of the VF protein in placental tissue.ResultsThe expression levels of VF and AHSG in maternal and umbilical cord serum and the expression level of VF in placental tissue in GDM pregnant women were higher than those in healthy pregnant women. The incidence of adverse outcomes in the GDM pregnant women was higher than that in healthy pregnant women, and these differences were statistically significant (P < 0.05). Those who had higher expression levels of VF or AHSG had a higher incidence of adverse outcomes (P < 0.05).ConclusionThe expression of VF and AHSG may participate in the development of GDM. A test of VF and AHSG in GDM pregnant women may have some predictive value for the occurrence of adverse outcomes.  相似文献

Psychological stress is strain affecting the intangible self, caused by problems in adaptation, perception, and emotions. Previous studies have demonstrated that arginine vasopressin (AVP) plays an important role in psychological stress. The goal of present study was to investigate the interaction between AVP release and cardiovascular functions by measuring AVP concentration and recording blood pressure or heart rate during psychological stress in human. The results showed that (1) psychological stress not only increased the systolic blood pressure, diastolic blood pressure and heart rate, but also elevated the cortisol and AVP concentration in both plasma and CSF in a stress level-dependent manner; (2) there was a positive relationship between plasma AVP concentration and systolic blood pressure, diastolic blood pressure, heart rate or plasma cortisol concentration; (3) there was also a positive relationship between AVP concentrations in plasma and CSF AVP. The data suggested that plasma AVP, which might come from the central nervous system, might influence the cardiovascular functions during psychological stress in human.  相似文献
