A food survey was carried out with the aim to investigate the levels of lead (Pb), zinc (Zn), copper (Cu) and chromium (Cr) in various fruits and vegetables sold in Algeria. Concentrations (mg/kg dry wt.) in selected foodstuffs were detected within the following ranges: 4–29.49, 11.17–49, 12.33–39.33 and 3–16.33 for Cu, Zn, Pb and Cr respectively. The food ingestion rate of the selected items was investigated by self-administered questionnaires which were filled by a total of 843 people randomly recruited at the exit of markets. The potential health risk for consumers was investigated by estimating the daily intake (EDI) and the target hazard quotient (THQ) for each heavy metal. For all foodstuffs, the EDI and the THQ were below the threshold values for Cu, Zn and Cr while they exceeded the thresholds for Pb (EDI: 15.66 μg Pb/kg body weight/day; THQ: 4.37), indicating an obvious health risk over a life time of exposure.  相似文献

Raltegravir (RAL) is the first licensed antiretroviral integrase inhibitor that may be used both for treatment-naïve human immunodeficiency virus type 1 (HIV-1) patients and for salvage therapy. The Brazilian public free access programme limits its use for salvage therapy, with scarce information regarding RAL resistance from patients failing a RAL-containing salvage regimen. This study evaluated RAL resistance mutations detected by population sequencing in 69 HIV-infected patients with advanced disease failing a RAL-containing regimen in a real-world setting. RAL resistance mutations were identified in 47/69 patients (68%). The most common salvage regimen, used by 56/69 patients (81%), included lamivudine, tenofovir, darunavir/ritonavir and RAL. At failure, major RAL resistance mutations included Q148H/R/K (21/47; 45%), N155H (14/47; 30%), Y143R/H/C (3/47; 6%) and E92Q (1/47; 2%). Most samples with Q148H/R/K also showed G140S/A/C (21/47; 45%). RAL resistance was significantly associated with less than two active drugs in the optimised background therapy regimen at failure [39/39 (100%) vs. 9/17 (53%); P < 0.001] and with a longer cumulative duration with detectable viraemia (viral load >50 copies/mL) (86 weeks vs. 32 weeks; P = 0.001). A high frequency of RAL mutations was observed in this study. In addition, these results reinforce the importance of close monitoring of RAL-containing regimens to reduce the time of failure and consequent resistance accumulation.  相似文献

Plant growth-promoting rhizobacteria (PGPR) not only promote growth and heavy metal uptake by plants but are promising biosorbents for heavy metals remediation. However, there exist arguments over whether extracellular adsorption (biosorption) or intracellular accumulation (bioaccumulation) play dominant roles in Cd(ii) adsorption. Therefore, three cadmium-resistant PGPR, Cupriavidus necator GX_5, Sphingomonas sp. GX_15, and Curtobacterium sp. GX_31 were used to study bioaccumulation and biosorption mechanisms under different initial Cd(ii) concentrations, using batch adsorption experiments, desorption experiments, scanning electron microscopy coupled with energy dispersive X-ray (SEM-EDX) spectroscopy, transmission electron microscopy (TEM), and Fourier-transform infrared (FTIR) spectroscopy. In this study, with the increase of the initial Cd(ii) concentrations, the removal efficiency of strains decreased and the adsorption capacity improved. The highest Cd(ii) removal efficiency values were 25.05%, 53.88%, and 86.06% for GX_5, GX_15, and GX_31 with 20 mg l−1 of Cd(ii), while the maximum adsorption capacity values were 7.97, 17.13, and 26.43 mg g−1 of GX_5, GX_15, and GX_31 with 100 mg l−1 of Cd(ii). Meanwhile, the removal efficiency and adsorption capacity could be ordered as GX_31 > GX_15 > GX_5. The dominant adsorption mechanism for GX_5 was bioaccumulation (50.66–60.38%), while the dominant mechanisms for GX_15 and GX_31 were biosorptions (60.29–64.89% and 75.93–79.45%, respectively). The bioaccumulation and biosorption mechanisms were verified by SEM-EDX, TEM and FTIR spectroscopy. These investigations could provide a more comprehensive understanding of metal-bacteria sorption reactions as well as practical application in remediation of heavy metals.

Borrelia burgdorferi s.s., the bacterium that causes Lyme disease in North America, circulates among a suite of vertebrate hosts and their tick vector. The bacterium can be differentiated at the outer surface protein C (ospC) locus into 25 genotypes. Wildlife hosts can be infected with a suite of ospC types but knowledge on the transmission efficiencies of these naturally infected hosts to ticks is still lacking. To evaluate the occupancy and detection of ospC types in wildlife hosts, we adapted a likelihood-based species patch occupancy model to test for the occurrence probabilities (ψ – “occupancy”) and transmission efficiencies (ε – “detection”) of each ospC type. We detected differences in ospC occurrence and transmission efficiencies from the null models with HIS (human invasive strains) types A and K having the highest occurrence estimates, but both HIS and non-HIS types having high transmission efficiencies. We also examined ospC frequency patterns with respect to strains known to be invasive in humans across the host species and phylogenetic groups. We found that shrews and to a lesser extent, birds, were important host groups supporting relatively greater frequencies of HIS to non-HIS types. This novel method of simultaneously assessing occurrence and transmission of ospC types provides a powerful tool in assessing disease risk at the genotypic level in naturally infected wildlife hosts and offers the opportunity to examine disease risk at the community level.  相似文献

BackgroundMany methods reportedly prevent contrast-induced nephropathy (CIN), but the effect of brain natriuretic peptide (BNP) on CIN is unknown. In this study we investigated recombinant BNP use before coronary angiography (CA) or nonemergent percutaneous coronary intervention (PCI) in patients with unstable angina.MethodsOne thousand patients with unstable angina were prospectively evaluated. The patients were randomly assigned to: group A, isotonic normal saline (NaCl 0.9%, 1 mL/kg/h) for 24 hours before CA or PCI; and group B, human recombinant BNP (rhBNP; 0.005 μg/kg/min). Serum creatinine (Scr) levels and estimated glomerular filtration rate were measured before and 24, 48, and 72 hours, and 7 days after the procedure. The primary outcome was CIN incidence defined according to a relative (≥ 25%) or absolute (≥ 0.5 mg/dL and 44 μmol/L, respectively) increase in Scr from baseline within 48 hours. The secondary end points were the changes in the Scr and estimated glomerular filtration rate, before and after the procedure.ResultsContrast volume, a history of diabetes mellitus, and BNP administration independently predicted CIN. The incidence of CIN was significantly greater in group A than in group B (14.8% vs 5.6%; P < 0.01). Renal function was less compromised in patients who received rhBNP. The Scr of all patients with CIN remained increased for 24 hours, but it was lower and recovered faster in patients who received rhBNP.ConclusionsrhBNP administration before CA or PCI protects renal function and can significantly decrease CIN incidence.  相似文献

During evolution, reproductive hormones and their receptors in the brain‐pituitary‐gonadal axis have been altered by genetic mechanisms. To understand how the neuroendocrine control of reproduction evolved in mammals, it is important to examine marsupials, the closest group to placental mammals. We hypothesised that at least some of the hormones and receptors found in placental mammals would be present in koala, a marsupial. We examined the expression of koala mRNA for the reproductive molecules. Koala cDNAs were cloned from brain for gonadotrophin‐releasing hormones (GnRH1 and GnRH2) or from pituitary for GnRH receptors, types I and II, follicle‐stimulating hormone (FSH)β and luteinising hormone (LH)β, and from gonads for FSH and LH receptors. Deduced proteins were compared by sequence alignment and phylogenetic analysis with those of other vertebrates. In conclusion, the koala expressed mRNA for these eight putative reproductive molecules, whereas at least one of these molecules is missing in some species in the amniote lineage, including humans. In addition, GnRH1 and 2 are shown by immunohistochemistry to be expressed as proteins in the brain.  相似文献

Phenylethanoid glycosides (PhGs) are generally water-soluble phenolic compounds that occur in many medicinal plants. Until June 2020, more than 572 PhGs have been isolated and identified. PhGs possess antibacterial, anticancer, antidiabetic, anti-inflammatory, antiobesity, antioxidant, antiviral, and neuroprotective properties. Despite these promising benefits, PhGs have failed to fulfill their therapeutic applications due to their poor bioavailability. The attempts to understand their metabolic pathways to improve their bioavailability are investigated. In this review article, we will first summarize the number of PhGs compounds which is not accurate in the literature. The latest information on the biological activities, structure–activity relationships, mechanisms, and especially the clinical applications of PhGs will be reviewed. The bioavailability of PhGs will be summarized and factors leading to the low bioavailability will be analyzed. Recent advances in methods such as bioenhancers and nanotechnology to improve the bioavailability of PhGs are also summarized. The existing scientific gaps of PhGs in knowledge are also discussed, highlighting research directions in the future.  相似文献

Exendin-4 (EX4) has been reported to attenuate myocardial ischemia and reperfusion (I/R) injury and inflammatory and oxidative responses. Nuclear DNA-binding protein high-mobility group box 1 (HMGB1) protein acts as a late mediator of severe vascular inflammatory conditions. However, the effect of EX4 on HMGB1-induced inflammatory response has not been studied. First, we accessed this question by monitoring the effects of posttreatment EX4 on lipopolysaccharide (LPS) and cecal ligation and puncture (CLP)-mediated release of HMGB1 and HMGB1-mediated regulation of proinflammatory responses in human umbilical vein endothelial cells (HUVECs) and septic mice. Posttreatment EX4 was found to suppress LPS-mediated release of HMGB1 and inhibited HMGB1-mediated hyperpermeability and leukocyte migration in septic mice. EX4 also induced downregulation of CLP-induced release of HMGB1, production of IL-6, and mortality. Collectively, these results suggest that EX4 may be regarded as a candidate therapeutic agent for treatment of vascular inflammatory diseases via inhibition of the HMGB1 signaling pathway.  相似文献
