目的:结合紫外光谱(UV-s)和惩罚岭型线性判别(PRLDA)模式识别方法建立短串联重复序列(STR)的基因分型方法。方法:以D16S539基因座的9-9,12-13两种基因型为研究对象,研究包含该多态性位点的DNA片段的聚合酶链反应(PCR)扩增条件和紫外检测条件,建立标准化扩增条件和检测条件,以此获得基因分型的标准物及其标准UV-s。以标准光谱为识别变量,建立了9-9与12-13基因型的PRLDA判别模型。结果:所建立的PRLDA模型有较大的类间距和较小的类内距,表明模型具有良好的稳健性,且预测样本均分布在校正样本集的范围之内,对预测样本也表现出了极强的判别能力。结论:该方法不需任何检测前处理,而只需一步PCR扩增和UV-s检测即可实现STR的基因分型,具有简单、快速、低成本等优点。 相似文献

Tetrazolium violet (TV), a tetrazolium salt, was synthesized as a novel and potent anticancer agent with a broad spectrum
of anticancer activity against many cancer cells. A previous study showed that tetrazolium violet inhibited cell growth, and
induced cell cycle arrest and apoptosis in C6 Rat glioma cells. It also showed that treatment of cells with TV for 24 h resulted
in a dramatic up-regulation of p53, and an increase in the activity of caspase-3, accompanied with a significant increase
of Bax/Bcl-2 ratio. In this study, we further investigated which Fas/FasL and caspase were activated by TV during the apoptosis.
Annexin-V-propidium iodide (PI) binding assay and nucleosome ELISA assay further indicated that TV induced a typical apoptosis,
in a time-dose-dependent manner. The data showed that the activity of Fas/FasL and caspase-8 and -9 were significantly enhanced
by the compound, which suggested that TV might be used as a Fas/FasL and caspases promoter to initiate brain cancer cell apoptosis. 相似文献

We report a complication occurring after the use of a bioceramic material for a three-dimensional reconstructive cranioplasty. A patient who had a satisfactory aesthetic result after frontal skull bone reconstruction with bioceramic injured his head after falling out of bed. The bioceramic material fragmented and the three fragments migrated over each other to form a lump. The bioceramic material had to be surgically removed. Care must be taken when using the fragile bioceramic material for reconstructive purposes, especially when patients are at risk of frequent falls. 相似文献

目的考察巫山淫羊藿悬浮培养细胞的生长特性,为巫山淫羊藿资源利用提供参考。方法选用悬浮培养所获得的均匀悬浮培养细胞为研究对象,在光照条件下进行悬浮培养,每隔3 d取样,测定相关的生理生化指标。结果巫山淫羊藿悬浮细胞的生长曲线呈"S"型,培养基中蔗糖、磷酸盐、硝酸盐、铵盐量均呈急剧下降趋势,最后变化趋于平缓,细胞合成黄酮最佳光照时间为每天12 h,黄酮合成量为27.854 82 mg/L。结论巫山淫羊藿细胞生长特性的研究为其大规模悬浮培养生产天然活性成分奠定了基础。 相似文献

Background and aimsReducing consumption of sugar-sweetened beverages (SSBs) is a global public health priority because of their limited nutritional value and associations with increased risk of obesity and metabolic diseases. Gut microbiota-related metabolites emerged as quintessential effectors that may mediate impacts of dietary exposures on the modulation of host commensal microbiome and physiological status.Methods and resultsThis study assessed the associations among SSBs, circulating microbial metabolites, and gut microbiota–host co-metabolites, as well as metabolic health outcomes in young Chinese adults (n = 86), from the Carbohydrate Alternatives and Metabolic Phenotypes study in Shaanxi Province. Five principal component analysis-derived beverage drinking patterns were determined on self-reported SSB intakes, which were to a varying degree associated with 143 plasma levels of gut microbiota-related metabolites profiled by untargeted metabolomics. Moreover, carbonated beverages, fruit juice, energy drinks, and bubble tea exhibited positive associations with obesity-related markers and blood lipids, which were further validated in an independent cohort of 16,851 participants from the Regional Ethnic Cohort Study in Northwest China in Shaanxi Province. In contrast, presweetened coffee was negatively associated with the obesity-related traits. A total of 79 metabolites were associated with both SSBs and metabolic markers, particularly obesity markers. Pathway enrichment analysis identified the branched-chain amino acid catabolism and aminoacyl-tRNA biosynthesis as linking SSB intake with metabolic health outcomes.ConclusionOur findings demonstrate the associations between habitual intakes of SSBs and several metabolic markers relevant to noncommunicable diseases, and highlight the critical involvement of gut microbiota-related metabolites in mediating such associations. 相似文献
