目的利用四环素调控复制缺陷性单纯疱疹病毒I型重组载体(QR9TO-LacZ)感染体外原代培养的大鼠皮层神经元,探讨其介导半乳糖苷酶(LacZ)基因的表达情况及基因调控效能;并检测该载体对神经元活力的影响。方法用RUL9-8细胞系扩增QR9TO-LacZ病毒株,收集后采用噬斑法测定病毒滴度;体外原代培养大鼠皮层神经元。将培养的原代神经元分为强力霉素(DOX)组和对照组。两组细胞均用QR9TO-LacZ转染,强力霉素组加入四环素衍生物强力霉素(0.5μg/ml),对照组加入等剂量空白对照液。同时根据感染复数(MOI),即MOI=3、5、10、30 PFU/cell,将两组培养细胞进一步分为亚组。病毒转染48h后行X-gal染色;光镜下观察神经元形态,随机选取10个高倍镜视野(×400),计算每高倍镜视野中平均蓝染细胞率,比较不同MOI病毒感染神经元的效率。采用四甲基偶氮唑盐微量酶反应比色法(MTT法)检测感染病毒48 h(MOI同上)后神经元的活性。结果在含有DOX的培养环境中,不同MOI亚组(3、5、10、30 PFU/cell)QR9TO-LacZ感染神经元出现蓝染细胞百分比分别为5.35%±0.84%、10.64%±1.92%、19.73%±2.87%、34.41%±2.58%;在无DOX的培养环境中,各亚组均未观察到蓝染细胞。上述MOI感染的神经元细胞存活率分别为:90.24%±8.16%、88.00%±5.69%、83.95%±3.82%、78.90%±5.49%。结论 QR9TO-LacZ载体能有效地将目的基因导入原代培养神经元中并表达;目的基因表达的开启受强力霉素调控;QR9TO-LacZ对体外原代培养神经元毒性较低。 相似文献

Cataloguing the effects of different types of stress on behaviour and physiology in rodent models has not been comprehensively attempted. Here, we systematically review whether chronic exposure to physical stress, psychosocial stress, or both types of stress can induce different behavioural and neurobiological outcomes in male and female rodents. We found that physical stress consistently increased depressive-like behaviour, impaired social interaction and decreased body weight, while psychosocial stress consistently increased both anxiety- and depressive-like behaviour, impaired social interaction and learning and memory, increased HPA axis activity, peripheral inflammation and microglial activation, and decreased hippocampal neurogenesis in male rodents. Moreover, we found that the combined effect of both stress types resulted in a more severe pathological state defined by increased anxiety- and depressive-like behaviour, impaired social interaction and learning and memory, increased HPA axis activity and central inflammation, and reduced hippocampal neurogenesis and neural plasticity in male rodents. Phenotypes for females were less consistent, irrespective of the type of stress exposure, on account of the limited number of studies using females. This review highlights that the type of stress may indeed matter and will help animal researchers to more appropriately choose a stress/depression model that fits their research purposes. 相似文献

Background and aimA prominent hallmark of tumors is aberrant lipid metabolism, and various peroxisome-related genes (PRGs) are associated with aberrant tumoral metabolic signaling. However, the influence of PRGs on the prognosis of hepatocellular carcinoma (HCC) patients remains debatable. Thus, the current study was designed to evaluate the effect of PRGs on HCC and construct a prognostic model for predicting survival.MethodsWe initially acquired HCC-related gene expression profiles from the Cancer Genome Atlas and International Cancer Genome Consortium databases. We then utilized Cox analysis and Lasso regression to identify suitable PRGs for the risk model. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses were conducted to clarify the functional roles of PRGs. Single-sample gene set enrichment analysis (ssGSEA) was conducted to confirm the relationship between PRGs and immunity.ResultsFour PRGs were correlated with HCC patient survival: 2 risk genes (MPV17, and ABCD1) and 2 protective genes (ACSL1 and ACSL6). We derived risk scores based on PRGs to construct a predictive model that could accurately predict overall survival (OS) among HCC patients. Furthermore, GO and KEGG analyses revealed that these PRGs were potentially involved in lipid metabolism and ferroptosis in HCC. Moreover, ssGSEA results demonstrated that high PRG scores were associated with immune suppressor activation, which caused the suppression of immune effectors (CD8+ T-cells, B cells, and NK cells) and the attenuation of the immune-mediated antitumor effect.ConclusionPRGs act as key regulators in tumorigenesis and tumor progression by affecting lipid synthesis and utilization, which we used to predict the outcome of HCC patients. Moreover, PRGs have been shown to promote tumoral immune resistance by serving as a vital bridge between metabolism and immunity. Thus, a personalized treatment approach targeting PRGs would clinically benefit patients by blocking the interaction between tumor metabolism and immunity. 相似文献

Chondrosarcoma is the second most common type of primary bone malignancy following up osteosarcoma, characterized by resistance to conventional chemotherapeutic agents and radiation regimens. The p160 family members steroid receptor coactivator-1 and -3 (SRC-1 and SRC-3) have been implied in the regulation of cancer growth, migration, invasion, metastasis and chemotherapeutic resistance; but we still lack detailed information about the levels of SRCs in chondrosarcoma. In this study, expression of SRC-1 and SRC-3 in chondrosarcoma was examined by immunohistochemistry with tissue microarrays; the four score system (0, 1, 2 and 3) was used to evaluate the staining. The results showed that there were no gender-, site- or age-differences regarding the expression of SRC-1 or SRC-3 (p > 0.05); organ (bone or cartilage) -differences were only detected for SRC-1 but not SRC-3 (p < 0.05). Significant higher levels of SRC-1 and SRC-3 were detected in MDC and PDC when compared to WDC. Our study clearly demonstrated differentiation-dependant expression of SRC-1 and SRC-3 in chondrosarcoma, may be novel targets for the prognosis and/or treatment of chondrosarcoma, would have opened a new avenue and established foundation for studying chondrosarcoma. 相似文献

目的探讨两种无创正压通气在新生儿呼吸窘迫综合征(NRDS)初始治疗中的应用,及对其早期脑功能发育的影响。方法 2016年6月-2018年8月安徽医科大学第三附属医院NICU收治的符合入选标准的NRDS患儿50例,给予气管插管、使用肺表面活性物质、拔管,按照随机数字表法随机分为两组:nCPAP组(25例)与n Bi PAP组(25例)。根据TcSO2或血气分析调整FiO2等参数,记录各种临床指标。评估正压通气后24 h Tc SO2、血气参数及FiO2,应用aEEG评估脑成熟度。结果正压通气后24 h nBiPAP组FiO2(%)低于nCPAP组(P=0. 003),TcSO2(%)高于nCPAP组(P=0. 003);n BiPAP组aEEG得分明显高于n CPAP组,且a EEG各项评分与FiO2呈负相关(均P<0. 01),与TcSO2呈正相关(均P<0. 01)。结论在NRDS初始治疗中,nBiPAP组较nCPAP组更好地促进了肺氧合,减低了对FiO2的需求,改善了早产儿的早期脑功能。 相似文献
