目的:观察腹腔镜联合胆道镜治疗胆囊结石合并胆总管结石的疗效。方法将2012年1月~2014年1月我院应用腹腔镜联合胆道镜治疗胆囊结石患者合并胆总管结石50例患者作为观察组,另选择同期应用传统开腹手术治疗的胆囊结石患者合并胆总管结石患者50例作为对照组,观察比较两组患者的手术时间、术中出血量、置入引流管时间、住院时间及并发症发生率。结果观察组患者的手术时间、置入引流管时间、住院时间均显著短于对照组,术中出血量显著少于对照组,差异有统计学意义(P <0.05或 P<0.01)。观察组并发症的发生率4.0%,明显低于对照组并发症的发生率20.0%,组间比较,差异具有显著性(x2=11.237,P <0.05)。结论腹腔镜联合胆道镜治疗胆囊结石合并胆总管结石疗效确切,可以缩短手术时间及住院时间、降低并发症的发生率,值得推广和应用。  相似文献

目的:对阴虚血热出血大鼠含鲜地黄血清进行分析,探索鲜地黄清热凉血药效的物质基础。方法:给予血热出血症模型大鼠鲜地黄,采集血清,用高效液相色谱法分析大鼠含药血清、正常血清以及鲜地黄药材的色谱,研究鲜地黄在血中移行成分以及鲜地黄对大鼠血清中固有成分的影响。结果:在血热出血症模型大鼠含药血清中检测到了鲜地黄的有效成分——梓醇的色谱峰;且发现血清中和药材中有3种相同的共有成分;鲜地黄对模型大鼠血清中1种固有成分的升高有抑制作用。结论:鲜地黄中梓醇及3种未明成分可能是其清热凉血物质基础;鲜地黄对动物血清中固有成分代谢的影响可能是鲜地黄清热凉血药效的机制  相似文献

目的:考察异长春花苷内酰胺( strictosamide,STR)的抗菌、抗病毒活性,为其用于治疗上呼吸道感染提供一定的实验依据.方法:采用试管法与平皿法测定其对标准金黄色葡萄球菌等10株菌株的体外抑制作用,并观察对金黄色葡萄球菌和肺炎双球菌感染小鼠的保护作用,即将小鼠分为正常组,模型组,热毒宁对照组(生药剂量8.7g·kg-)及异长春花苷内酰胺低、高剂量(20,40 mg· kg -)组,每天ip给药1次,连续7d,末次给药后1h,腹腔注射金黄色葡萄球菌(或肺炎双球菌)菌液,观察小鼠7d内死亡情况;接种甲型流感病毒[A/PR8/34( H1 N1)]和乙型流感病毒(B/京防98-76)于MDCK(狗肾细胞),观察其体外抗病毒作用,并采用甲型流感病毒感染小鼠模型考察其体内抗病毒作用,即将小鼠分为正常组,模型组,热毒宁对照组(生药剂量8.7 g·kg-1)及异长春花苷内酰胺低、高剂量( 20,40 mg· kg-)组,每天ip给药1次,连续5d,于给药第1天以病毒尿囊液滴鼻感染小鼠,继续给药4d后观察肺部病变,作病理组织学检查,计算肺指数与抑制率.结果:异长春花苷内酰胺对标准大肠杆菌等7株菌株具有抑制作用,MIC均为5 g·L-1;质量浓度为10 g·L-1时,对肺炎双球菌等5株菌株的抑菌圈直径为13~25 mm;40 mg· kg -时可分别使金黄色葡萄球菌和肺炎双球菌感染小鼠的存活率达70%和55%;其体外对甲型与乙型流感病毒的IC50分别为0.649,0.323 g·L-1;对甲型流感病毒所致的小鼠肺指数升高与肺组织病变亦有明显降低和改善作用,40 mg· kg -时的肺指数抑制率为39.8%.结论:异长春花苷内酰胺具有抗菌、抗病毒活性.  相似文献

AimThe present work aims to assess the effectiveness of lentiviral vector (LV) mediated Toll-like receptor 2 (TLR2) gene silencing in the survival of transplanted corneal allografts, against immune rejection, in rats.MethodsLV mediated TLR2 small interference RNA (SiRNA) with enhanced green fluorescent protein (eGFP) [LV-TLR2-siRNA-eGFP] was realised and transfected to both rat corneal epithelial (EC) and stromal cells (SC). Multiplicity of infection (MOI) was optimized for transfection efficiency using flow cytometric (FCM) analysis. Viability of transfected cells and the success rate of TLR2 gene silencing were respectively determined by CCK-8 assay and western blot assay. The in-vivo experiments were subjected to a penetrating keratoplasty (PK) performed between host Sprague Dawley (SD) and donor Wistar/SD rats, randomly dividing them into 4 groups including the allograft, isograft, allograft treated with LV-eGFP (LV blank control) and allograft treated with LV-TLR2-siRNA-eGFP (LV treated group). The rejection index (RI) was then recorded under a slit lamp, every day following surgery. Expression of the TLR2 and Myeloid differentiation primary response gene 88 (MyD88) were detected by using immunohistochemistry on day 9 post-surgery, whereas grafts’s TLR2 and MyD88 mRNA were determined on day 5, 9, and 14 post-surgery performing RT-PCR and, normal rat corneas were included as additional controls.ResultsTransfected cells showed the strongest eGFP expression when MOI was 200 with an efficiency of 77.5% for EC and 76.3% for SC. CCK-8 assay, as measured at 72 and 96 h post transfection, showed no significant changes in the cell viability (both EC and SC) between the transfected and the control group (p > 0.05, p > 0.05). Western blot results demonstrated a successful down regulation of TLR2 expression by LV-TLR2-SiRNA-eGFP, in both EC and SC. In vivo, compared to allograft group, LV treated group demonstrated less edema, opacity and neovascularization. Immunohistochemical analysis revealed a lower expression of TLR2 and MyD88 in isograft and LV treated group as compared to allograft group. TLR2 and MyD88 mRNA were detected in all grafts, and increased over time. With its highest expression in allograft group (peak on day 9), the mRNA expression for TLR2 and MyD88 showed a significant difference amongst the groups, on both day 9 and 14 (p < 0.001).ConclusionsLV mediated TLR2 siRNA could effectively down regulate the TLR2 expression via RNA interference and prolong the survival of corneal grafts, although not necessarily able to prevent the rejection.  相似文献

The circadian timing system orchestrates daily variations in physiology and behavior through coordination of multioscillatory cell networks that are highly plastic in responding to environmental changes. Over the last decade, it has become clear that this plasticity involves structural changes and that the changes may be observed not only in central brain regions where the master clock cells reside but also in clock-controlled structures. This review considers experimental data in invertebrate and vertebrate model systems, mainly flies and mammals, illustrating various forms of structural circadian plasticity from cellular to circuit-based levels. It highlights the importance of these plastic events in the functional adaptation of the clock to the changing environment.  相似文献

Ethnopharmacological relevance

Curcuma zedoaria (Berg.) Rosc., a traditional Chinese herb, was used widely but absolutely prohibited for the pregnant in clinic. Based on that there is abundant angiogenesis in endometrium and placenta during gestation period, we hypothesized that some components from it could inhibit angiogenesis and then damaged the supply of oxygen and nutrition to the embryo, which finally led to gestation failure.

Aim of the study

This study was set to demonstrate whether essential oil, major components of Curcuma zedoaria had anti-angiogenic effect.

Materials and methods

Essential oil of Curcuma zedoaria (EO-CZ) was abstracted by steam distillation extraction. Cell proliferation assay and two angiogenic models, rat aortic ring assay and chick embryo chorioallantoic membrane assay were presented. Furthermore, melanoma growth and experimental lung metastasis assay in mice were performed to evaluate its anti-angiogenesis effect in vivo. Immunohistochemical analysis and enzyme-linked immunosorbent assay (ELISA) were used to respectively detect the expression of CD34 and matrix metalloproteinases (MMPs).

Results

EO-CZ exhibited anti-proliferative effect on B16BL6 and SMMC-7721 cells, the IC50, respectively was 41.8 μg/ml and 30.7 μg/ml, and on HUVEC (Human Umbilical Vein Endothelial Cells) cells with IC50 of far more than 120 μg/ml. Both 20 μg/ml and 40 μg/ml EO-CZ indicated significant suppression on sprouting vessels of aortic ring and formation of microvessels in chick embryo chorioallantoic membrane in vitro. Moreover, solid melanoma grown in left oxter of mice was obviously inhibited after oral intake of 100 and 200 mg/kg of EO-CZ a day for 28 days, and CD34 expression indicating angiogenesis in melanoma reduced significantly compared with control; melanoma metastatic nodules in lung were detected to be inhibited, as well as MMP-2 and MMP-9 expression in serum.

Conclusions

Essential oil, a fat-soluble fraction of Curcuma zedoaria, presented anti-angiogenic activity in vitro and in vivo, resulting in suppressing melanoma growth and lung metastasis. And this was associated with down-regulating MMPs.  相似文献
