AIM: To explore the effect of histone deacetylase inhibitor, trichostatin A (TSA) on the growth of biliary tract cancer cell lines (gallbladder carcinoma cell line and cholangiocarcinoma cell line) in vivo and in vitro, and to investigate the perspective of histone deacetylase inhibitor in its clinical application. METHODS: The survival rates of gallbladder carcinoma cell line (Mz-ChA-l cell line) and cholangiocarcinoma cell lines (QBC939, KMBC and OZ cell lines) treated with various doses of TSA were detected by methylthiazoy tetrazolium (MTT) assay. A nude mouse model of transplanted gallbladder carcinoma (Mz-ChA-l cell line) was successfully established, and changes in the growth of transplanted tumor after treated with TSA were measured. RESULTS: TSA could inhibit the proliferation of gallbladder carcinoma cell line (Mz-ChA-l cell line) and cholangiocarcinoma cell lines (QBC939, KMBC and OZ cell lines) in a dose-dependent manner. After the nude mouse model of transplanted gallbladder carcinoma (Mz- ChA-l cell line) was successfully established, the growth of cancer was inhibited in the model after treated with TSA. CONCLUSION: TSA can inhibit the growth of cholangiocarcinoma and gallbladder carcinoma cell lines in vitro and in vivo. 相似文献

Effects of splenectomy in patients with cirrhosis undergoing hepatic resection for hepatocellular carcinoma 总被引:5,自引:0,他引:5
AIM: To study the effects of splenectomy in patients with cirrhosis undergoing hepatic resection for hepatocellular circinoma.METHODS: Twenty-six patients with HCC associated with cirrhosis were divided into hepatectomy with splenectomy group (splenectomy group, n=11) and hepatectomy without splenectomy group (non-splenectomy group, n=15). T lymphocyte subsets such as CD4, CD8, CD4/CD8, helper T (Th) lymphocyte cytokines such as interferon γ(IFN-γ),interleukin 2(IL-2), interleukin 10(IL-10) and white blood cell (WBC), platelet (PLT), total bilirubin (T-Bil) were measured and used as parameters to evaluate the effects of splenectomy.RESULTS: There was no significant difference in CD4, CD8,CD4/CD8, IL2, IFN-γ, IL10, WBC, PLT, T-Bil levels between two groups before surgery. Two months after operation,the levels of CD4 (41.2 %±4.2 % vs 34.7 %±3.8 %), CD4/CD8 (1.7±0.2 vs 1.0±0.2), IFN-γ (102.3±15.9 pg/ml vs 86.5±14.8 pg/ml), IL-2(97.2±15.6 pg/ml vS77.6±14.5 pg/ml) were increased and those of CD8 (25.6±3.9 vs32.8 %±4.1%),IL-10 (56.9±10.4 pg/ml vs 72.6±15.3 pg/ml) were decreased in splenectomy groups as compared with those in nonsplenectomy group (P<0.05). WBC and PLT counts in the splenectomy group were 8.9±1.6×109 and 310±32×109,respectively, which were significantly higher than those in non-splenectomy group (3.7±1.4×109 and 104±41×109) respectively on the 14th post-operative day. T-Bil concentration in the splenectomy group (24±7 μmol/L) was significantly lower than that in the non-splenectomy group (37±13 μmol/L) on the 7th post-operative day (P<0.05).CONCLUSION: Splenectomy combined with hepatectomy for HCC associated with cirrhosis is helpful for the recovery of T-lymphocyte subsets and the maintenance of Th:L/Th2 cytokine balance. 相似文献

目的探索3D打印个体化人工椎体在胸腰椎肿瘤整块切除后脊柱稳定性重建中的临床疗效。方法回顾性分析华中科技大学同济医学院附属同济医院2015年7月至2020年6月收治的接受一期后路全脊椎整块切除术的胸腰椎肿瘤患者28例,分为2组,每组14例。其中3D组使用3D打印个体化人工椎体重建,常规组使用钛笼重建。对比两组的住院时间、手术时间、术中出血量,记录术前、术后7 d及末次随访时视觉模拟评分(VAS),Frankel分级情况,测量节段高度及角度,评估临床疗效。结果两组患者住院时间、手术时间、术中出血量和VAS评分比较,差异无统计学意义。共8例患者Frankel评分获得一个等级的改善(28.6%)。3D组椎间高度丢失(1.9±2.2)mm,内植物沉降2例,沉降率14.3%;常规组椎间高度丢失(6.6±5.5)mm,内植物沉降8例,沉降率57.1%;两组比较差异有统计学意义(<0.05)。在节段角度丢失方面组间差异无统计学意义(=0.571)。3D组所有患者内固定良好,常规组1例患者发生断棒情况。结论 3D打印人工椎体可以更好地维持节段高度,降低椎间隙塌陷和内固定失败的风险。 相似文献

目的:研究归肾经方对阿霉素(adriamycin,ADR)诱导的肾小球足细胞损伤标志蛋白nephrin及自噬相关蛋白Beclin-1、LC3Ⅱ/Ⅰ、p-mTOR等表达的影响。 方法:以ADR体外诱导的方法制作足细胞损伤的细胞模型。除空白组外均以ADR诱导足细胞,分为5组:空白组(control group,10%空白血清)、模型组(model group,10%空白血清+ADR 0.5 μg·mL-1)、中药高剂量组(H-TCM group,10%高剂量归肾经方含药血清+ADR 0.5 μg·mL-1)、中药低剂量组(L-TCM group,10%低剂量归肾经方含药血清+ADR 0.5 μg·mL-1)、替米沙坦组(TMST group,10%替米沙坦含药血清+ADR 0.5 μg·mL-1)。CCK-8检测各组细胞活性改变,Western blot法检测各组足细胞标志蛋白nephrin、desmin及自噬相关蛋白beclin-1、LC3Ⅱ/Ⅰ等表达。 结果:①与空白组比较,模型组细胞活力显著降低(P<0.01),与模型组比较,中药高剂量组及替米沙坦组细胞活性显著增高(P<0.05);②Western blot检测结果显示,与control组比较,model组nephrin显著降低(P<0.01),desmin、LC3Ⅱ/Ⅰ、Beclin-1、p-Akt、p-mTOR显著增高(P<0.01);与Model组比较,H-TCM组及TMST组nehprin表达显著增高(P<0.05),desmin表达显著降低(P<0.05),H-TCM、L-TCM及TMST组Beclin-1、LC3Ⅱ/Ⅰ、p-Akt、p-mTOR表达显著降低(P<0.01);与H-TCM组比较,L-TCM组LC3Ⅱ/Ⅰ表达显著增高(P<0.05)。结论:归肾经方可以修复ADR诱导的足细胞损伤,改善细胞内的自噬水平,与上调细胞内nephrin、Beclin-1、p-Akt、p-mTOR的表达有关。 相似文献

目的: 建立液相色谱-质谱联用(HPLC-MS/MS)法测定人血浆中富马酸单甲酯的方法,并应用于检测中国健康受试者血浆中富马酸单甲酯的浓度。方法: 血浆样品用醋酸铵稀释后经过96孔固相萃取板前处理,以含0.5%甲酸的水溶液和甲醇作为流动相,采用梯度洗脱方式在ACE Excel 3 C18-AR(2.1 mm×50 mm)色谱柱上进行分离,每个样品分析时间为4 min。在电喷雾负电离模式下,多反应监测检测离子对m/z 129.0→m/z 85.0(富马酸单甲酯)和m/z 134.0→m/z 90.0(内标,富马酸单甲酯-d5)。结果: 富马酸单甲酯线性范围为4~1 200 ng·mL-1(r2>0.99),定量下限为4 ng·mL-1,日内和日间精密度RSD ≤ 9.7%,准确度相对偏差均在±4.2%以内,平均回收率RSD ≤ 6.6%,基质效应在98.3%~100.9%之间。稳定性实验中,各种储存条件不影响对药品浓度的准确测定。结论: 本方法运行时间短,灵敏度高,重复性好,适用于富马酸单甲酯血浆浓度定量检测。 相似文献

AIM: To evaluate the effects of interferon-α-2b (IFN- α-2b) on expression of cyclooxygenase-2 (COX-2) and vascular endothelial growth factor (VEGF) in human hepatocellular carcinoma (HCC) inoculated in nude mice and to study the underlying mechanism of IFN-α- 2b against HCC growth. METHODS: Thirb/-two nude mice bearing human HCC were randomly divided into four groups (n = 8). On the 10th day after implantation of HCC cells, the mice in test groups (groups A, B and C) received IFN-α- 2b at a serial dose (10000 IU for group A, 20000 IU for group B, 40000 IU for group C sc daily) for 35 d. The mice in control group received normal saline (NS). The growth conditions of transplanted tumors were observed. Both genes and proteins of COX-2 and VEGF were detected by RT-PCR and Western blot. Apoptosis of tumor cells in nude mice was detected by TUNEL assay after treatment with IFN-α-2b. RESULTS: Tumors were significantly smaller and had a lower weight in the IFN-α-2b treatment groups than those in the control group (P 〈 0.01), and the tumor growth inhibition rate in groups A, B and C was 27.78%, 65.22% and 49.64%, respectively. The expression levels of both genes and proteins of COX-2 and VEGF were much lower in the IFN-α-2b treatment groups than in the control group (P 〈 0.01). The apoptosis index (AI) of tumor cells in the IFN-α-2b treatment groups was markedly higher than that in the control group (P 〈 0.01). Group B had a higher inhibition rate of tumor growth, a lower expression level of COX-2 and VEGF and a higher AI than groups A and C (P 〈 0.05), but there was no significant difference between groups A and C. CONCLUSION: The inhibitory effects of IFN-α-2b on implanted tumor growth and apoptosis may be associated with the down-regulation of COX-2 and VEGF expression. There is a dose-effect relationship. The medium dose of IFN-α-2b for inhibiting tumor growth is 20 000 IU/d. 相似文献
