子宫内膜息肉是妇科常见疾病,现代中医认为本病主要病理因素是瘀滞,临床常见气滞血瘀、痰湿瘀阻、湿热瘀滞、气虚血瘀、阳虚血瘀诸证,运用中医药辨证治疗、中医外治法,可有效改善临床症状,降低息肉复发率,提高妊娠率。本文将近十年中医药治疗子宫内膜息肉的临床研究作以综述。  相似文献

Linkage studies indicate close associations of certain HLA alleles with autoimmune diseases. To better understand how specific HLA alleles are related to disease pathogenesis, we have generated an HLA DR3/DQ2 transgenic mouse utilizing a 550-kb yeast artificial chromosome (YAC) construct containing the complete DRalpha, DRbeta1, DRbeta3, DQalpha, and DQbeta regions. The transgenic mouse (4D1/C2D) in an I-Abeta(o) background appears healthy with no signs of autoimmune diseases. Lymphoid tissues as well as CD4(+) T cells develop normally. Characterization of the transgene expression demonstrates that approximately 90% of B cells express high levels of DR3 and 50-70% of B cells express DQ2. CD11c(+) dendritic cells express high levels of DR and DQ. Approximately 12-18% of resting T cells are positive for DR expression, and further up-regulation to 40-50% expression is seen upon activation with anti-CD3/anti-CD28 mAb. These results suggest that the transgenic construct confers a high fidelity to the normal human temporal and spatial expression profile. Analysis of T cell receptor repertoire in transgenic mice confirms that DR3/DQ2 are able to mediate thymic selection. Furthermore, transgenic mice respond to a DR3-restricted antigen, demonstrating antigen processing and presentation by antigen-presenting cells (APC). Purified T cells from ovalbumin (OVA)-immunized 4D1 mice respond to human APC co-cultured with OVA, suggesting appropriate antigen/DR3 or DQ2 recognition by murine T cells. Immunoglobulin isotype switching is also observed, indicating functional T-B cognate interactions. Thus, the DR3/DQ2 transgenic mouse has normal lymphoid development and functionality that are mediated by HLA transgenes and can be used to investigate HLA-associated immunological questions.  相似文献

The most common models of CD4 T-cell deficiency are mice exogenously injected with anti-CD4 antibody (Ab), CD4 knockout (CD4−/−) and major histocompatibility complex (MHC) class II knockout (class II−/−) mice. We recently described the anti-CD4 Ab transgenic mouse (GK) as an improved CD4 cell-deficient model. This review compares this new GK mouse model with the widely available class II−/− and CD4−/− mice, when exposed to complex antigens (foreign grafts and during bacterial or viral infection). We highlight here the cytometric and functional differences (including Ab isotype, viral or bacterial clearance, and graft survival) among these CD4 cell-deficient models. For example, whereas grafts are generally rejected in class II−/− and CD4−/− mice as quickly as in wild-type mice, they survive longer in GK mice. Also, CD4−/− mice produce IgG against both simple model and complex antigens, but class II−/− and GK mice produce small amounts of IgG2a against complex antigens but not simple model antigens. These differences harbinger the caveats in the use of these various mice.  相似文献

以小鼠心肌组织异位移植和混合淋巴细胞反应为整体和离体模型,观察了阿片受体阻断剂纳曲酮对移植排异反应的影响。结果显示:给动物从术前开始腹腔注射纳曲酮共10天(每日二次,每次5mg/kg)可明显延长移植心肌组织的存活时间;加入纳曲酮(10-4~10-8mol/L)对混合淋巴细胞反应有抑制作用并呈量效关系。同时还观察到,给正常小鼠腹腔注射纳曲酮3天以上,可引起动物脾细胞由ConA诱导的淋巴细胞转化反应受抑制。以上结果说明纳曲酮可抑制移植排异反应,此作用有可能是通过阻断内源性阿片肽所致。  相似文献

目的 分析中药治疗糖尿病肾脏疾病(DKD)随机对照试验中结局指标的使用情况和其他试验设计要素,为中药治疗DKD临床试验的设计及核心指标集的构建提供依据。方法 计算机检索知网、万方、维普、中国生物医学等7个医学数据库及2个临床试验注册中心(clinicaltrials.gov、chinadrugtrials.org.cn)近5年收录的中药治疗DKD随机对照试验,运用Cochrance手册对纳入的文献进行偏倚风险评估,对结局指标及其他试验设计要素进行统计与分析。结果 共纳入97项随机对照试验(RCTs),包含5项试验注册方案,研究发现,97项研究总体偏倚风险较高;DKD Ⅲ期(36项,41.38%)和气阴两虚兼血瘀证(16次,26.23%)为应用最多的DKD分期及中医证型;92项RCTs的疗程最多96周,最少2周,其中52项(56.52%)研究疗程为12周;92项RCTs及5项注册试验共采用152种结局指标,使用1 040次,共分为8大类,实验室检查(血液)、实验室检查(尿液)、临床疗效、中医证候积分、生活质量量表、生命体征、其他指标及其他事件,应用频率较高的依次是血肌酐(68次,70.10%)、临床有效率(55次,56.70%)、空腹血糖(51次,52.58%)、尿素氮(48次,49.48%)、总胆固醇(47次,48.45%)、24 h尿蛋白定量(43次,44.33%)等;56项RCTs及2项注册试验共采用安全性指标53种,频数227次,使用频率较高的依次是,不良反应(49次,84.48%)、肝功能(28次,48.28%)、血常规(24次,41.38%)、心电图(17次,29.31%)、尿常规(14次,24.14%);10项RCTs及5项注册试验报告了主要结局指标;54项RCTs报告了临床有效率指标。结论 目前中药治疗DKD的RCTs结局指标设计尚不规范,未来应积极构建突显出中医药特色的核心结局指标集,提升临床研究的质量,提高试验结果的应用价值。  相似文献

To investigate the effect of inhibiting transforming growth factor‐β (TGF‐β1)/Smad2/3 signaling on rotator cuff (RC) healing. A bilateral supraspinatus tendon detachment‐repair model of Sprague‐Dawley (SD) rats was utilized. A total of 120 SD rats were randomly assigned to six groups and each group received the subacromial injection of normal saline, empty vectors, or lentiviral vectors containing small interfering RNA against TGF‐β1, Smad2, Smad3 at the bone‐tendon junction. Biomechanical and histological analyses were performed to evaluate bone‐tendon junction healing quality at 8 weeks after repair. Histologically, scar healing was found in all surgical groups. Animals with inhibited Smad3 exhibited better bone‐tendon junction structures with higher density, parallel orientation, and collagen fiber continuity than other surgical group animals. Immunohistochemistry revealed that the protein expression level of collagen I in animals with inhibited Smad3 was more prominent compared with all other surgical groups. Biomechanically, Animals with inhibited Smad3 showed better results in the maximum load at 4, 6, and 8 weeks after surgery compared with other surgical groups. Besides, C3H10T1/2 (Smad3?) cells increased TT‐D6 cell migration and tendon‐associated genes expression (scleraxis, tenascin C, collagen I) in coculture system. We conclude that inhibition of Smad3 promotes RC tendon healing in the rat supraspinatus model.  相似文献

Hepatic ischemia–reperfusion injury (HIRI) is of common occurrence during liver surgery and transplantation. Pinocembrin (PIN) is a kind of flavonoid monomer extracted from the local traditional Chinese medicine Penthorum chinense Pursh (P. chinense). However, the effect of PIN on HIRI has not determined. We investigated the protective effect and potential mechanism of PIN against HIRI. Model mice were subjected to partial liver ischemia for 60 min, experimental mice were pretreated with PIN orally for 7 days, and H2O2-induced oxidative damage model in AML12 hepatic cells was established in vitro. Histopathologic analysis and serum biochemical levels revealed that PIN had hepatoprotective activities against HIRI. The variation of GSH, SOD, MDA, and ROS levels indicated that PIN treatments attenuated oxidative stress in tissue. PIN pretreatment obviously ameliorated apoptosis, and restrained the expression of HMGB1 and TLR4 in vivo. In vitro, compared with H2O2 group, the contents of ROS, mitochondrial membrane potential, apoptotic cells, and Bcl-2 protein were decreased, while the Bax protein expression was increased. Moreover, HMGB-1 small interfering RNA test and western blotting showed that PIN pretreatment reduced HMGB1 and TLR4 protein levels. In conclusion, PIN pretreatment effectively protected hepatocytes from HIRI and inhibited the HMGB1/TLR4 signaling pathway.  相似文献

CYP2C9 is an important member of the cytochrome P450 enzyme superfamily with some 12 CYP2C9 alleles (*1-*12) being previously reported. Recently, we identified a new CYP2C9 allele with a Leu90Pro mutation in a Chinese poor metabolizer of lornoxicam [Si D, Guo Y, Zhang Y, Yang L, Zhou H, and Zhong D (2004) Pharmacogenetics 14:465-469]. The new allele, designated CYP2C9*13, was found to occur in approximately 2% of the Chinese population. To examine enzymatic activity of the CYP2C9*13 allele, kinetic parameters for lornoxicam 5'-hydroxylation were determined in COS-7 cells transiently transfected with pcDNA3.1 plasmids carrying wild-type CYP2C9*1, variant CYP2C9*3, and CYP2C9*13 cDNA. The protein levels of cDNA-expressed CYP2C9*3 and *13 in postmitochondrial supernatant (S9) from transfected cells were lower than those from wild-type CYP2C9*1. Mean values of Km and Vmax for CYP2C9*1, *3, and *13 were 1.24, 1.61, and 2.79 microM and 0.83, 0.28, and 0.22 pmol/min/pmol, respectively. Intrinsic clearance values (Vmax/Km) for variant CYP2C9*3 and CYP2C9*13 on the basis of CYP2C9 protein levels were separately decreased to 28% and 12% compared with wild type. In a subsequent clinical study, the AUC of lornoxicam was increased by 1.9-fold and its oral clearance (CL/F) decreased by 44% in three CYP2C9*1/*13 subjects, compared with CYP2C9*1/*1 individuals. This suggests that the CYP2C9*13 allele is associated with decreased enzymatic activity both in vitro and in vivo.  相似文献
