A fundamental shift to a total system approach for crop protection is urgently needed to resolve escalating economic and environmental consequences of combating agricultural pests. Pest management strategies have long been dominated by quests for “silver bullet” products to control pest outbreaks. However, managing undesired variables in ecosystems is similar to that for other systems, including the human body and social orders. Experience in these fields substantiates the fact that therapeutic interventions into any system are effective only for short term relief because these externalities are soon “neutralized” by countermoves within the system. Long term resolutions can be achieved only by restructuring and managing these systems in ways that maximize the array of “built-in” preventive strengths, with therapeutic tactics serving strictly as backups to these natural regulators. To date, we have failed to incorporate this basic principle into the mainstream of pest management science and continue to regress into a foot race with nature. In this report, we establish why a total system approach is essential as the guiding premise of pest management and provide arguments as to how earlier attempts for change and current mainstream initiatives generally fail to follow this principle. We then draw on emerging knowledge about multitrophic level interactions and other specific findings about management of ecosystems to propose a pivotal redirection of pest management strategies that would honor this principle and, thus, be sustainable. Finally, we discuss the potential immense benefits of such a central shift in pest management philosophy.  相似文献

Severe GN involves local neutrophil extracellular trap (NET) formation. We hypothesized a local cytotoxic effect of NET-related histone release in necrotizing GN. In vitro, histones from calf thymus or histones released by neutrophils undergoing NETosis killed glomerular endothelial cells, podocytes, and parietal epithelial cells in a dose-dependent manner. Histone-neutralizing agents such as antihistone IgG, activated protein C, or heparin prevented this effect. Histone toxicity on glomeruli ex vivo was Toll-like receptor 2/4 dependent, and lack of TLR2/4 attenuated histone-induced renal thrombotic microangiopathy and glomerular necrosis in mice. Anti–glomerular basement membrane GN involved NET formation and vascular necrosis, whereas blocking NET formation by peptidylarginine inhibition or preemptive anti-histone IgG injection significantly reduced all aspects of GN (i.e., vascular necrosis, podocyte loss, albuminuria, cytokine induction, recruitment or activation of glomerular leukocytes, and glomerular crescent formation). To evaluate histones as a therapeutic target, mice with established GN were treated with three different histone-neutralizing agents. Anti-histone IgG, recombinant activated protein C, and heparin were equally effective in abrogating severe GN, whereas combination therapy had no additive effects. Together, these results indicate that NET-related histone release during GN elicits cytotoxic and immunostimulatory effects. Furthermore, neutralizing extracellular histones is still therapeutic when initiated in established GN.  相似文献

Background

We present the results of open reduction and internal fixation of intra-articular calcaneal fractures with Kirschner wires with some modifications.

Material and methods

A longitudinal study was done on 37 patients who had fallen from certain heights; the study subjects were predominantly construction workers. Forty-seven fractures of the study subjects were treated using a modified approach that includes incision and some tips in operative technique. Details of modification and their evaluation on the basis of restoration of heel shape with special attention to the lateral wall, radiological assessment of Bohler’s angle, varus and valgus movement of the heel, pain while walking, patient satisfaction assessment upon return to pre-injury work are elaborated in this paper. Duration of study was from January 2000 to May 2011.

Results

The fractures healed well with good radiological and functional outcomes, and patients returned to pre-injury work status in 4–6 months.

Conclusion

The need to change to the Kocher approach for the treatment of calcaneal fracture with right-angled incision and elevation of the lateral flap is not necessary if the surgical tips showed in this paper are followed meticulously to achieve a good soft tissue outcome initially and, eventually, a good bony radiological and functional outcome. As implants used are very cheap and affordable to workers of low socioeconomic status, the authors have developed this method and presently believe that this method is best for all classes of patients with the said problem.  相似文献

Sliding wear performance of thermal spray WC-based coatings has been widely studied. However, there is no systematic investigation on the influence of test conditions on wear behaviour of these coatings. In order to have a good understanding of the effect of test parameters on sliding wear test performance of HVAF-sprayed WC–CoCr coatings, ball-on-disc tests were conducted under varying test conditions, including different angular velocities, loads and sliding distances. Under normal load of 20 N and sliding distance of 5 km (used as ‘reference’ conditions), it was shown that, despite changes in angular velocity (from 1333 rpm up to 2400 rpm), specific wear rate values experienced no major variation. No major change was observed in specific wear rate values even upon increasing the load from 20 N to 40 N and sliding distance from 5 km to 10 km, and no significant change was noted in the prevailing wear mechanism, either. Results suggest that no dramatic changes in applicable wear regime occur over the window of test parameters investigated. Consequently, the findings of this study inspire confidence in utilizing test conditions within the above range to rank different WC-based coatings.  相似文献

Genomic locations such as promoter, exon, intron, telomeric and non-telomeric regions are rich in GC-rich sequences with the potential to form G- and C-tetraplexes on both strands independently. Herein, we employed biophysical and biochemical methods to study a 34-mer C-rich DNA sequence of the myosin heavy chain β gene (MYH7β) promoter, namely HM34C for humans and the rabbit counterpart, RM34C, which differs from HM34C at three positions (three bases). Circular dichroism (CD), UV-thermal denaturation and native gel electrophoresis studies demonstrated that both the C-rich promoter segments form C-tetraplex (i-motif) structures. The CD studies revealed that HM34C forms the i-motif structure at acidic pH (5.2) in the presence of 0.1 M NaCl but remains unstructured at physiological pH. Interestingly, RM34C can form the stable i-motif structure in acidic as well as physiological pH. A shift in the positive peak from 280 nm to 275 nm with the increase in temperature from 4 °C to 30 °C was observed in temperature-dependent CD studies. UV-melting studies showed a biphasic transition for RM34C, indicating the existence of two structural species at neutral pH. In view of these findings we suggest that at physiological pH, the RM34C sequence exists in equilibrium between two structural motifs, i.e. the i-motif and homoduplex structure. This study may add to the understanding of the i-motif/homoduplex in equilibrium in physiological environments.

Background

The incidence of incisional hernias after stoma reversal is not well reported. The aim of this study was to systematically review the literature reporting data on incisional hernias after stoma reversal. We evaluated both the incidence of stoma site and midline incisional hernias.

Methods

A systematic review identified studies published between January 1, 1980, and December 31, 2012, reporting the incidence of incisional hernia after stoma reversal at either the stoma site or at the midline incision (in cases requiring laparotomy). Pediatric studies were excluded. Assessment of risk of bias, detection method, and essential study-specific characteristics (follow-up duration, stoma type, age, body mass index, and so forth) was done.

Results

Sixteen studies were included in the analysis; 1613 patients had 1613 stomas formed. Fifteen studies assessed stoma site hernias and five studies assessed midline incisional hernias. The median (range) incidence of stoma site incisional hernias was 8.3% (range 0%–33.9%) and for midline incisional hernias was 44.1% (range 8.7%–58.1%). When evaluating only studies with a low risk of bias, the incidence for stoma site incisional hernias is closer to one in three and for midline incisional hernias is closer to one in two.

Conclusion

Stoma site and midline incisional hernias are significant clinical complications of stoma reversals. The quality of studies available is poor and heterogeneous. Future prospective randomized controlled trials or observational studies with standardized follow-up and outcome definitions/measurements are needed.  相似文献
