Neurotransmitter spillover is a form of communication not readily predicted by anatomic structure. In the cerebellum, glutamate spillover from climbing fibers recruits molecular layer interneurons in the absence of conventional synaptic connections. Spillover-mediated signaling is typically limited by transporters that bind and reuptake glutamate. Here, we show that patterned expression of the excitatory amino acid transporter 4 (EAAT4) in Purkinje cells regulates glutamate spillover to molecular layer interneurons. Using male and female Aldolase C-Venus knock-in mice to visualize zebrin microzones, we find larger climbing fiber-evoked spillover EPSCs in regions with low levels of EAAT4 compared with regions with high EAAT4. This difference is not explained by presynaptic glutamate release properties or postsynaptic receptor density but rather by differences in the glutamate concentration reaching receptors on interneurons. Inhibiting glutamate transport normalizes the differences between microzones, suggesting that heterogeneity in EAAT4 expression is a primary determinant of differential spillover. These results show that neuronal glutamate transporters limit extrasynaptic transmission in a non–cell-autonomous manner and provide new insight into the functional specialization of cerebellar microzones.SIGNIFICANCE STATEMENT Excitatory amino acid transporters (EAATs) help maintain the fidelity and independence of point-to-point synaptic transmission. Whereas glial transporters are critical to maintain low ambient levels of extracellular glutamate to prevent excitotoxicity, neuronal transporters have more subtle roles in shaping excitatory synaptic transmission. Here we show that the patterned expression of neuronal EAAT4 in cerebellar microzones controls glutamate spillover from cerebellar climbing fibers to nearby interneurons. These results contribute to fundamental understanding of neuronal transporter functions and specialization of cerebellar microzones.  相似文献

Preferential looking (PL), as a technique for assessing visual acuity, was designed for use with young infants. There are occasions when a practitioner may need to use a PL test with an adult who is unable to participate in acuity measures with conventional optotype tests (such as an adult with learning disabilities, stroke damage or dementia). In preparation for the development of an adult-appropriate PL test, this study compared scores with the Cardiff Acuity Test (CAT, which was designed as a PL test) and a standard LogMAR-based test, the Lea chart. One hundred and one adults with learning disabilities, attending vision screening at Special Olympics, took part in acuity measures with both tests. Athletes subsequently found to have uncorrected refractive errors were excluded and analysis was confined to 72 athletes. There was no significant difference in mean acuity with the two tests, but CAT tended to overestimate scores for poorer acuities. CAT was less sensitive than the Lea chart to interocular acuity differences. Although CAT may be a successful way to assess acuity in patients unable to participate in conventional acuity tests, the practitioner should be cautious in interpreting results. There is the potential with CAT to overestimate acuity and to miss small interocular differences, so criteria for determining 'abnormality' may need to be adjusted.  相似文献

The enteric nervous system (ENS) controls gastrointestinal (GI) functions, including motility and digestion, which are impaired in ENS disorders. Differentiation of enteric neurons is mediated by factors released by the gut mesenchyme, including smooth muscle cells (SMCs). SMC‐derived factors involved in adult enteric neural progenitor cells (NPCs) differentiation remain elusive. Furthermore, physiologically relevant in vitro models to investigate the innervations of various regions of the gut, such as the pylorus and lower oesophageal sphincter (LES), are not available. Here, neural differentiation in bioengineered innervated circular constructs composed of SMCs isolated from the internal anal sphincter (IAS), pylorus, LES and colon of rabbits was investigated. Additionally, SMC‐derived factors that induce neural differentiation were identified to optimize bioengineered construct innervations. Sphincteric and non‐sphincteric bioengineered constructs aligned circumferentially and SMCs maintained contractile phenotypes. Sphincteric constructs generated spontaneous basal tones. Higher levels of excitatory and inhibitory motor neuron differentiation and secretion of bone morphogenic protein 2 (BMP2) were observed in bioengineered, innervated, sphincteric constructs compared to non‐sphincteric constructs. The addition of BMP2 to non‐sphincteric colonic SMC constructs increased nitrergic innervations, and inhibition of BMP2 with noggin in sphincteric constructs decreased functional relaxation. These studies provide: (a) the first bioengineered innervated pylorus and LES constructs; (b) physiologically relevant models to investigate SMCs and adult NPCs interactions; and (c) evidence of the region‐specific effects of SMCs on neural differentiation mediated by BMP2. Furthermore, this study paves the way for the development of innervated bioengineered GI tissue constructs tailored to specific disorders and locations within the gut. With rapid industrialization, there is an ever-increasing demand for iron oxides, calcium oxides, aluminum oxides, silica, and zeolites as raw materials for various industries, but reserves of such metal oxides are continuously diminishing. Therefore, there is an urgent need to explore new alternatives for such value-added minerals. One such material is incense stick ash (ISA), which is among the most unexplored byproducts from residential and holy places. Currently, ISA is of no use and it is disposed of in millions of tons (MTs) in rivers and other water bodies in India due to its sacred value. The major chemical composition of ISA is calcium, silica, alumina, ferrous minerals, magnesium, and traces of Na, K, P, Ti, etc. Major fractions of ISA, i.e., 50–60%, are made up of calcium and magnesium oxides; 20–30% of ISA is made up of silica, alumina, and ferrous minerals, as revealed by X-ray fluorescence spectroscopy (XRF). In the present research work, methods of recovery of value-added micro and nano minerals from ISA are suggested, using cost-effective techniques and an eco-friendly approach. Firstly, magnetic fractions were recovered by a magnetic separation method; then, alumina, silica, and calcium oxides were synthesized from non-magnetic fractions. The confirmation of the synthesized and extracted nanomaterials was done by Fourier transform infrared spectroscopy (FTIR), particle size analyzer (PSA), X-ray diffraction (XRD), field emission scanning electron microscopy with electron diffraction spectroscopy (FESEM-EDS), and transmission electron microscopy (TEM). The purity of synthesized particles varied from 40–80%. In the future, ISA will prove to be an alternative resource material for Fe, Ca, Si, C, Al, and zeolites, which will minimize solid waste pollution and water pollution arising due to the disposal of ISA into water bodies.  相似文献

Purpose:Marfan syndrome (MFS) is a genetic disorder associated with considerable morbidity and mortality. Presently, well-documented information on this condition is not available in India.Methods:In this retrospective cohort study, we recruited patients with clinically diagnosed MFS who presented to the outpatient department using revised Ghent nosology. We retrieved complete ophthalmic information, including vision, anterior and posterior segments, exported from electronic medical records, and relevant investigations, surgical details, and follow-up data were obtained in a specific, pretested format.Results:Our cohort consisted of 86 eyes of 43 patients and had a male preponderance. The prevalence was 20.5 per 100,000 individuals. The mean age of the patients was 23.9 years. All eyes were treated either optically with refraction or surgically using lensectomy and vitrectomy with suture supported scleral fixated intraocular lens (IOL), which significantly affected the visual outcome (P = 0.000).Conclusion:Although considered a rare disease, MFS is commonly found in the ophthalmological setting. Refraction and surgery (lensectomy with scleral fixated IOL) may improve the vision significantly.  相似文献

Potyviral coat protein (CP) and helper component-proteinase (HCPro) play key roles in both the regulation of viral gene expression and the formation of viral particles. We investigated the interplay between CP and HCPro during these viral processes. While the endogenous HCPro and a heterologous viral suppressor of gene silencing both complemented HCPro-less potato virus A (PVA) expression, CP stabilization connected to particle formation could be complemented only by the cognate PVA HCPro. We found that HCPro relieves CP-mediated inhibition of PVA RNA expression likely by enabling HCPro-mediated sequestration of CPs to particles. We addressed the question about the role of replication in formation of PVA particles and gained evidence for encapsidation of non-replicating PVA RNA. The extreme instability of these particles substantiates the need for replication in the formation of stable particles. During replication, viral protein genome linked (VPg) becomes covalently attached to PVA RNA and can attract HCPro, cylindrical inclusion protein and host proteins. Based on the results of the current study and our previous findings we propose a model in which a large ribonucleoprotein complex formed around VPg at one end of PVA particles is essential for their integrity.  相似文献

Purpose:To study the clinical presentation, mycological profile, and risk factors of fungal keratitis (FK) cases presenting at two tertiary-care centers, one each at North (Chandigarh) and Northeast (Assam) India, and to compare the spectrum of fungi recovered from the clinical and environmental samples at both locations.Methods:All patients with suspected FK were enrolled from both the centers between January 2018 and December 2019. Corneal samples were collected and processed as per standard laboratory protocols. Demographic details and clinical and mycological profiles were noted in all patients. Environmental sampling from the soil, air, and the vegetative matter was performed from both locations and neighboring districts.Results:Of the 475 suspected cases, 337 (71%) were diagnosed as FK (median age: 50 years; 77.2% males). The presence of diabetes, hypertension, blurred vision, and corneal discoloration was significantly higher in patients with FK compared to those without FK. Aspergillus sp. (52.1%) and Fusarium sp. (47.61%) were the predominant etiological agents isolated from cases in North and Northeast India, respectively. FK due to melanized fungi was associated with diabetes, trauma with animal tail, and corneal discoloration. A similar spectrum of fungi was seen in environmental and clinical samples in both the regions.Conclusion:The difference in etiological agents of FK and environmental fungal isolates in North and Northeast India highlights the need to identify the ecological niche of potential fungal pathogens. Prospective, multicenter studies, systematic environmental sampling, and the evaluation of the differences in causative agents and clinical presentation of FK from different parts of the country can substantially improve our understanding of its region-specific clinico-epidemiological profile.  相似文献
