Correction of T-cell defects by either thymic hormone treatment or thymus transplantation has proven to be more difficult clinically than historically anticipated. Because the precise action of thymic hormones is unknown and because these hormones act upon post-thymic cells, therapeutic attempts may fail owing to lack of sufficient substrate population. Results of thymic transplantation suggest that this procedure may be best utilized for the treatment of mild T-cell defects, rather than as complete replacement treatment for severe deficiency. Future clinical trials of thymic transplantation or thymic hormone appear justified in narrowly circumscribed and well-characterized conditions. 相似文献

Mouse monoclonal antibodies to Hepatitis B surface antigen(HBsAg) were prepared and their functional capabilities tested by the method of solid phase enzyme linked immuno sorbent assay(ELISA). HBsAg binding studies indicated that one monoclonal antibody 6E-1-1 bound more HBsAg at a faster rate than the other monoclonal antibodies. Also, for the binding inhibition studies with the selected monoclonal antibody 6E-1-1, one monoclonal antibody 8D-3-6 didn’t exhibit binding inhibition for HBsAg. Then, a simultaneous ELISA method was developed for the immunodiagnosis of HBsAg. Different combinations of two monoclonal antibodies as solid phase and horseradish peroxidase(HRPO) labeled phase were studied. The combination of monoclonal antibody of higher affinity constant (6E-1-1) immobilized in a solid phase and monoclonal antibody of lower affinity constant (8D-3-6) as a HRPO labeled phase was more sensitive when two monoclonal antibodies of different affinity constants for HBsAg were prepared. 相似文献

7,8-Dihydroxyflavone (DHF) is a naturally occurring flavonoid that has been reported to protect against a variety of pathologies. Chronic administration of DHF prevents high-fat diet (HFD)-induced obesity in female, but not male, mice. However, the mechanisms underlying this sexual dimorphism have not been elucidated. We have discovered that oral DHF supplementation significantly attenuates fat mass, hepatic lipid accumulation, and adipose tissue inflammation in female mice. In contrast, male mice were not protected from adiposity, and had a paradoxical worsening of hepatic lipid accumulation and adipose tissue inflammation upon DHF supplementation. Consistent with these sexually dimorphic effects on body weight and metabolic health, 7,8-DHF induced early and stable remodeling of the female intestinal microbiome. DHF supplementation significantly increased gut microbial diversity, and suppressed potentially detrimental bacteria, particularly Desulfovibrionaceae, which are pro-inflammatory and positively associated with obesity and inflammation. Changes in the female gut microbiome preceded alterations in body weights, and in silico analyses indicated that these early microbial changes were highly predictive of subsequent weight gain in female mice. While some alterations in the intestinal microbiome were also observed in male DHF-supplemented mice, these changes were distinct from those in females and, importantly, were not predictive of subsequent body weight changes in male animals. The temporality of microbial changes preceding alterations in body weight in female mice suggests a role for the gut microbiome in mediating the sexually dimorphic effects of DHF on body weight. Given the significant clinical interest in this flavonoid across a wide range of pathologies, further elucidation of these sexually dimorphic effects will aid the development of effective clinical therapies. 相似文献

目的:为研究Bio-Normalizer(BN)对大脑神经细胞缺氧损伤的保护作用,并探讨其机制。方法:本研究采用无血清体外细胞培养方法,培养鼠胚大脑神经细胞,将神经细胞造成缺氧损伤,在无血清培养液中加入不同剂量的BN,观察BN对缺氧损伤的保护作用。将细胞分为4组,Ⅰ组为缺氧BN浓度为0mg/ml组;Ⅱ组为缺氧BN浓度为0.1mg/ml组,Ⅲ组为缺氧BN浓度为0.5mg/ml组;Ⅳ组为非缺氧BN浓度为0mg/ml组。在倒置显微镜下观察细胞生长发育的情况,于细胞培养后的第4天收集细胞,测定生化指标,同时进时光镜和电镜病理检查。结果:缺氧0.1mgBN/ml组的神经细胞MTT及NSE活性均高于缺氧0mgBN/ml组,并有显著差别(P<0.05);形态学观察结果也显示缺氧0.1mgBN/ml组和0.5mgBN/ml组的神经细胞缺氧损伤后恢复明显好于缺氧0mgBN/ml组。结论:BN对大脑神经细胞缺氧损伤恢复具有明显的促进作用。 相似文献

目的 研究人工设计合成的多肽PLNG在不同作用环境条件下的跨膜运动现象。方法 体外培养不同组织来源的鼠细胞及CHO细胞、BEL细胞,用免疫荧光观察不同浓度、不同温度、不同反应时间条件下,PLNG的穿膜能力及PLNG对不同类型的细胞(CHO细胞、BEL细胞、成年大鼠肝细胞、幼大鼠肝细胞、成年大鼠心肌细胞、幼大鼠心肌细胞、成年大鼠神经细胞、幼大鼠神经细胞)的穿膜特性。结果 PLNG在不同作用环境条件下对细胞膜都有穿透作用,且进入细胞的量近乎相同。结论 实验观察到PLNG具有广谱的穿膜能力,这种穿膜能力在一定范围内对温度、时间及PLNG浓度不敏感;而且这种穿膜能力不受组织特异性的限制。 相似文献
