Associations have previously been reported between a 124-bp allele at D2S2944 and recurrent, early-onset depression in women. These earlier reports also noted but did not emphasize a possible association between this allele and alcohol-specific depression. We have analyzed the Iowa Adoption Study data to test this association. D2S2944 allele typing was available for 247 subjects from the Iowa Adoption Studies. Information on lifetime affective, alcohol, drug, and antisocial personality (ASPD) disorders was available from a structured interview. We used logistic regression to analyze adjusted and interactive D2S2944 associations with depression. Controlling other risk factors, the 124-bp allele had a strong association with DSM-IV major depression specific to those with histories of alcohol abuse/dependence and/or ASPD. (Overlap between the two prevented further specification of the interaction.) The association was not gender-specific and was not limited to alcohol-related depressive episodes. This relationship appears stronger than the D2S2944 association previously reported for early-onset depression in women. We note that, in combination with additional liabilities towards depression in women and alcoholism/sociopathy in men, the 124-bp allele may provide a basis for the phenomenon of depressive spectrum illness proposed by Winokur. 相似文献
Renal hypodysplasia (RHD) is characterized by a reduced nephron number, small kidney size, and disorganized renal tissue. A hereditary basis has been established for a subset of affected patients, suggesting a major role of developmental genes that are involved in early kidney organogenesis. Gene mutations that have dominant inheritance and cause RHD, urinary tract anomalies, and defined extrarenal symptoms have been identified in TCF2 (renal cysts and diabetes syndrome), PAX2 (renal-coloboma syndrome), EYA1 and SIX1 (branchio-oto-renal syndrome), and SALL1 (Townes-Brocks syndrome). For estimation of the prevalence of these events, an unselected cohort of 99 unrelated patients with RHD that was associated with chronic renal insufficiency were screened for mutations in TCF2, PAX2, EYA1, SIX1, and SALL1. Mutations or variants in the genes of interest were detected in 17 (17%) unrelated families: One mutation, two variants, and four deletions of TCF2 in eight unrelated patients; four different PAX2 mutations in six families; one EYA1 mutation and one deletion in two patients with branchio-oto-renal syndrome; and one SALL1 mutation in a patient with isolated RHD. Of a total of 27 patients with renal cysts, six (22%) carried a mutation in TCF2. It is interesting that a SIX1 sequence variant was identified in two siblings with renal-coloboma syndrome as a result of a PAX2 mutation, suggesting an oligogenic inheritance. Careful clinical reevaluation that focused on discrete extrarenal symptoms and thorough family analysis revealed syndrome-specific features in nine of the 17 patients. In conclusion, 15% of patients with RHD show mutations in TCF2 or PAX2, whereas abnormalities in EYA1, SALL1, and SIX1 are less frequent. 相似文献

2. 2. [3H]PCP labels a single class of site in rat brain (kd = 46 nM; Bmax = 10.5 fold/slice). Ligand selectivity pattern strongly suggests that [3h]pcp binds to sites relevant for its pharmacological actions.
3. 3. Chronic PCP treatment (10 mg/kg/day for 14 days) decreases the number of sites (Bmax) for [3H]PCP and [3H]spiperone binding but not for [3H]dihydromorphine. These modifications could be related to the development of tolerance and dependence to PCP.
4. 4. Visualization of [3H]PCP binding sites shows high densities of receptors in cortical areas and hyppocampus. Lower densities are observed in caudate-putamen, nucleus accumbens, and amygdala. Negligible quantities of receptors are seen in brain stem and over white matter.
5. 5. The presence of specific [3h]pcp binding sites in rat brain suggests the possible existence of an endogenous ligand for this unique receptor.
Research has shown insecure attachment style is associated with ineffective emotional regulation leading to maladaptive behaviors in adulthood. In the present study, we examined the association between attachment style and illicit substance use within a sample of adoptees (n=148). It was predicted that insecure attachment style would be associated with a higher incidence of lifetime illicit substance use and that perceived social support would mediate this association. Logistic regression analyses showed higher prevalence of illicit substance use among both insecure attachment groups as compared to the secure group. No difference was found between the two insecure types. Perceived social support was found to mediate the association between attachment style and illicit substance use for the insecure-preoccupied group only. The findings from the present study further implicate attachment style in the risk for illicit substance use, as well as preventions designed to identify those at risk for use. 相似文献
A dengue vaccine effective against all four serotypes is urgently needed. However, safety and immunogenicity could be affected by prior exposure to flaviviruses. This open, controlled, phase IIa study was conducted in 35 healthy adults who had received monovalent, live attenuated Vero cell-derived dengue vaccine against dengue virus 1 (VDV1) or 2 (VDV2) or yellow fever (YF) vaccine 1 year before or who were flavivirus-naïve. All participants received one subcutaneous injection of tetravalent dengue vaccine (TDV) and were followed for 180 days. Previous vaccination did not increase reactogenicity, laboratory abnormalities, or incidence of vaccine viremia, but it did increase the neutralizing antibody response to dengue virus that persisted at day 180. There was no increase in YF antibodies in participants previously immunized with YF vaccine. Prior exposure to YF or monovalent dengue vaccines had no adverse effects on the safety or incidence of viremia associated with this TDV, but it increased immunogenicity. 相似文献
There is no confirmed strategy for treating painful bladder syndrome/interstitial cystitis (PBS/IC) with unclear etiology. Therefore, a pilot study was carried out to evaluate the efficacy and safety of hyperbaric oxygen (HBO) therapy in treatment-resistant PBS/IC patients.
Methods
HBO treatment (2.0 ATA for 60 minutes/day × 5 days/week for 2 or 4 weeks) was performed on 11 patients with severe symptoms that had not been improved by previous therapy regimens between December 2004 and July 2009.
Results
Seven of the 11 patients demonstrated persistent improvement in symptoms during the 12 months after HBO treatment. These responders demonstrated a decrease in the pelvic pain scale and urgency scale from 7.7 ± 1.0 and, 6.6 ± 0.9 to 3.4 ± 2.5 and 4.3 ± 2.4 after 12 months, respectively (p < 0.05). The total score of the interstitial cystitis symptom index and 24-hour urinary frequency demonstrated a significant sustained decrease from the baseline. Two responders, who received an additional course of HBO 12 and 13 months after initial treatment, respectively, did not suffer impairment for more than two years. There was one case of transient eustachian tube dysfunction and three cases of reversible exudative otitis media as a consequence of HBO treatment.
Conclusions
HBO is a potent treatment for PBS/IC patients resistant to conventional therapy. It was well tolerated and provided maintained amelioration of pain, urgency and urinary frequency for at least 12 months. 相似文献
