From emf values of galvanic cells of the types {Na-glass  NaCl (mB), HZ± (mA)  AgCl(S)  Ag}; {glass-H  HCl (mC=0.01 mol kg?1), HZ± (mA), NaCl (mB)  AgCl(S)  Ag} and {glass-H  HZ± (mA), NaZ (mD=0.09982 mol kg?1), NaCl (mB)  AgCl(S)  Ag} (where HZ± stands for dl-α-amino-n-butyric acid), activity coefficients for NaCl and the aminoacid were determined, at various relative concentrations of both compounds, in the ionic strength range of 0.1–4 mol kg?1 and 298.15 K. Likewise, activity coefficients for both the pure aminoacid hydrochloride (up to 0.5 mol kg?1) and the sodium salt of the aminoacid (up to 0.1 mol kg?1) were also determined, together with the thermodynamic ionisation equilibrium constants for these latter compounds.  相似文献

OBJECTIVE: Dietary intake of omega-3 polyunsaturated fatty acids (PUFA) like alpha-linolenic acid (ALA) is antiarrhythmic and cardioprotective. PUFA may also be beneficial in hypertension. Altered Na(+)-Ca(2+) exchanger (NCX) activity has been implicated in arrhythmias, hypertension and heart failure and may be a target for PUFA. Thus, we tested the effects of ALA and other distinct fatty acids on the cardiac (NCX1.1) and vascular (NCX1.3) NCX isoforms. METHODS: HEK293 cells stably expressing NCX isoforms were ramped from +60 to -100 mV (over 1600 ms) in the absence and presence of 25 microM oleic acid (OA, omega-9), linoleic acid (LA, omega-6), ALA (omega-3), or eicosapentaenoic acid (EPA, omega-3). NiCl(2) (5 mM) was used to inhibit and therefore identify the NCX current. The effect of 25 microM ALA on NCX1.1 and NCX1.3 activity was also assessed in adult rat ventricular cardiomyocytes and rabbit aortic vascular smooth muscle cells (VSMC) by measuring [Ca(2+)](i) following substitution of [Na(+)](o) with Li(+). RESULTS: Application of Ni(2+) had no effect in non-transfected cells. ALA and EPA (25 microM) reduced the Ni(2+)-sensitive forward NCX1.1 current (at -100 mV) by 64% and reverse current (at +60 mV) by 57%, and inhibited the Ni(2+)-sensitive NCX1.3 forward and reverse currents by 79% and 76%, respectively. Neither OA nor LA (25 microM) affected the NCX1.1 currents, but both partially inhibited the forward and reverse mode NCX1.3 currents. Inhibition of NCX1.3 by ALA occurred at a much lower IC(50) ( approximately 19 nM) than for NCX1.1 ( approximately 120 nM). In cardiomyocytes and VSMC, ALA significantly reduced the Li(+)-induced rise in intracellular [Ca(2+)]. CONCLUSIONS: NCX1.3 is more sensitive to inhibition by ALA than NCX1.1. In addition, only omega-3 PUFA inhibits NCX1.1, but several classes of fatty acids inhibit NCX1.3. The differential sensitivity of NCX isoforms to fatty acids may have important implications as therapeutic approaches for hypertension, heart failure and arrhythmias.  相似文献

The palpation and enucleation of occult insulinomas (less than 15 mm) can be a difficult surgical problem even with good arteriographic localization. In the authors' limited experience, confirmation of arteriographic findings by pancreatic venous sampling provided little additional localizing information. However, if arteriography is negative or equivocal, venous sampling can indicate the segment of pancreas to be "blindly" resected if the adenoma is not palpable. Venous sampling may be misleading in polyendocrine syndromes because of the frequency of multiple adenomas and variable hormone production.  相似文献

Clinical and molecular similarities between canine mammary tumours and human breast cancer have been described in recent decades. Clinically, the similarities are very strong: spontaneous tumours, hormonal aetiology, age of onset and an identical course of the disease. The clinical characteristics that have an impact on the clinical outcome are also identical: tumour size, lymph node invasiveness and clinical stage. Nowadays, as far as human medicine is concerned, the goal is to identify prognostic factors, mainly at the molecular level, such as those involved in metastasis, which could be used as therapeutic targets to support a better outcome. Moreover, in this area, canine mammary tumours seem to mimic human breast cancer, as a range of similarities are found at the molecular level concerning the overexpression of steroid receptors, proliferation markers, epidermal growth factor, p53 supressor gene mutations, metalloproteinases, cyclooxygenases, among many others. Clinical and molecular data that support canine mammary tumours as a model to study human breast cancer are analysed in this review. Additionally, it is shown that some recent molecular targets in canine mammary tumours may be seen as indicators for similar research to be performed in the corresponding human disease.  相似文献

A case-control study was performed to determine whether an association exists between cancellation of ambulatory cataract surgery in adults, preoperative test results, and systemic diseases present at clinical examination. Among 500 adults who were to undergo ambulatory cataract surgery, cancellation of surgery due to medical events was not associated with results of preoperative testing (hemoglobin, serum glucose, and electrocardiogram). The rate of surgery cancellation was higher only in patients having a history of arterial hypertension. Presented at the 1999 Joint Meeting of the American Academy of Ophthalmology and the Pan-American Association of Ophthalmology, Orlando, Fla, 1999. Intravenous digital subtraction renal angiography (DSRA) has been compared with conventional angiography only in small, selected series of hypertensive patients. The authors prospectively examined with intravenous DSRA 94 patients at increased risk for renovascular hypertension and compared these studies with conventional angiography. A stenosis of at least one main renal artery was identified with intravenous DSRA in 22 patients and confirmed in 20 patients. No significant stenoses were seen with conventional angiography in any of the 64 patients in whom lesions were not seen with intravenous DSRA. Since inadequate DSRA studies were considered positive for renal artery stenosis, the sensitivity of intravenous DSRA was 100% (25 of 25); specificity, 93% (64 of 69); positive predictive value, 83% (25 of 30); and negative predictive value, 100% (64 of 64). The authors conclude that intravenous DSRA is a sensitive test for identifying stenosis of the main renal arteries and is appropriate to use as a screening test among patients at increased risk for renovascular hypertension.  相似文献

Cyclosporin nephrotoxicity in heart and lung transplant patients   总被引:1,自引:0,他引:1  
Twenty-two patients with heart, lung or heart and lung transplants maintained on cyclosporin for periods ranging from 3 months to 10 years developed renal insufficiency which was investigated by renal biopsy. The histopathological changes were: (i) severe vascular and glomerular damage due to thrombotic microangiopathy (TM); (ii) a form of focal segmental glomerulosclerosis (FSGS); (iii) glomerular ischaemia. Rather than being separate entities, these changes appeared to represent a spectrum of pathology, some biopsies showing all three forms of glomerular injury. In all cases the glomerular changes were accompanied by arteriolar and arterial pathology, and we identified novel ultrastructural changes in the arteriolar endothelial basal lamina. Tubular atrophy was a consistent feature, the severity of which reflected the severity of the glomerular sclerosis, and which appeared to be a consequence of glomerular loss. Our findings are consistent with the nephrotoxic effects of cyclosporin being mediated chiefly via damage to preglomerular vessels and glomerular capillary endothelium. From an analysis of the clinical aspects of these cases, the effects of cyclosporin appear to be to some extent idiosyncratic, and therefore not entirely preventable, but strict monitoring of blood cyclosporin levels is essential to minimize the risk of permanent renal damage. Monitoring urinary protein in addition to plasma creatinine may detect the onset of FSGS, as proteinuria precedes creatinine elevation.   相似文献

Antigen recognition by clonotypic B cell receptor (BcR) is the first step of B lymphocytes differentiation into plasmocytes. This B cell function is dependent on efficient major histocompatibility complex (MHC) class II-restricted presentation of BcR-bound antigens. In this work, we analyzed the subcellular mechanisms underlying antigen presentation after BcR engagement on B cells. In quiescent B cells, we found that MHC class II molecules mostly accumulated at the cell surface and in an intracellular pool of tubulovesicular structures, whereas H2-M molecules were mostly detected in distinct lysosomal compartments devoid of MHC class II. BcR stimulation induced the transient intracellular accumulation of MHC class II molecules in newly formed multivesicular bodies (MVBs), to which H2-M was recruited. The reversible downregulation of cathepsin S activity led to the transient accumulation of invariant chain-MHC class II complexes in MVBs. A few hours after BcR engagement, cathepsin S activity increased, the p10 invariant chain disappeared, and MHC class II-peptide complexes arrived at the plasma membrane. Thus, BcR engagement induced the transient formation of antigen-processing compartments, enabling antigen-specific B cells to become effective antigen-presenting cells.  相似文献
