Electron microscopic studies of the DNA of defective and standard pseudorabies virions.

DNA from standard and defective virus particles was analyzed by electron microscopy. Upon partial denaturation the DNA molecules from standard pseudorabies virus showed distinct denaturation patterns. The molecules are asymmetrical with respect to adenine-thymidine (A-T) content, having several A-T-rich regions on the “left” side and only one on the “right” side of the molecule. The fact that a distinct denaturation map was obtained indicates that the molecules are not circularly permuted. On the extreme left of the molecule a segment of approximately 0.5 μm invariably contained denatured regions, whereas this was not evident on the right side of the molecule. Thus, if the molecule is terminally redundant, the redundant region covers a relatively small part of the viral genome. The contour length of the DNA molecules present in defective, interfering particles was found to be similar to that of standard DNA. Under partial denaturation conditions a distinct segment of DNA (1.1 μm in length) was found to be tandemly repeated in defective DNA. The number of repeats in different molecules varied. In some of the DNA molecules as many as 17 consecutive repeats were found, whereas in others no repeats were seen. Repeated segments were not detected in standard DNA.     

An ultrastructural study of normal human mammary epithelial cells in culture

The ultrastructure of normal human mammary cells cultured from post-weaning breast fluids is described. Cells from confluent monolayers in two week old cultures were studied. The epithelial nature of these cells was established by the demonstration of a well developed system of cell-to-cell interdigitation and numerous desmosomes. These cells also share with breast epithelial cells in vivo, polarity, with blunt short microvilli on the apical surface and an oriented arrangement of organelles in the basal and apical portions of the cells. The Golgi apparatus, which is the most highly developed organelle, is localized in the apical pole and contains substantial quantities of secretory material in the cisternae and vesicles. A variegated palisade of finely granular material mixed with tonofilaments is seen in the basal portion of the cells; many of these tonofilaments end in the terminal web of the desmosomes. The regular occurrence of these cells in breast fluids during the terminal phases of lactation suggests that their separation is a part of normal breast involution.     

Pseudorandom testing technique for the characterization of local hemodynamic control

Summary In order to investigate the low frequency properties of renal and femoral hemodynamic variables, pseudorandom testing techniques were used. The arterial flow of each bed, in separate experiments, was modulated by a low amplitude signal based on a pseudorandom binary sequence (PRBS) generated by digital computer.The cross-correlation functions between input flow and arterial pressure, venous pressure, and venous flow exhibit damped oscillations in all cases. These responses are parameterized in terms of a damping ratio () and an undamped natural frequency _n for a second order model. The parameters of the model are dependent upon the state of the bed as defined by mean arterial and venous pressures, mean flow through the bed, resistance, and oxygen consumption.The results of this study offer further insight into the dynamic low frequency autoregulation phenomenon for the renal and femoral beds of the dog.Supported by NIH Grant HE 11747.     

In vitro stimulation by prostate extracts of rat ventral prostate stromal and epithelial cell division

We examined the effect of prostate cell extracts on the replication of highly enriched rat ventral prostate stromal and epithelial cells cultured in RPMI-1640. Extracts from normal rat prostates completely inhibited cell division in both fractions, while a 10% (v/v) extract of Dunning R-3327G adenocarcinoma inhibited replication of stromal cells but permitted that of epithelial cells. The cytotoxic effect of prostate extracts was dialyzable, heat stable, unaffected by proteases, soluble in acid/alcohol, and insoluble in chloroform. These properties are consistent with those of polyamines incubated in the presence of fetal calf serum. Dialyzed extracts from Dunning adenocarcinomas, and from human benign hypertrophic and carcinomatous prostates stimulated rat prostate stromal and epithelial cell division, in keeping with other reports of "growth factors" in prostate tissue. This mitogenic activity was stable to temperature (70 degrees C, 4 hours), and marginally if at all affected by exposure to trypsin or to pronase coupled to Sephadex. Short-term culture of separated prostate cells should provide a useful assay system for detecting putative autocrine or paracrine stromal and/or epithelial cell growth factors and identifying suspected homo- or heterotypic cellular interactions in normal and diseased prostates.     

The physician's role in an aging America: taking the team approach to eldercare

With the advent of an aging patient population, the healthcare community is sharpening its focus on geriatric medicine and on how current resources will fare under the demands of an aging America. One approach to effective eldercare is that of a "team" of physician and nonphysician providers, cooperating to meet the multifaceted needs of the elderly patient. As the traditional primary care provider, the physician is a natural choice for the role of team leader.
