Nearly 70-75% of patients with localized juvenile periodontitis (JP) have abnormal polymorphonuclear leukocytic (PMN) chemotaxis. The objective of this study was to determine whether the lower chemotactic response in PMNs from JP patients is associated with a defect in intracellular signal transduction, as measured by stimulus-induced changes in free cytosolic calcium (Ca2+) mobilization. We report that peptide chemoattractants such as N-formyl-methionyl-leucyl-phenylalanine (fMLP) and the complement fragment C5a in direct comparative studies induced lower amounts of initial Ca2+ mobilization in PMNs from JP patients than healthy controls, as monitored by intracellular fura-2 fluorescence. The initial resting levels of free cytosolic Ca2+ in PMNs from JP patients and normal individuals were found to be similar. fMLP and C5a both mobilized Ca2+ in PMNs in a dose-dependent manner. Treatment of PMNs from 0.16 to 20 nM fMLP and 0.2 to 20 nM C5a resulted in elevated levels of free cytosolic Ca2+. However, above 20 nM fMLP and 5 nM C5a concentrations the extent of total Ca2+ mobilization did not differ significantly. Although fMLP and C5a caused Ca2+ mobilization in PMN cells from JP and healthy control subjects, fMLP stimulation induced higher levels of free cytosolic Ca2+ mobilization in PMN cells from healthy control subjects (141.29 +/- 25.55 nM/2 x 10(6) PMNs), than PMNs from JP patients (62.33 +/- 23.76 nM/2 x 10(6) PMNs). Similarly C5a induced higher levels of Ca2+ mobilization in PMNs from healthy control individuals (130.43 +/- 18.26 nM Ca2+/2 x 10(6) PMNs)O, when compared to JP patients (49.92 +/- 14.92 nM Ca2+/2 x 10(6) PMNs).(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Such stimulation of rat parotid acinar cells in vitro modulated the rate of processing of N-linked oligosaccharides in a high-molecular weight (220 kdalton) secretory glycoprotein. Conversion of polymannose-type oligosaccharides to complex-type oligosaccharides was evaluated by sensitivity to endoglucosaminidase H and alpha-mannosidase, and with a specific inhibitor of glucosidases I/II. Oligosaccharide maturation in the 220 kdalton glycoprotein required one-third to half less time in cells exposed to the beta-adrenergic agonist isoproterenol than in controls.  相似文献

The enamel-covered primary and secondary coronal dentine in the molars of 90-day-old and 360-day-old rats was examined using microradiography. Some preparations were subsequently examined by transmission and scanning electron microscopy (SEM). Fractured dentine surfaces and methacrylate casts of the tubular system in the primary dentine were also examined with SEM. No microradiographic evidence of a hypermineralized peritubular matrix, such as that seen in man and other species, was seen in either young or old rats. Transmission and SEM confirmed the microradiographic findings. The tubule obliteration and extensive intra-luminal mineral deposits which have been reported in the enamel-free coronal dentine of the rat molar were not seen in the enamel-covered coronal dentine but some evidence of tubule obliteration was seen in the secondary dentine.  相似文献

A prospective four-year-study of 945 fractures of the facial skeleton is presented. Details of facial trauma were collected in patients referred from the Bradford and Airedale Districts of West Yorkshire, England, an area with a population slightly in excess of half a million. Statistical analysis shows variations from previously published papers, the male to female ratio being under 2.6:1, with an average of 1.15 fractures per patient. A relatively high incidence of sports fractures is reported but the number of industrial accidents is surprisingly low. The majority of patients seen were in their third decade, the mean age being just over 25 years. The analysis also reveals that a high percentage of fractures were treated conservatively.  相似文献

Adenyl cyclase.     
Certain hormones regulate the activity of their target cells by stimulating adenyl cyclase, which is an enzyme located within the target cell's plasma membrane. Adenyl cyclase catalyzes the formation of cyclic AMP, which is released into the cell and modulates cell functions. In this communication the characteristics of adenyl cyclase are reviewed. The coupling between hormone receptors and this enzyme is discussed as is the ability of agents such as hormones, ATP, magnesium, calcium, guanine nucleotides and prostaglandins to alter cyclase activity. Several diseases that result from derangements of the adenyl cyclase system are known and the molecular bases for these diseases are discussed in this review.  相似文献
