目的:调查分析就业与未就业大学生的人格特质差异及就业的大学生人格特质是否有性别差异。方法:①纳入某医科大学临床、检验、放射、口腔、高护等专业毕业已就业的大学生和未就业的大学生。于2005-07用随机抽样的方式,选取其中录取的202名,其中男生91名,女生111名。未就业的大学生204名,其中男103名,女101名。纳入对象均健康,且对调查项目知情意。②采用加利福尼亚心理调查表进行个体水平、人格特征和自我实现倾向测试。该量表共440个测试题,20个分量表,3个结构量表。主要包括4大因素。人际关系和适应性,包括支配性、进取能力、社交性、社交风度、自我接受;社会化成熟感、责任心和价值感,包括责任心、社会化、自我控制、好印象、同众性、适意感、宽容性;成就潜能和智力水平,包括顺从成就、独立成就、智力效率;个人生活态度和倾向,包括心理感受性、灵活性、女性化/男性化。另外还有2个通俗概念量表即独立性、通情。3个结构量表内外向、常规趋向和常规异向和自我实现。分数越高表明心理素质越适应就业。③计量资料间差异性比较采用t检验。结果:共发出问卷406份,回收填写完整、准确的答卷406份,回收率100%。①就业与未就业大学生加尼福尼亚心理调查表分量表评分比较:就业大学生加尼福尼亚心理调查表分量表支配性、进取能力、社交性、社交风度、自我接受、独立性、通情、责任心、社会化、自我控制、好印象、同众性、适意感、宽容性、顺从成就、独立成就、智力效率、心理感受性评分明显高于未就业大学生(t=3.81~12.62,P<0.01)。②就业男女大学生加尼福尼亚心理调查表分量表评分比较:差异不明显(t=0.04~1.73,P>0.05)。③加尼福尼亚心理调查表结构量表评分比较:就业大学生常规趋向和常规异向和自我实现评分明显高于未就业大学生(t=5.00~10.06,P<0.01)。结论:毕业后能够就业的大学生人格特征较为成熟,较未就业大学更具备适应就业的心理素质和人格特质,此种人格特质无性别差异。 相似文献

新型大肠杆菌-分枝杆菌穿梭载体的构建及卡那霉素抗性基因表达的研究 总被引:1,自引:0,他引:1
用 PCR(polymerase chain reaction)技术克隆了卡介苗(Bacille Calmette-Guerin,BCG)热休克蛋白 65(heat shock Protein 65, hsp65)启动子序列,构建成穿梭载体ME-65。该载体含有分枝杆菌和大肠杆菌的质粒复制基因、BCG的hsp65高效启动子序列以及在分枝杆菌和大肠杆菌中都能表达的卡那霉素抗性基因。将其转化到耻垢分枝杆菌和BCG中均可稳定复制并表达卡那霉素抗性。质粒对宿主菌的生长无明显的影响,未观察到质粒的丢失和突变。该载体具有分子量小、容量大、转化效率高、复制稳定、含有多克隆位点等优点,将外源基因插入到hsp65启动子的下游,可望在BCG中获得高效表达。为BCG和其它分枝杆菌发展成多价重组疫苗开辟了新途径。 相似文献

Summary The BALB/c mice were immunized with Hsp70 DNA and Hsp65 DNA vaccines in humanMycobacterium tuberculosis. Eight weeks after immunization, the eyeballs were removed, blood and spleen taken, and intraperitoneal macrophages were harvested.
The lymphocytic stimulating index (SI) was used to measure the cellular proliferating ability and NO release to measure the
phagocytic activity of the macrophages. With ELISA kit, the levels of interleukin-2 (IL-2) and interferon-γ (IFN-γ) in serum
and the splenic lymphocytic cultured supernatant were detected. The results showed that after the mice were immunized with
100 μg/mouse of Hsp70 DNA vaccine intramuscularly, the splenic lymphocytic proliferating ability in the mice was significantly
increased as compared with that in the control group, vector group and Hsp65 DNA vaccine group (P<0.01); The contents of NO in the intraperitoneal macrophages of the mice were significantly lower than in the control group
and Hsp65 DNA vaccine group (P<0.01); The levels of serum IL-2 in the mice were significantly higher than in the control group, but there was no statistical
difference between Hsp65 DNA group and vector group (P>0.05); The contents of serum IFN-γ in the mice were significantly higher than in the control group, but significantly lower
than in the Hsp65 DNA vaccine group (P< 0.05). It was indicated that immunization with Hsp70 DNA vaccine could obviously enhance the immune response, but its intensity
seemed inferior to Hsp65 DNA vaccine. The anti-infection mechanisms and clinical use in the future of the vaccines of Hsp70
DNA and Hsp65 DNA are worth further studying.

卡介苗(BCG)是全球应用最广泛的疫苗,也是一种能携带多种致病菌保护性抗原表达的疫苗载体。本研究以pUC19为骨架,分别克隆入BCG分泌抗原85-B的调节和信号肽序列、Neo基因序列(编码卡那霉素抗性)以及分枝杆菌质粒复制基因,构建成分枝杆菌-大肠杆菌穿梭质粒pBCG-8000。该重组质粒在分枝杆菌(含BCG)、大肠杆菌两个系统中均能稳定复制,并能稳定表达卡那霉素抗性。本研究为把BCG和其它分枝杆菌发展成为多价疫苗载体打下了基础。 相似文献

To better understand the prevalence of Gallibacterium anatis in different poultry species, a rapid and accurate method was developed to detect G. anatis using a TaqMan fluorescent quantitative polymerase chain reaction (qPCR). Specific primers and a TaqMan probe were designed based on the reference gtxA gene sequence. The qPCR standard curve showed a good linear relationship, and the method showed good reproducibility, sensitivity, and specificity, indicating its suitability for G. anatis identification and quantitative analysis. A comparison of the detection results in 160 clinical swab samples showed that the detection rate (54.4%) of the qPCR for G. anatis was better than that of two conventional methods: gyrB gene-based qPCR for G. anatis (51.9%) and culture-based identification (34.4%). G. anatis was detected in layer chicken (77.3%), Silkie chicken (72.7%), and duck (27.1%) with relatively high detection rates, whereas dove (8.8%) and quail (3.0%) showed lower detection rates, indicating the different prevalence of G. anatis in different fowl species. 相似文献
