Ribosome crystal structures have revealed that two small subunit proteins, S9 and S13, have C-terminal tails, which, together with several features of 16S rRNA, contact the anticodon stem-loop of P-site tRNA. To test the functional importance of these protein tails, we created genomic deletions of the C-terminal regions of S9 and S13. All of the tail deletions, including double mutants containing deletions in both S9 and S13, were viable, showing that Escherichia coli cells can synthesize all of their proteins by using ribosomes that contain 30S P sites composed only of RNA. However, these mutants have slower growth rates, indicating that the tails may play a supporting functional role in translation. In vitro analysis shows that 30S subunits purified from the S13 deletion mutants have a generally decreased affinity for tRNA, whereas deletion of the S9 tail selectively affects the binding of tRNAs whose anticodon stem sequences are most divergent from that of initiator tRNA.  相似文献

Dengue is highly endemic in southern Vietnam and all four serotypes of dengue virus have already been identified. To determine the age-specific prevalence of dengue and associated risk factors, we conducted a serological study at two primary schools and assessed risk factors by analysing children's questionnaires and household surveys. Sera were collected from 961 primary schoolchildren in Binh Thuan Province and tested for the presence of dengue virus serum antibodies using an indirect immunoglobulin G (IgG) enzyme-linked immunosorbent assay (ELISA). The antibody prevalence of the total population was 65.7% (n=631) which increased from 53.0 to 88.2% with age. The annual incidence of a first dengue infection, estimated by binary regression of the seroprevalence by age, was 11.7%. Interestingly, the prevalence of dengue IgG antibodies was significantly higher in children who confirmed using a pit latrine (RR 1.467, 95% CI: 1.245-1.730) and whose domestic environment contained discarded cans (RR 1.238, 95% CI: 1.042-1.470) and pigs (RR 1.228, 95% CI: 1.002-1.504). The epidemiology of dengue in southern Vietnam is stable with a constantly high annual incidence of first infections. Transmission occurs mainly peri-domestically, which has important public health implications.  相似文献

Lipid accumulation has been described in the duodenal lamina propria of human immunodeficiency virus (HIV)-infected patients with diarrhea and malabsorption. Using light and electron microscopy, we studied duodenal biopsies obtained from 54 consecutive HIV-infected patients by means of upper gastrointestinal endoscopy after an overnight fast. The presence of diarrhea and weight loss were recorded, and all the patients had standard stool study for ova, parasites, and bacteria. Serum levels of albumin, triglycerides, and cholesterol were obtained within one week of the endoscopy. Fecal fat and fecal 1-antitrypsin clearance were measured in 11 patients. Lipid accumulation was observed in nine patients (16.6%). Fat droplets were seen in enterocytes, in their basolateral membrane spaces, and in the lamina propria. The mean serum levels of triglycerides (1.85±0.20 mmol/liter) and cholesterol (2.81±0.30 mmol/liter) were significantly lower in the patients with enteric steatosis than in patients without this anomaly (respectively, 3.38±0.39 and 3.97±0.18 mmol/liter,P<0.001P<0.01). The mean amount of fecal fat in the three patients with lipid accumulation (16±1.60 g/24 hr) was significantly larger than in the eight patients without lipid accumulation (4.50±0.62 g/24 hr,P<0.01). These findings suggest that fat malabsorption in HIV-infected individuals is due to a blockage of transport through the duodenal mucosa. The frequency of diarrhea, weight loss, or identified enteric pathogens did not differ significantly between patients with and without enteric steatosis. Both the etiology and the pathophysiology of these alterations remain to be documented.  相似文献

In Vietnam, dengue hemorrhagic fever has been detected since the 1950s. In Southeast Asia, urban centers expanded rapidly in an uncontrolled and unplanned way. The Aedes aegypti populations and dengue viruses thrived in these new ecological and demographic settings. The result of these changes was a greatly extended geographic distribution, increased densities of Ae. aegypti and the maintenance of the four dengue serotypes leading to a dramatic increase in dengue transmission. To assess the role of the vector in the changing pattern of the disease in Southeast Asia, we studied the ecology of Ae. aegypti, genetic differentiation, variability in competence as a vector for dengue 2 virus, and resistance to insecticides.  相似文献

We report a simple, inexpensive and user-friendly capillary viscometer based on the measurement of pressure drop in capillary tubing using the principle of ideal gas law. Viscosity is an important physical property of a fluid that provides molecular information of the fluid''s behavior under flow conditions. Measuring viscosity, however, generally requires relatively large fluid volume samples and is expensive with commercial viscometers. Microfluidic viscometers at different levels of complexity can measure fluids at different flow rates with a small sample volume but the cost of commercially available microfluidic viscometers is still high. The reported capillary viscometer is cost-effective, uses small amounts of sample fluid and can measure viscosity under various shear rates. According to the Hagen–Poiseuille equation, the pressure drop of laminar flows in a capillary at a given flow rate is proportional to the viscosity of the fluid. When an enclosed air volume is connected to the upstream of the capillary, the pressure drop can be calculated with the change of the connected air volume, which is reflected by the displacement change of the air–liquid interface in the connecting capillary to the enclosed air volume. Based on these principles, the viscometer was assembled with readily accessible materials, and required no internal sensors or extensive programming. Measurements were successfully performed for five liquids including water, acetone, 2% fat milk, glycerin 30% and glycerin 40%. Except for acetone, the difference between measured and known viscosity was within 4% and highly consistent, well within the 13% uncertainty errors of readily accessible laboratory materials. Overall, the simple viscometer was easily assembled with low cost materials, was portable and accurate, and provided an alternative to expensive commercial viscometers. Finally, the simple capillary viscometer was a good outreach project for K-12 students to understand fluid behavior.

A Cu2(OBA)2(BPY) metal–organic framework was utilized as a productive heterogeneous catalyst for the synthesis of 3-aroylquinolines via one-pot domino reactions of 2-aminobenzylalcohols with propiophenones. This Cu-MOF was considerably more active towards the one-pot domino reaction than a series of transition metal salts, as well as nano oxide and MOF-based catalysts. The MOF-based catalyst was reusable without a significant decline in catalytic efficiency. To the best of our knowledge, the transformation of 2-aminobenzylalcohols to 3-aroylquinolines was not previously reported in the literature, and this protocol would be complementary to previous strategies for the synthesis of these valuable heterocycles.

This work aimed to investigate the application of a solid acid catalyst, a replacement for mineral acids or enzymes, to biomass conversion for further applications. Sulfonated zeolite, HSO3-ZSM-5, was successfully synthesized and characterized by several analysis techniques. The obtained catalyst showed high activity and efficiency in the hydrolysis of pretreated corn cob. Moreover, the acidity of the zeolite product positively influenced the biomass conversion. The influences of reaction parameters such as catalyst loading, reaction time and temperature on the hydrolysis were also established. Under suitable conditions, a hydrolysis yield of ∼54% was achieved. This recyclable solid acid catalyst provided a promising potential for applications in many industrially important hydrolysis processes of biomass.
