A case of persistent bloodstream infection with Kocuria rhizophila related to a damaged central venous catheter in a 3-year-old girl with Hirschsprung's disease is reported. The strain was identified as K. rhizophila by 16S rRNA gene sequencing and matrix-assisted laser desorption ionization-time of flight mass spectrometry. Arbitrarily primed PCR analysis showed a clonal strain. The repeated septic episodes were resolved with the catheter repair.  相似文献

A single-step multiplex PCR (here referred to as a duplex PCR) has been developed for simultaneous detection and diagnosis of Fasciola hepatica and F. gigantica. These species overlap in distribution in many countries of North and East Africa and Central and Southeast Asia and are similar in egg morphology, making identification from fecal samples difficult. Based on a comparative alignment of mitochondrial DNA (mtDNA) spanning the region of cox1-trnT-rrnL, two species-specific forward primers were designed, FHF (for F. hepatica) and FGF (for F. gigantica), and a single reverse primer, FHGR (common for both species). Conventional PCR followed by sequencing was applied using species-specific primer pairs to verify the specificity of primers and the identity of Fasciola DNA templates. Duplex PCR (using three primers) was used for testing with the DNA extracted from adult worms, miracidia, and eggs, producing amplicons of 1,031 bp for F. hepatica and 615 bp for F. gigantica. The duplex PCR failed to amplify from DNA of other common liver and intestinal trematodes, including two opisthorchiids, three heterophyids, an echinostomid, another fasciolid, and a taeniid cestode. The sensitivity assay showed that the duplex PCR limit of detection for each Fasciola species was between 0.012 ng and 0.006 ng DNA. Evaluation using DNA templates from 32 Fasciola samples (28 adults and 4 eggs) and from 25 field-collected stools of ruminants and humans revealed specific bands of the correct size and the presence of Fasciola species. This novel mtDNA duplex PCR is a sensitive and fast tool for accurate identification of Fasciola species in areas of distributional and zonal overlap.  相似文献

Injected therapeutics, such as cells or biological molecules, may have enhanced efficiency when delivered within a scaffold carrier. Here, we describe a dual-component Dock-and-Lock (DnL) self-assembly mechanism that can be used to construct shear-thinning, self-healing, and injectable hydrogels. One component is derived from the RIIα subunit of cAMP-dependent kinase A and is engineered as a telechelic protein with end groups that dimerize (docking step). The second component is derived from the anchoring domain of A-kinase anchoring protein (AD) and is attached to multi-arm crosslinker polymers and binds to the docked proteins (locking step). When mixed, these two DnL components form robust physical hydrogels instantaneously and under physiological conditions. Mechanical properties and erosion rates of DnL gels can be tuned through the AD peptide sequence, the concentration and ratio of each component, and the number of peptides on the cross-linking polymer. DnL gels immediately self-recover after deformation, are resistant to yield at strains as high as 400%, and completely self-heal irrespective of prior mechanical disruption. Mesenchymal stem cells mixed in DnL gels and injected through a fine needle remain highly viable (>90%) during the encapsulation and delivery process, and encapsulated large molecules are released with profiles that correspond to gel erosion. Thus, we have used molecular engineering strategies to develop cytocompatible and injectable hydrogels that have the potential to support cell and drug therapies.  相似文献

Layers 5 and 6 of primate primary visual cortex (V1) harbor morphologically diverse cell groups that have corticocortical and corticosubcortical projections. Layer 6 middle temporal area (MT)-projecting neurons are particularly interesting, as they are the only deep-layer cortical neurons that provide both corticocortical feedforward inputs (to area MT) and corticosubcortical feedback projections (to superior colliculus [SC]) (Fries et al. [1985] Exp Brain Res 58:613-616). However, due to limitations in anatomical tracing techniques, little is known about the detailed morphologies of these cells. We therefore applied a genetically modified rabies virus as a retrograde tracer to fill the dendrites of projection neurons with green fluorescent protein (GFP) (Wickersham et al. [2007] Nat Methods 4:47-49). We injected virus into SC or area MT of macaque monkeys and examined labeled cells in V1. Two-thirds of labeled neurons following SC injections were found in layer 5, consisting of "tall-tufted" and "nontufted" cells; the remaining cells were layer 6 "nontufted." Area MT injections labeled neurons in layers 4B and 6, as previously described (Shipp and Zeki [1989] Eur J Neurosci 1:309-332). The layer 6 neurons projecting to MT were remarkably similar to the layer 6 SC-projecting neurons. In contrast to the dense and focused dendritic arbors of layer 5 "tall-tufted" pyramids, all "nontufted" cells had sparse, but unusually long basal dendrites. The nontufted cells closely resemble Meynert cells (le Gros Clark [1942] J Anat 76:369-376; Winfield et al. [1983] Proc R Soc Lond B Biol Sci 217:129-139), but the full in vivo reconstructions presented here show that their basal dendrites can extend much further (up to 1.2 mm) and are less asymmetric than inferred from Golgi reconstructions.  相似文献

Road traffic injuries (RTIs) are increasing in developing countries where accurate routine data are usually not available. Although a capture-recapture technique has increasingly been employed in studies of human populations to provide reliable estimates of the magnitude of problems, it has rarely been used in road traffic injury research. We applied two sample capture-recapture methods using hospital and traffic police records to estimate non-fatal road traffic injuries in Thai Nguyen City during the years 2000-2004. We generated a conservative adjusted estimate of non-fatal RTIs using data from the two sources matched by name, surname, sex of victims and at least one of the other matching variables, of age, address of victim and date of injuries. We then compared the estimated rates with those reported based on police and hospital data. The results show that during years 2000-2004, the police reported 1,373 non-fatal RTIs, while hospital records revealed 6,069 non-fatal RTIs. Most reported victims on both hospital and police reports were males (67.3 % and 74.4%, respectively). More than half the victims on both hospital and police reports were drivers (77.5% and 66.1%, respectively) or pedestrians (10.6% and 7.1%, respectively). Youth and young adults (ages 15-34) constituted the majority of the victims on the hospital and police reports (52.8% and 63.7%, respectively). The capture-recapture analysis estimated that 11,140 (95% CI: 10,626-11,654) subjects were involved in RTIs during the study period. In comparison to the estimated figure, official sources accounted for only 21.9 to 60.1% of total non-fatal RTIs. Estimated rates of non-fatal RTIs were 105.5 injuries/10,000 population per year and 393 injuries/10,000 vehicles. Given the fact that under reporting of RTIs has been a major limitation of routine official data sets in developing countries, we suggested the capture-recapture method be used as a tool to provide affordable and reliable estimates of RTIs in resource-poor countries.  相似文献
