ObjectiveThe purpose of this study was to clarify the interaction of vascular endothelial growth factors (VEGFs)-C and -D with cell surface foetal liver kinase-1 (Flk-1) and fms-like tyrosine kinase-4 (Flt-4) receptors in the induction and activity of osteoclasts in cultured human peripheral blood mononuclear cells (PBMCs).DesignPBMCs were cultured on chamber slides or on ivory discs for 2 or 3 weeks in the presence of macrophage-colony stimulating factor (M-CSF), VEGF-A, -C or -D, or placental growth factor (PlGF) with or without receptor activator of nuclear factor kappa-B ligand (RANKL). The number of osteoclasts in each group was counted and the area of ivory resorption was measured. In addition, osteoclast differentiation was further analysed under the same conditions, but with the addition of specific neutralizing antibodies against Flk-1 and Flt-4.ResultsRANKL was essential for the induction of osteoclasts in PBMCs. However, significant differences were found in the number of osteoclasts induced by VEGF-A, -C, -D or M-CSF with RANKL compared with control groups lacking or containing RANKL. Blocking of either Flk-1 or Flt-4 resulted in a reduction in the enhancement of osteoclast differentiation in PBMCs by VEGF-C or -D with RANKL. The osteoclasts induced by VEGF-A, -C, -D or M-CSF with RANKL formed significantly larger resorption lacunae than those formed by osteoclasts induced by RANKL alone.ConclusionsThis study showed that VEGF-C and -D play a role in the induction of osteoclast differentiation through both Flk-1 and Flt-4 receptors and influence the area of the ivory resorption in PBMCs.  相似文献

Objectives:We investigated the topographical representation of the masticatory muscles, primarily in the trigeminal motor nucleus (TMN) of the house musk shrew (Suncus murinus) and miniature pig, in a series of mammalian comparative anatomical studies. Additionally, correlations between motoneurons and muscles were investigated in order to examine the functionality of each muscle in the trigeminal motor system.Methods:Motoneurons were labeled using horseradish peroxidase (HRP) injection into each muscle innervated by the trigeminal nerve. In the shrew, the sizes and numbers of HRP-labeled neurons, muscle weight, and numbers of muscle fibers were measured in the innervated muscles, and correlation coefficients for the relationships among these parameters were calculated.Results:The motoneuron cluster of each muscle was arranged in the TMN in a manner similar to that observed in other previously reported animals, but the distribution of the lateral pterygoid motoneurons varied between species. In addition, considerably higher overlap was observed in each pig jaw-closer motoneuron cluster compared with other animals. The approximate innervation ratios were as follows: masseter, 337; temporal, 322; anterior digastric, 137; medial pterygoid, 110; lateral pterygoid, 79; mylohyoid, 42; tensor veli palatini, 42; transverse mandibular, 16; and tensor tympani muscles, 5.ConclusionsThe distribution pattern of the masticatory motoneurons was clearly observed in the shrew and pig TMN, and various correlations between the motoneuron and innervated muscles were determined.  相似文献

Background: Single inhaled corticosteroids and long-acting beta-agonists (ICS/LABA) are clinically effective and safe. However, if local oropharyngeal and laryngeal adverse effects (LOLAE) appear, adherence to the use of ICS is impaired. To minimize the development of adverse effects, it is essential to identify the underlying risk factors.Methods: The study included 481 asthmatic patients who were prescribed ICS/LABA for the first time in their life between January and September of 2010. Patients ranged in age from 14 to 86 years old and consisted of 281 never smokers and 200 smokers. All data were collected retrospectively by respirologists.Results: Seventy-three out of 481 patients suffered from one or more adverse effects, with 54 of these exhibiting LOLAE. Patients with LOLAE (51.4 ± 16.2 yrs) were significantly older than those without LOLAE (43.7 ± 15.9 yrs) (p = 0.0011) and were also prescribed a significantly higher dose of ICS. The pack-years of patients with LOLAE (2.1 ± 4.9) were significantly lowerthan those without LOLAE (6.0 ± 13.0) (p = 0.0087). The type of administered ICS was also significantly associated with a risk of developing LOLAE.Conclusions: Our survey indicated that a greater age, a higher dose of ICS, and the type of ICS were potential risk factors of LOLAE. The identified factors should be considered in a clinical setting in order to prevent the development of LOLAE and provide optimal treatment to patients.  相似文献

A clear understanding of the electron energy structure of TiO2(B)/anatase is needed to study the related catalytic reactions and design new composite photocatalysts. In this study, the electron energy structures of TiO2(B) and anatase were estimated by analyzing the energy-resolved distribution of electron traps measured by reversed double-beam photoacoustic spectroscopy. In the mixture of TiO2(B) and anatase, interfacial charge-transfer excitation from anatase to electron traps of TiO2(B) was suggested. By analyzing this for TiO2(B), the electron level with a relatively high density of states was found to be located ∼0.07 eV deeper than that for anatase. Furthermore, a similar electron energy structure was suggested for a composite photocatalyst having a mixed phase of TiO2(B) and anatase.
