As a consequence of the high prevalence of TorqueTeno virus (TTV) in blood donors, thalassemia patients frequently acquire various genotypes of this virus through therapeutic blood transfusions. At present, the clinical consequences of TTV infection remain indeterminate for these patients. Here, several hundred thalassemia patients were tested for the presence of TTV and its genotypes using a combination of PCR and clone-based DNA sequencing. Approximately 10% (12/118) of the patients aged 2-20 years remained negative for TTV including eight genotypes of SENV. Ferritin, aspartate-aminotransferase (AST) and alanine-aminotransferase (ALT) levels were invariably lower in TTV-negative patients (P = 0.02, <0.01, and 0.06, respectively) than in TTV-positive patients. Patients with TTV-HCV co-infection showed elevated ferritin and ALT levels compared with patients with TTV infection alone (P < 0.02 and P < 0.01). AST and ALT levels were within the normal range for all TTV-negative patients, whereas abnormal levels of AST and ALT were seen in a significant proportion of TTV-positive patients (30.7% and 33.6%, respectively) and patients with TTV-HCV co-infections (70.0% and 56.6%, respectively). Only TTV-positive patients (28.0%) and patients with TTV-HCV co-infections (36.3%) had hyper-ferritin levels (> or =3,000 ng/ml). The genotype(s) of TTV responsible for the liver dysfunction could not be determined. However, high levels of AST and ALT were found to be correlated with detection of a higher number of TTV genotypes in the patients. The data suggests that frequent and persistent TTV infection through blood transfusion is associated with hepatic dysfunction and/or damage in transfusion dependent thalassemia patients. 相似文献

IntroductionBlastocystis is a common intestinal parasite of human and animal hosts. The parasite has 17 subtypes, and among those at least nine subtypes (ST1-ST9) are found in human hosts.ObjectiveThe aim of the present study was to investigate the presence of different subtypes of Blastocystis spp. among the patients referred to Velayat hospital of Qazvin province, Iran.MethodsOverall, 864 stool samples were examined by using formalin-ethyl acetate concentration method and Trichrome staining. All specimens were cultured in clotted fetal bovine medium. Later, DNA extraction and PCR amplification of 18S ribosomal RNA gene region was conducted and phylogenetic tree constructed.ResultsThe results revealed 7.9% (68/864) of the study population were infected with Blastocystis. Intestinal symptoms were observed in 61% (36/59) of individuals positive for Blastocystis, with abdominal pain in 58% (21/36) of cases which was more frequent than other intestinal signs. No significant relationship was observed among the study variables. By molecular and phylogenetic analysis, three subtypes ST1 (45%), ST2 (30%) and ST3 (23%) of parasite were identified.ConclusionThis study showed ST1 subtype was the predominant subtype among the positive specimens, meanwhile the highest haplotype and nucleotide diversity were clarified in ST3 subtype. 相似文献

A total of 114 isolates of methicillin-resistant Staphylococcus aureus (MRSA) were collected from hospitals in Tehran, Iran. A multiplex PCR was designed to examine the presence of six different prophage classes. The results showed high diversity of bacteriophages, with four different prophage types and eight prophage patterns. An important S. aureus phage coding for several virulence factors, Φ-77-like phage, was detected in 97?% of the isolates. We found a high rate of resistance of MRSA isolates to penicillin, ciprofloxacin, tobramycin and kanamycin. This is the first study showing high prevalence and diverse bacteriophage populations in MRSA strains in Iranian hospitals. 相似文献

The purpose of this study is to investigate the applicability of the current surgical guideline stating that the main facial foramina that transmit cutaneous nerves to the face (supraorbital notch/foramen, infraorbital foramen, and mental foramen) are equidistant from the midline in European and Hispanic populations. Previous studies suggest this surgical guideline is not applicable for all ethnicities; however, to our knowledge, no data have been published regarding the accuracy of this guideline pertaining to the Hispanic population. An experimental study was performed on 67 cadavers donated to the Human Anatomy Program at UT Health San Antonio. The supraorbital, infraorbital, and mental foramina were dissected and midline structures including the crista galli, internasal suture, anterior nasal spine, and mandibular symphysis were identified. The distance from each foramen to midline was recorded using a digital caliper. For all cadavers/ethnicities studied, the supraorbital, infraorbital, and mental foramina were 25.32 mm, 29.57 mm, and 25.55 mm to the midline, respectively. Thus, the infraorbital foramen is located significantly more lateral compared to the supraorbital (p < 0.0001) and mental foramina (p < 0.0001). After dividing the sample based on ethnicity, this relationship was also true for the European sample and tended to be true for the Hispanic sample. Significant anatomical variations exist in the current surgical guideline stating that the supraorbital foramen, infraorbital foramen, and mental foramen are equidistant from the midline. Clinicians may need to adjust their methodology during surgical procedures of the face in order to optimize patient care. 相似文献

Cytokines are mediators for polarization of immune response in vaccines. Studies show that co‐immunization of DNA vaccines with granulocyte‐macrophage colony‐stimulating factor (GM‐CSF) can increase immune responses. Here, experimental mice were immunized with HIV‐1tat/pol/gag/env DNA vaccine with GM‐CSF and boosted with recombinant vaccine. Lymphocyte proliferation with Brdu and CTL activity, IL‐4, IFN‐γ, IL‐17 cytokines, total antibody, and IgG1 and IgG2a isotypes were assessed with ELISA. Results show that GM‐CSF as adjuvant in DNA immunization significantly increased lymphocyte proliferation and IFN‐γ cytokines, but CTL response was tiny increased. Also GM‐CSF as adjuvant decreased IL‐4 cytokine vs mere vaccine group. IL‐17 in the group that immunized with mixture of DNA vaccine/GM‐CSF was significantly increased vs DNA vaccine group. Result of total antibody shows that GM‐CSF increased antibody response in which both IgG1 and IgG2a increased. Overall, results confirmed the beneficial effect of GM‐CSF as adjuvant to increase vaccine immunogenicity. The hallmark result of this study was to increase IL‐17 cytokine with DNA vaccine/GM‐CSF immunized group. This study for the first time provides the evidence of the potency of GM‐CSF in the induction of IL‐17 in response to a vaccine, which is important for control of infection such as HIV‐1. 相似文献

Milk is considered a nutritious food because it contains several important nutrients including proteins and vitamins. Conversely, it can be a vehicle for several pathogenic bacteria such as Staphylococcus aureus. The aim of the present study was to investigate the presence of classical enterotoxins of S. aureus in raw milk in Iran. For this purpose, from January 2011 to October 2011, a total of 88 raw milk samples from sheep (n?=?28), goat (n?=?27), camel (n?=?14), and water buffalo (n?=?19) were collected from 64 randomly selected herds in Isfahan, Chaharmahal va Bakhtiari, and Khuzestan provinces, Iran, and tested for staphylococcal enterotoxins (SEs) presence by enzyme-linked immunosorbent assay method. Out of 88 samples studied, 11 samples (12.5%) were positive for at least one SE. The prevalence rate of SEs contamination in raw sheep, goat, and water buffalo milk samples were 10.7%, 18.5%, and 15.8%. No classical SEs was founded from raw camel milk samples. Of these, four were positive for staphylococcal enterotoxin A (SEA), four for SEC, two SEA and SEC, and one for SEA and SED. None of the samples was positive for SEB and SEE. Statistical evaluation showed that there were no significant differences (P?>?0.05) between the presence of SEs raw sheep, goat, and water buffalo milk samples tested. The quality of Iranian milk is still low, and efforts from the government and the entire productive chain are required to attain consumer safety. 相似文献

The objective of this study was to determine the Salmonella prevalence, the serotypes involved, and antimicrobial susceptibility patterns of Salmonella isolates recovered from fish, shrimp, lobster, and crab in Iran. A total of 384 samples of fish, shrimp, lobster, and crab were collected in three provinces along the Persian Gulf in the south coast of Iran. Samples were collected at the end of each month from September 2009 to May 2011. All samples were evaluated for the presence of Salmonella, stereotyped and tested for antimicrobial susceptibility. There was an overall Salmonella prevalence of 5%. Salmonella was isolated from a significantly larger number of fish (10.4%) than shrimp (1.8%; P?≤?0.05). No Salmonella was isolated from lobster and crab samples. Salmonella isolates recovered from fish and shrimp samples were of five different serotypes including Salmonella typhimurium, Salmonella enteritidis, Salmonel la typhi, Salmonella paratyphi B, and Salmonella newport. Susceptibilities of Salmonella isolates were determined for 12 antimicrobial drugs using the disk-diffusion method. Resistance to nalidixic acid was the most common finding (47.4%), followed by resistance to tetracycline (36.8%), streptomycin (15.8%), trimethoprim (15.8%), and ciprofloxacin (5.3%). To our best knowledge, this is the first study on prevalence of Salmonella in fish, shrimp, lobster, and crab and the first report on the isolation of Salmonella spp. from retail fish and shrimp in Iran. 相似文献

Human anxiety is frequently accompanied by depression, and when they co-occur both conditions exhibit greater severity and resistance to treatment. Little is known, however, about the molecular processes linking these emotional and mood disorders. Based on previously reported phosphorylation patterns of extracellular signal-regulated kinase (ERK) in the brain, we hypothesized that ERK's upstream activators intertwine fear and mood regulation through their hippocampal actions. We tested this hypothesis by studying the upstream regulation of ERK signaling in behavioral models of fear and depression. Wild-type and ERK1-deficient mice were used to study the dorsohippocampal actions of the putative ERK activators: mitogen-activated and extracellular signal-regulated kinase (MEK), protein kinase C (PKC), and cAMP-dependent protein kinase (PKA). Mice lacking ERK1 exhibited enhanced fear extinction and reduced depression caused by overactivation of ERK2. Both behaviors were reversed by inhibition of MEK, however the extinction phenotype depended on hippocampal, whereas the depression phenotype predominantly involved extrahippocampal MEK. Unexpectedly, inhibition of PKC accelerated extinction and decreased depression by ERK-independent mechanisms, whereas inhibition of PKA did not produce detectable molecular or behavioral effects in the employed paradigm. These results indicate that, contrary to fear conditioning but similar to mood stabilization, extinction of fear required upregulation of MEK/ERK and downregulation of ERK-independent PKC signaling. The dissociation of these pathways may thus represent a common mechanism for fear and mood regulation, and a potential therapeutic option for comorbid anxiety and depression. 相似文献

我们报道成功治疗波氏假阿利什菌引发的真菌性角膜炎1例,患者只留有很小的角膜瘢痕。一位71岁女性患者,有疼痛、红肿和异物感病史,来到我们第三眼科中心就诊。最初角膜刮片显示有真菌成分,2d后真菌培养为阳性,5d后显示生长物为波氏假阿利什菌。患者仅用药物疗法取得临床治愈,留下微小的角膜瘢痕,最终视力为3/10。 相似文献

The antimicrobial activity of the methanol extracts of the dried flowering aerial parts of Stachys byzantina, S. inflata, S. lavandulifolia and S. laxa (Labiatae) were studied using the disc diffusion method and determination of minimum inhibitory concentration (MIC) values against Staphylococcus aureus, Streptococcus sanguis, Escherichia coli, Pseudomonas aeroginosa, Klebsiella pneumoniae, Aspergilus niger and Candida albicans. The extracts of plants exhibited concentration-dependent antibacterial activity against the bacteria tested. The extracts were more active against Gram-positive microorganisms.
