PurposeRetinal microglia promote angiogenesis and vasculopathy in oxygen-induced retinopathy (OIR); however, its specific molecular mechanism in the formation of retinal angiogenesis remains unclear. The lectin galactoside-binding soluble 3 binding protein (LGALS3BP), a member of the scavenger receptor cysteine-rich (SRCR) domain protein family, is involved in tumor neovascularization, and we therefore hypothesized that LGALS3BP plays an active role in microglia-induced angiogenesis.MethodsThe expression of LGALS3BP in microglia was detected by immunofluorescence, RT-qPCR, and western blotting. Functional assays of human umbilical vein endothelial cells (HUVECs) such as migration, proliferation, and tube formation were measured by Transwell, EdU, and Matrigel assays. Angiogenesis-related factors and PI3K/AKT levels were detected by western blotting. The relationship between LGALS3BP and PI3K or HIF-1α was investigated by immunoprecipitation.ResultsOur results showed that the expression of LGALS3BP was significantly increased in microglia surrounding neovascularization of the OIR mice and was also upregulated in human microglial clone 3 (HMC3) cells after hypoxia. Moreover, HUVECs co-cultured with hypoxic HMC3 cells showed increased migration, proliferation, and tube formation, as well as levels of angiogenesis-related factor. However, the proangiogenic ability and angiogenesis-related factor expression of HMC3 cells was suppressed after silencing LGALS3BP. LGALS3BP induces the upregulation of angiogenesis-related factors through the PI3K/AKT pathway and then promotes angiogenesis in microglia.ConclusionsCollectively, our findings suggest that LGALS3BP in microglia plays an important role in angiogenesis, suggesting a potential therapeutic target of LGALS3BP for angiogenesis. 相似文献

目的 了解实习护生工作场所欺凌现状及其影响因素,为针对性干预工作场所欺凌行为提供参考。 方法 以便利抽样方法抽取实习护生315名,采用中文版负性行为问卷进行调查。 结果 实习护生工作场所欺凌得分为31.43±9.92,在过去6个月中遭受工作场所欺凌的实习护生107人(33.97%)。不同学历层次、生源地、学习成绩、就读护理专业原因的实习护生感受的工作场所欺凌得分比较,差异有统计学意义(均P<0.01)。多元逐步回归分析显示,就读护理专业原因、生源地、学历层次是实习护生感受工作场所欺凌的主要影响因素(P<0.05,P<0.01)。 结论 实习护生感知工作场所欺凌的比例偏高,护理管理者和教育者应当予以重视,采取有效防范措施和针对性教育,减少工作场所欺凌的发生。 相似文献

Objective: Recent studies have shown that the local expression of soluble interleukin (IL) -1 receptor type Ⅱ (slL-1 R Ⅱ ) in endometrial tissue of women with endometriosis is decreased, and the depression of IL-1 R Ⅱ was more significant in infertile women than that in fertile women with endometriosis. In this research, we investigated the remedial effect of slL-1-R Ⅱ administration on endometriosis in the nude mouse model. Methods: Nineteen nude model mice with endometriosis were randomly divided into three groups: group A was treated by intraperitoneal administration with only slL-1 R Ⅱ for two weeks, group B was similarly treated with only IL- 1, and group C (control) was administered saline. After 2 weeks, the size of the ectopic endometrial lesions was calculated, and the expression of vascular endothelial growth factor (VEGF) and B-cell lymphoma leukemia-2 (Bcl- 2) were detected by immunohistochemistry. The IL-8 and VEGF levels in the peritoneal fluid (PF) and serum were also measured by enzyme-linked immunosorbent assay (ELISA). Results: The mean size of ectopic endometrial lesion did not differ between the three groups (P 〉 0.05). Compared with the control, the expression of VEGF and Bcl-2 was significantly lower in group A, and higher in group B. In the three groups, the levels of IL-8 in the PF and serum were highest in group A, and lowest in group B. Conclusion: slL-1 R Ⅱ may suppresse hyperplasia of ectopic endometriosis, perhaps by reducing the expression of certain cytokines, such as VEGF, IL-8, and Bcl-2, which could provide a new clinical strategy for the treatment of endometriosis. 相似文献

目的:观察吉奥在老年进展期胃癌患者中的应用及对AEG-1/MT qDH、NF-κB、MMP-9水平的影响.方法:取胃癌患者100例,随机分为对照组(n=45)和观察组(n=45).对照组采用胃切除术治疗,观察组联合替吉奥化疗治疗,采用免疫组织化学法检测2组患者AEG-1/MT qDH、NF-κB、MMP-9水平,比较两组临床疗效及对AEG-1/MT qDH、NF-κB、MMP-9水平的影响.结果:观察组治疗后1年生存率、3年生存率及5年生存率,高于对照组;观察组治疗后AEG-1/MT qDH、NF-κB、MMP-9水平,显著低于对照组;观察组治疗后并发症发生率为22.00%,与对照组的18.00%相比差异无统计学意义.结论:胃癌患者在胃切除治疗基础上联合替吉奥治疗效果理想,能降低AEG-1/MT qDH、NF-κB、MMP-9水平,值得推广应用. 相似文献

为评价检测血清前清蛋白(PA)代替血清总胆汁酸(TBA)作为肝功能检测的可行性,对156份标本(43例正常人及59例肝炎、37例肝硬化、17例肝癌患者)血清进行PA与TBA的检测比较,结果显示两种方法的相符率为77.56%,而且两种方法的敏感性、特异性、准确率均无显著差异,但综合评价检测PA的可用度明显优于检测TBA(P<0.05),而且检测方法简便、价格便宜、试剂来源方便且不需复杂的设备。 相似文献

目的:研究TRAP基因在人肿瘤组织中的表达及其与人端粒酶逆转录酶(hTERT)的作用相关性,探讨其在人肿瘤发生中的作用。方法:通过原核表达TRAP蛋白免疫新西兰白兔,制备特异性多克隆抗体;免疫组化检测TRAP及hTERT在人肿瘤组织中的表达;通过构建TRAP真核表达载体、基因转染,观察TRAP对细胞hTERT转录、端粒酶活性和癌细胞生长及成瘤性的影响。结果:通过大肠杆菌表达的TRAP蛋白免疫、制备多克隆抗体,经Westem blot鉴定与TRAP具有特异结合性;免疫组化显示247例恶性肿瘤中有213例TRAP阳性,表达率为86.2%,而在所有癌旁组织中TRAP为阴性。另外,交界性及癌前病变与良性病变组织中TRAP表达差异也有显著性:进一步检测卵巢生发上皮肿瘤、乳腺癌、肝癌及中枢神经系统肿瘤组织hTERT表达,显示hTERT与TRAP表达的结果相一致。TRAP正义转染使hTERT及端粒酶活性升高,细胞增殖加快;而反义转染时,两者均下降,细胞增殖受抑。裸鼠成瘤性实验表明:正义TRAP使癌细胞成瘤性增强,而反义则抑制移植瘤的生长。结论:TRAP表达存在于人癌组织及部分癌前病变,与癌细胞恶性表型相关,并与hTERT表达一致。TRAP基因参与对端粒酶的调节作用,可能与人肿瘤发生有关。 相似文献

Photothermal therapy (PTT) has become one of the most promising therapies in cancer treatment as its noninvasiveness, high selectivity, and favorable compliance in clinic. However, tumor thermotolerance and distal metastasis reduce its efficacy, becoming the bottleneck of applying PTT in clinic. In this study, a chidamide-loaded magnetic polypyrrole nanocomposite (CMPP) has been fabricated as a visualized cancer photothermal agent (PTA) to counter tumor thermotolerance and metastasis. The efficacy of CMPP was characterized by in vitro and in vivo assays. As a result, this kind of magnetic polypyrrole nanocomposites were black spherical nanoparticles, possessing a favorable photothermal effect and the suitable particle size of 176.97 ± 1.45 nm with a chidamide loading rate of 12.92 ± 0.45%. Besides, comparing with PTT, CMPP exhibited significantly higher cytotoxicity and cellular apoptosis rate in two tumor cell lines (B16-F10 and HepG2). In vivo study, the mice showed obvious near-infrared (NIR) and magnetic resonance imaging (MRI) dual-modal imaging at tumor sites and sentinel lymph nodes (SLNs); on the other hand, magnetic targeting guided CMPP achieved a cure level on melanoma-bearing mice through preventing metastasis and thermotolerance. Overall, with high loading efficiency of chidamide and strong magnetic targeting to tumor sites and SLNs, CMPP could significantly raise efficiency of PTT by targeting tumor thermotolerance and metastasis, and this strategy may be exploited therapeutically to upgrade PTT with MPP as one of appropriate carriers for histone deacetylase inhibitors (HDACis). 相似文献
