Water frogs, Pelophylax perezi, that are introduced in the Azores, were screened for parasites using PCR primers known to amplify Apicomplexa parasites, and using nematode-specific primers. With the former, three different organisms were detected: Hepatozoon, a trichodinid protozoan ciliate and a possible Stramenopile. Using the latter set of primers, a single unknown spirurid nematode was also detected. Phylogenetic analyses indicate that Hepatozoon detected within amphibian hosts appear to form a clade, although relationships of these parasites do not match the vertebrate intermediate host phylogeny. Regarding the possible Stramenopile, it is unclear whether this organism was actually present on the amphibian or in the water on the surface of the tissue sample. Our findings highlight that many different organisms can be detected with these primers and that they can be used to screen introduced host populations to detect parasites that have been brought with them.  相似文献

BackgroundOsteoarthritis (OA) is a degenerative disease that induces peri-articular tissue degradation. OA induces an imbalance between synthesis and degradation of the extracellular matrix components in favor of catabolic events, promoting pathological remodeling and involving degradative enzymes, such as matrix metalloproteinases (MMPs).ObjectiveThis study aimed to investigate the effects of 8-weeks resistance training (RT) on MMP-2 activity in the quadriceps tendon and patellar tendon in an OA model.MethodsTwenty-four Wistar rats were randomly divided into six groups: Control, Exercise, Sham, Sham with Exercise, OA, and OA with Exercise (OAE). The OA model was performed by anterior cruciate ligament transection surgery on the left knee. The 8-week RT consisted of climbing a 1.1-m vertical ladder three times per week with progressive weights secured to the animals’ tails. MMP-2 activity was analyzed by zymography.ResultsThe OAE group displayed lower pro, intermediate, and active MMP-2 activity in the quadriceps tendon compared with the OA group (p < 0.05). For the patellar tendon, there was no significant difference between the OAE group compared with the other groups (p > 0.05) for pro, intermediate, and active MMP-2 activity. Moreover, MMP-2 activity differed between tissues, the OA and OAE groups presented lower pro, intermediate, and active MMP-2 activity in the quadriceps tendon compared to the patellar tendon.ConclusionRT induced down-regulated MMP-2 activity in the quadriceps tendon. RT is a potential therapeutic approach to minimize the deleterious effects of extracellular matrix degeneration.  相似文献

ObjectiveTo compare the performance, reliability, and validity of functional tests between women with and without patellofemoral pain.MethodsTwenty women with a diagnosis of patellofemoral pain between 18 and 40 years of age and 20 age-matched pain-free controls participated in the study. All participants performed a set of five function tests: sitting-rising test, sit-to-stand in 30 seconds, stair-climb test, stair descent test, and six-minute step test. To investigate reliability, participants were assessed on two different days, seven days apart, by two independent investigators blinded to the results of the other investigator. Validity was evaluated through associations with the results on the Anterior Knee Pain Scale.ResultsPerformance in the tests was worse in women with patellofemoral pain than in the control group for the sit-to-stand in 30 seconds (mean difference [MD] 3.4reps; 95%CI: 0.4, 6.4), stair-climb test (MD: 0.36 s; 95%CI: 0.1, 0.63), and six-minute step test (MD: 45reps; 95%CI: 20, 70). No differences were observed for the sitting-rising and stair descent tests. All tests in both groups showed moderate to excellent intra- and inter-rater reliability (intraclass correlation coefficients: 0.61 to 0.91 and 0.72 to 0.96, respectively). Finally, only the results on the sit-to-stand in 30 seconds test correlated with the Anterior Knee Pain Scale (r = 0.44, p = 0.047) in the patellofemoral pain group.ConclusionWomen with patellofemoral pain present lower performance on some functional tests. Functional tests are reliable in patients with patellofemoral pain, although they are not associated with the results on the Anterior Knee Pain Scale self-questionnaire.  相似文献

PURPOSE

We aimed to evaluate the added value of diffusion-weighted imaging (DWI) to standard magnetic resonance imaging (MRI) for detecting post-treatment cervical cancer recurrence. The detection accuracy of T2-weighted (T2W) images was compared with that of T2W MRI combined with either dynamic contrast-enhanced (DCE) MRI or DWI.

METHODS

Thirty-eight women with clinically suspected uterine cervical cancer recurrence more than six months after treatment completion were examined with 1.5 Tesla MRI including T2W, DCE, and DWI sequences. Disease was confirmed histologically and correlated with MRI findings. The diagnostic performance of T2W imaging and its combination with either DCE or DWI were analyzed. Sensitivity, positive predictive value, and accuracy were calculated.

RESULTS

Thirty-six women had histologically proven recurrence. The accuracy for recurrence detection was 80% with T2W/DCE MRI and 92.1% with T2W/DWI. The addition of DCE sequences did not significantly improve the diagnostic ability of T2W imaging, and this sequence combination misclassified two patients as falsely positive and seven as falsely negative. The T2W/DWI combination revealed a positive predictive value of 100% and only three false negatives.

CONCLUSION

The addition of DWI to T2W sequences considerably improved the diagnostic ability of MRI. Our results support the inclusion of DWI in the initial MRI protocol for the detection of cervical cancer recurrence, leaving DCE sequences as an option for uncertain cases.Cervical cancer is the fourth most frequent cancer in women worldwide (1). Early stage disease is treated with surgery or chemoradiotherapy and has a good prognosis. However, around 30% of all patients treated for cervical carcinoma develop progressive or recurrent tumors (2).Recurrent cervical cancer is defined as local tumor regrowth or the development of distant organ/lymph node metastases at least six months after regression of the initial lesion. Approximately two-thirds of recurrences appear within the first two years following initial treatment, with 90% recurring by five years post-treatment (3). Risk factors for recurrence include histopathologic features, depth of tumor invasion, and nodal status (4).Pelvic recurrence can be located centrally (cervix, uterus, vagina, parametria, ovaries, bladder, or rectum) or in the pelvic sidewalls. Extrapelvic recurrence most commonly involves the para-aortic lymph nodes, lungs, liver, or bone (46).Treatment of recurrent cancer depends on the primary treatment approach, location, and extension. Patients with locally recurrent disease can be offered salvage treatments with curative potential (chemoradiotherapy, if not given previously, or pelvic exenteration in patients who already received chemoradiotherapy). Distant metastases, however, are nearly always incurable (3).In patients who successfully completed primary treatment, surveillance has been advocated to detect the residual or recurrent disease at curable stages (7). The use of imaging studies such as magnetic resonance imaging (MRI) is indicated on the basis of clinical suspicion (8).T2-weighted (T2W) imaging is the reference sequence for cervical cancer staging (9). Recurrent tumors are known to show high signal intensity on T2W MRI, contrasting with the low signal intensity of the cervical stroma. However, some benign conditions such as necrosis, inflammation, and edema may also increase signal intensity on T2W images, representing a potential challenge to the radiologist, particularly after radiotherapy (1013).Moreover, post-treatment changes can result in areas of fibrosis that are also difficult to differentiate from recurrence (14). MRI has proven to be superior to computed tomography (CT) in distinguishing fibrosis and scarring from active disease, but imaging findings are sometimes indeterminate, complicating the evaluation of recurrent disease (3).In recent years, the functional MRI techniques such as dynamic multiphase contrast-enhanced (DCE) MRI and diffusion-weighted imaging (DWI) have emerged as fundamental tools in female pelvic imaging evaluation (15). Although DCE was shown to be more accurate than T2W alone for tumor recurrence identification, the use of both sequences is recommended (10).Recently, DWI has been added to pelvic MRI protocols to increase diagnostic accuracy in tumor staging. This technique is a functional tool that relies on tissue water displacement to create a contrasted image. For correct evaluation and avoidance of pitfalls, the generated images must be interpreted alongside anatomical sequences. The apparent diffusion coefficient (ADC) map is also needed to reduce image misinterpretation, for example due to the T2 shine-through effect (15). In highly cellular tissues, water movement is restricted and such lesions appear bright at high b-values (1000 s/mm2) and have low ADC value, appearing dark gray on ADC maps in contrast to areas of freely moving water such as urine in the bladder (14). Some recent studies have suggested that DWI and ADC maps can be potentially useful in oncologic follow-up (14, 16).The purpose of this study was to compare the accuracy of T2W/DWI with that of conventional anatomical sequences alone and T2W/DCE imaging sequences in the evaluation of recurrent disease in patients treated for uterine cervical carcinoma.  相似文献

Male‐to‐female transsexual persons use oestrogens + antiandrogens to adapt their physical bodies to the female sex. Doses are usually somewhat higher than those used by hypogonadal women receiving oestrogen replacement. Particularly in cases of self‐adminstration of cross‐sex hormones, doses may be very high. Oestrogens are powerful stimulators of synthesis and release of prolactin and serum prolactin levels are usually somewhat increased following oestrogen treatment. Prolactinomas have been reported in male‐to‐female transsexual persons, both after use of high and conventional doses of oestrogens but remain rare events. We report two new cases of prolactinomas in male‐to‐female transsexual persons, one in a 41‐year‐old subject who had used nonsupervised high‐dose oestrogen treatment since the age of 23 years and another one in a 42 year old who had initiated oestrogen treatment at the age of 17 years. Their serum prolactin levels were strongly increased, and the diagnosis of a pituitary tumour was confirmed by imaging techniques. Both cases responded well to treatment with cabergoline treatment whereupon serum prolactin normalised. Our two cases are added to the three cases of prolactinomas in the literature in persons who had used supraphysiological doses of oestrogens.  相似文献
