反义胰岛素样生长因子—1mRNA治疗大鼠脑胶质瘤模型的动态MR研究

目的：动态观察大鼠Ｃ６胶质瘤模型的自然生长过程及反义ＩＧＦ－１ｍＲＮＡ对胶质瘤的治疗效果和治疗机制。方法：将３２只Ｗｉｓｔａｒ大鼠分成４组,每组８只,（１）对照组（Ｃ组）于右尾状核接种Ｃ６鼠胶质瘤细胞。（２）治疗组（Ｔ组）：于右尾状核接种Ｃ６细胞后的第７、１０天在接种点注射脂质体包裹的质粒Ｐ－ａｎｔｉ－ＩＧＦ－１,对胶质瘤进行治疗于右尾状核接种经反义ＩＧＦ－１ｍＲＮＡ转染的Ｃ６胶质瘤细胞。（４）Ｋ     

复方云芝胶囊对丝裂霉素C诱发突变及老年自发突变的抑制作用

采用体外培养人淋巴细胞微核检测法,探讨复方云芝胶囊抗突变作用。结果表明,复方云芝胶囊（１０ｍｇ／Ｌ～５０ｍｇ／Ｌ）可显著抑制丝裂霉素Ｃ（ＭＭＣ）诱发的微核形成,并可拮抗老年人自发微核,增加细胞增殖活性。     

用LC／MS／MS和RP—HPLC法对泻心汤煎煮产生的沉淀物的研究

目的：用ＬＣ／ＭＳ／ＭＳ和ＲＰ－ＨＰＬＣ法研究沔心汤煎煮产生的沉淀物。方法：利用ＬＣ－ＭＳ－ＭＳ法对淀心肠沉淀物进行了初步研究,又利用ＲＰ－ＨＰＬＣ法,测定了淀心汤沉淀物中小檗碱在小鼠体内的吸收。结果：发现其中含有小檗碱、巴马丁碱、黄连碱、药根碱,但在给小鼠灌服淀心汤沉淀物后２４ｈ内的几个时间段中,均未能从血浆中测到小檗碱。结论：淀心汤沉淀物进入体内后小檗碱并未进入血液,推测如果淀心汤沉淀物具有恬     

Dielectric Analysis of Phosphorylcholine Head Group Mobility in Egg Lecithin Liposomes

Purpose. A knowledge of the interfacial properties of lecithin underpins our understanding of many of the physicochemical characteristics of drug delivery systems such as liposomes and lecithin stabilized microemulsions. In order to further this understanding, a high frequency dielectric study of the interfacial properties of egg lecithin liposomes was undertaken. Methods. The effect of temperature, lecithin concentration and probe sonication on the interfacial dielectric properties of liposomal suspensions was investigated by high frequency dielectric relaxation spectroscopy between 0.2–6 GHz. Results. The frequency dependent permittivity of each suspension exhibited a dielectric dispersion centred around 100 MHz, corresponding to the relaxation of zwitterionic head groups. The activation energy for head group reorientation was estimated as H = 6.3 kJ mol^–1. There was an increase in extent of inter-head group interactions on increasing the liposome volume fraction, whereas the effect of probe sonication showed that: (i) head groups in both the outer and inner lamellae contribute to the dielectric response; (ii) the head groups may be less restricted in liposomes of high surface curvature with few lamellae; (iii) the high frequency permittivity of the suspension increased on sonication, as a result of a reduction in the amount of (depolarized) interlamellar water following a reduction in the number of lamellae per liposome. Conclusions. Dielectric analysis of the zwitterionic head groups of lecithin therefore provides a means for investigating the surface of lecithin liposomes, and may be used to investigate the effect of drugs and other solutes on membranes.     

Anti-tumor ribonuclease, combined with or conjugated to monoclonal antibody MRK16, overcomes multidrug resistance to vincristine in vitro and in vivo

Onconase, a ribonuclease isolated from Rana pipiens oocytes and early embryos, is a member of the RNase A superfamily. Onconase has anti-neoplastic properties both in vitro and in vivo, and is undergoing clinical evaluation. In the present study, Onconase was combined with or conjugated to MRK16, an anti-P-glycoprotein (Pgp) monoclonal antibody. The interaction of these combinations with vincristine (VCR) against parental and multidrug resistant (MDR), Pgp expressing, human colon carcinoma cells caused increased VCR cytotoxicity in vitro and enhanced survival of athymic nude mice given transplants of drug resistant HT-29(mdr1) cells in vivo. The results suggest that combination treatment with Onconase and other agents that modulate the chemosensitivity of Pgp-expressing human tumor cells has the potential to overcome MDR.     

槐角的避孕成分研究(Ⅱ)——化合物Ⅴ～Ⅷ的分离和鉴定

本文报道从槐角中分离到的化合物Ⅴ～Ⅷ的鉴定。经紫外、核磁共振光谱分析及物理化学常数的测定,证明化合物Ⅴ～Ⅷ分别为山萘酚-3-槐糖甙、染料木素-7-双葡萄糖甙、山萘酚-3-鼠李糖-双葡萄糖甙和染料木素-7-双葡萄糖-鼠李糖甙。这些甙水解后的甙元山萘酚和染料木素对小鼠具有抗生育活性。     

Shortlasting, Unilateral, Neuralgiform Headache Attacks With Conjunctival Injection, Tearing, and Subclinical Forehead Sweating ("Sunct" Syndrome): II. Changes in Heart Rate and Arterial Blood Pressure During Pain Paroxysms.

The recently described "Sunct" syndrome is a rare picture of unilateral, shortlasting headache attacks accompanied by autonomic phenomena (conjunctival injection, tearing, etc.) on the symptomatic side. Heart rate and blood pressure were monitored in two elderly "Sunct" patients during and outside headache attacks. An ultrasound Doppler servo method was used for the non-invasive, continuous, beat-to-beat determination of instantaneous arterial blood pressure. In a third patient, systolic and diastolic blood pressure, both outside and during pain paroxysms, were assessed using the standard Korotkoff method. Heart rate was found to be significantly decreased during pain paroxysms. Systolic blood pressure was observed to be significantly increased during attacks, when compared with the inter-attack period, while a less consistent pattern was observed for diastolic blood pressure. Some of the changes in the cardiovascular system seemed to start prior to pain onset. Therefore, it seems unlikely that these changes were caused by pain activation of the sympathetic nervous system or the oculocardiac reflex.     

Properties of hemoglobin and dextran-hemoglobin rightshifted by oxidized inositol tetrakisphosphate

Phytate was digested by wheat bran phytase to yield inositol tetrakisphosphate. Periodate-oxidized inositol tetrakisphosphate (oxyIP4) was coupled by means of reductive alkylation to hemoglobin and the covalent dextran-hemoglobin conjugate to yield the rightshifted (rs) compounds rsHb and rsDxHb, respectively. The variations of the oxygen dissociation curves of these molecules with pH and temperature were compared to those of hemoglobin. The variations with pH were found to be less pronounced for these rightshifted forms. An extensive decrease in the half-saturation oxygen tension was observed, however, with both rsHb and rsDxHb, as in the case of unmodified Hb. Modification of hemoglobin by oxyIP4 at the polyphosphate site was suggested by the lack of a further rightshifting effect of phytate on rsHb, and by the similarity between the difference spectrum of rs-methemoglobin and the difference spectrum induced by the addition of phytate.     

高效液相色谱法测定复方苯甲酸乳膏中苯甲酸和水杨酸的含量

目的:建立高效液相色谱法(HPLC)测定复方苯甲酸乳膏中苯甲酸和水杨酸的含量。方法:色谱柱为Hypersil C_(18)柱(4.6nm×250mm,5μm),流动相为甲醇-水(44:56),流速0.8mL·min~(-1),检测波长为226nm。结果:苯甲酸和水杨酸分别在15.20～76.00mg·L~(-1)和7.64～38.20mg·L~(-1)范围内线性良好(r=0.9999);日内、日间RSD在0.030～4.79％,高、中、低浓度回收率在98.54％～101.20％。分析了3批样品,结果满意。结论:本方法简便,快速,准确,可作为复方苯甲酸乳膏的质量控制方法。
