本研究探讨五聚体及胞内因子染色在定性定量检测抗原特异性T细胞中的应用价值。用RT—PCR方法检测受试者WT1表达水平;用HLA—A*0201/wTl五聚体及胞内IFNγ染色检测14例表达HLA—A*020l的白血病患者全相合移植前后及其供者外周血中的WT1特异性T细胞。结果表明:14例供者中2例有低水平WT1表达,白血病患者均有不同水平的WT1表达。14例供者均未检测到WT1+CD8^+CTL和WT1^+IFNγ^+细胞;移植前14例患者中2例可检出WT1^+CD8^+CTL,3例检出WTl^+IFNγ^+细胞,二者间无明显差异(P=0.357),而移植后均可检出WT1^+CD8^+CTL和WT1^+IFNγ^+细胞,明显高于移植前(P值均〈0.01)。移植后大部分患者均在30天内检出WT1^+CD8^+CTL和WT1^+IFNγ+细胞,但后者检出率高于前者(P=0.014)。14例患者中WT1^+CD8^+CTL峰值中位数为0.18%,而WT1^+IFNγ^+细胞峰值中位数为0.83%,明显高于前者(P=0.005);WT1^+CD8^+CTL峰值中位时间为移植后75天,WT1^+IFNγ^+细胞峰值中位时间为移植后105天,但二者间无明显差异(P=0.455)。结论:五聚体及胞内IFNγ染色能有效检测白血病患者外周血中白血病抗原特异性T细胞;两种方法联合检测有助于抗原特异性T细胞的准确定性定量检测。  相似文献

糖尿病视网膜病变(diabetic retinopathy,DR)是糖尿病导致的严重微血管并发症之一。目前,对于DR确切的发病机制仍无定论。近期研究表明,炎症与自身免疫反应在DR的发生发展中起着重要作用。核苷酸结合寡聚化结构域样受体蛋白3(nucleotide-binding domain and leucine-rich repeat receptor containing a pyrin domain 3,NLRP3)炎症小体能触发各种炎症反应,并在多种慢性炎症性疾病中发挥重要作用。现已有不少研究证实NLRP3炎症小体通过参与多条通路在DR中发挥促炎作用,提示通过抑制NLPR3炎症小体或者其参与的通路或许可成为DR治疗新靶点。本文就NLRP3炎症小体在DR发病中的作用作一综述。  相似文献

目的鉴定分离自江苏省先天性畸胎的弓形虫虫株(KS株)和HIV-弓形虫共感染患者刚地弓形虫的基因型。方法抽提江苏省先天性畸胎弓形虫分离株(KS株)速殖子和1例HIV-弓形虫共感染患者的全血DNA,选择11个基因位点,采用聚合酶链式反应-限制性片段长度多态性分析(PCR-RFLP)进行基因型鉴定。结果自江苏省先天性畸胎分离的弓形虫虫株和1例HIV-弓形虫共感染患者的全血DNA样本中均获得完整分型条带,经鉴定均为弓形虫基因型Ⅰ型。结论Ⅰ型刚地弓形虫可能是江苏地区引起妊娠异常结局妇女和HIV-弓形虫共感染患者的优势基因型虫株。  相似文献

In 2003, after three pilot projects successfully implemented WHO's Health-Promoting Schools (HPS) concept, officials in Zhejiang Province, China, expanded to additional 51 schools (93,000 students and their families and 6800 school personnel). Each school identified a health issue to begin HPS development, followed by conceptual orientation, resource mobilization, teacher training, surveys, interventions, outreach and evaluation. This study focused on the extent to which participating schools implemented the HPS concept and improved their psycho-social environments (PSEs). Forty-nine of the 51 schools met China's HPS criteria. Schools with fewer resources and with substantial resources, i.e. schools in both rural and urban areas, met the criteria. Schools' PSEs, as measured by the PSE Profile, improved as they became HPS. Findings from interviews and observations identified strong encouragement and support from officials, school personnel, students, parents and community leaders, and consistency of HPS with the national policy on quality education, as success factors.  相似文献

Adult-onset Still's disease (AOSD) is a rare, systemic inflammatory disorder, characterized by spiking high fever, fever-associated evanescent rash, arthritis, myalgia, serositis and hepatosplenomegaly. White blood cell count, neutrophilic cell count, and serum ferritin level are markedly elevated in the active stage of the disease. Neurological complications of AOSD commonly were cranial nerve palsies, seizures, aseptic meningoencephalitis, peripheral neuropathy and Miller-Fisher syndrome. We report a previously healthy 60-year-old Chinese man who fulfilled the criteria for AOSD and had a combination of focal and peripheral neurological symptoms. Magnetic resonance angiography (MRA) and transcranial Doppler ultrasonography (TCD) showed narrowness of cerebral blood vessel. Peripheral neuropathy was confirmed by electromyography and sural nerve biopsy. His generalized neuropathy and other symptoms were rapidly improved by receiving glucocorticoid therapy. We do a literature review about neurological manifestations observed in AOSD patients.  相似文献

From the roots of Securidaca inappendiculata, one new sterol glycoside securisteroside (1) has been isolated, along with two known sterols, spinasterol (2) and 3-O-beta-D-glucopyranosyl-spinasterol (3). AIM: To detect the possible role of interleukin (IL)-26 in diabetic retinopathy (DR) patients. METHODS: Subjects were divided into diabetes without retinopathy (DWR) group (n=20), non-proliferative diabetic retinopathy (NPDR) group (n=20), proliferative diabetic retinopathy (PDR) group (n=20) and normal control group (n=20). The protein expression of IL-26 in the serum and vitreous fluid were measured by enzyme-linked immunosorbent assay (ELISA). The mRNA change of IL-26 in peripheral blood mononuclear cells (PBMCs) was assessed by real-time polymerase chain reaction. RESULTS: The serum expression of IL-26 in PDR group was significantly elevated compared with the normal control group, DWR group and NPDR group. The vitreous fluid concentration of IL-26 in PDR patients (without anti-VEGF therapy) was also higher compared to normal controls. However, no obvious significance was found concerning the expression of IL-26 in vitreous fluid between PDR after anti-VEGF therapy and normal controls. In PDR group, the mRNA level of IL-26 significantly increased compared with the normal controls and DWR patients in the PBMCs. CONCLUSION: Protein and mRNA expression of IL-26 are increased in serum, vitreous fluid and PBMCs in PDR patients, suggesting that IL-26 may be associated with the pathogenesis of PDR.  相似文献

目的 喉外伤是耳鼻咽喉科的急重病,本研究的目的 是探讨喉外伤的临床特点及其相应的治疗措施.方法 回顾性研究122例喉外伤的临床观察及治疗.结果 122例患者均救治成活.1例喉返神经损伤致一侧声带固定,57例气管切开术后均顺利拨管,无喉气管狭窄发生.结论 治疗喉外伤患者首先要去除气道阻塞和大出血两大致命因素,早期诊断及妥善治疗是防止喉气管狭窄的关键.  相似文献

Treatment outcome of acute myeloid leukemia (AML) in elderly patients remains unsatisfactory. It has been shown that the infusion of granulocyte colony-stimulating factor-mobilized donor peripheral blood stem cells (G-PBSCs) can enhance graft-versus-leukemia effects and speed hematopoietic recovery. Fifty-eight AML patients aged 60-88 years were randomly assigned to receive induction chemotherapy with cytarabine and mitoxantrone (control group; n = 28) or it plus human leukocyte antigen-mismatched G-PBSCs (G-PBSC group; n = 30). Patients who achieved complete remission received another 2 cycles of postremission therapy with intermediate-dose cytarabine or it plus G-PBSCs. The complete remission rate was significantly higher in the G-PBSC group than in the control group (80.0% vs 42.8%; P = .006). The median recovery times of neutrophils and platelets were 11 days and 14.5 days, respectively, in the G-PBSC group and 16 days and 20 days, respectively, in the control group after chemotherapy. The 2-year probability of disease-free survival was significantly higher in the G-PBSC group than in the control group (38.9% vs 10.0%; P = .01). No graft-versus-host disease was observed in any patient. Persistent donor microchimerism was successfully detected in all of the 4 female patients. These results indicate that G-PBSCs in combination with conventional chemotherapy may provide a promising treatment method for AML in elderly patients.  相似文献

Streptococcus mutans is a common Gram-positive bacterium and plays a significant role in dental caries. Tobacco and/or nicotine have documented effects on S. mutans growth and colonization. Sortase A is used by many Gram-positive bacteria, including S. mutans, to facilitate the insertion of certain cell surface proteins, containing an LPXTGX motif such as antigen 1/11. This study examined the effect of nicotine on the function of sortase A to control the physiology and growth of S. mutans using wild-type S. mutans NG8, and its isogenic sortase-defective and -complemented strains. Briefly, the strains were treated with increasing amounts of nicotine in planktonic growth, biofilm metabolism, and sucrose-induced and saliva-induced antigen I/ll-dependent biofilm formation assays. The strains exhibited no significant differences with different concentrations of nicotine in planktonic growth assays. However, they had significantly increased (P~〈 0.05) biofilm metabolic activity (2- to 3-fold increase) as the concentration of nicotine increased. Furthermore, the sortase-defective strain was more sensitive metabolically to nicotine than the wild-type or sortase-complemented strains. All strains had significantly increased sucrose-induced biofilm formation (2- to 3-fold increase) as a result of increasing concentrations of nicotine. However, the sortase-defective strain was not able to make as much sucrose- and saliva-induced biofilm as the wild-type NG8 did with increasing nicotine concentrations. These results indicated that nicotine increased metabolic activity and sucrose-induced biofilm formation. The saliva-induced biofilm formation assay and qPCR data suggested that antigen 1/11 was upregulated with nicotine but biofilm was not able to be formed as much as wild-type NG8 without functional sortase A.  相似文献
