Shigellae infect human intestine and cause intense inflammation and destruction of colonic and rectal mucosa. To model the interactions of shigella with human intestine in vivo, we have studied shigella infection in human intestinal xenografts in severe combined immunodeficient mice (SCID-HU-INT mice). Inoculation of shigella into human intestinal xenografts caused severe inflammation and mucosal damage, which was apparent as soon as 4 h following infection. Shigella infection was associated with human intestinal production of interleukin-1B (IL-1B) and IL-8 and a marked neutrophil influx into the graft. Depletion of neutrophils from SCID-HU-INT mice reduced inflammation in the human intestinal xenograft in response to shigella infection but failed to significantly alter tissue damage. However, the number of intracellular bacteria was more than 20-fold higher in the human intestinal xenografts from neutrophil-depleted SCID-HU-INT mice. Infection of human intestinal xenografts with an attenuated vaccine strain of shigella (CVD1203) induced lower levels of IL-1B and IL-8 than wild-type shigella and caused only moderate damage to the intestinal permeability barrier. Our studies establish the SCID-HU-INT mouse as a viable model for studying the interactions between shigella and human intestine and indicate that neutrophils are important for controlling the invasion of human intestine by shigella.  相似文献

Lymphangioma is an abnormal collection of lymphatics that are developmentally isolated from the normal lymphatic system. Lymphangioma rarely presents as a solitary pulmonary lesion. We report a rare case of intrapulmonary cystic lymphangioma involving the upper lobe of the right lung, which presented with dyspnea in a 2-month-old infant. High-resolution computed tomography (HRCT) of the chest demonstrated a well-circumscribed, multiseptate, cystic lesion in the upper lobe of the right lung, mimicking the feature of type I congenital cystic adenomatoid mal-formation. The tumor was removed by bilobectomy of the upper and middle lobes of the right lung, and its pathologic examination confirmed the diagnosis of an intra-pulmonary cystic lymphangioma.  相似文献

目的:制备并鉴定抗Ig融合蛋白Fc段的单克隆抗体(mAb),建立用于检测Ig融合蛋白的夹心ELISA法和纯化Fc融合蛋白的亲和层析法。方法:以hBCMA—Ig融合蛋白为抗原免疫BALB/c小鼠,通过细胞融合制备抗Fc段mAb,用ELISA等方法鉴定mAb的Ig亚类、表位以及种属特异性,建立用于检测Ig融合蛋白的夹心ELISA法;Western blot检测mAb对变性Ig融合蛋白的反应性。将mAb与Sepharose4B交联,制备亲和层析柱,对LAIR1-Ig融合蛋白进行纯化。结果:获得7株稳定分泌抗Fc段mAb的杂交瘤(FMUFcl-FMUFc7)。利用FMUFc4作为包被mAb,FMUFc5作为酶标记mAb,成功地建立了检测Ig融合蛋白的ELISA法,敏感度达到2μg/L;在7株mAb中,FMUFc6可用于Ig融合蛋白的western blot检测。用FMUFc 6mAb制备的亲和层析柱,可有效地纯化LAIRl—Ig融合蛋白。结论:成功地制备了抗Ig融合蛋白Fc段的mAb,建立了可用于Ig融合蛋白检测和纯化的方法,为Ig融合蛋白的应用提供了有力手段。  相似文献

In our study of the effects of hyposmotic swelling on the Ca(2+)-activated potassium currents [I(K(Ca))] and its mechanism, we employed the whole-cell patch clamp technique using the gastric antral circular myocytes of the guinea-pig. Hyposmotic swelling efficiently increased I(K(Ca)), and the extent of changes in I(K(Ca)) was sharply dependent on the osmolarity of the perfusion solutions. When the calcium-free solution (EGTA 10 microM added in calcium-free solution) was superfused, I(K(Ca)) was not increased by the hyposmotic swelling. Gadolinium (Gd(3+)) 100 nM, a blocker of the stretch-activated nonselective cation channel, blocked the activation of I(K(Ca)) induced by hyposmotic swelling, but nicardipine 5 microM (the L-type calcium channel blocker) did not. Heparin 3 mg/ml, a potent inhibitor of inositol triphosphate receptor (InsP(3)R), did not inhibit the response, and caffeine 1 mM (the agonist for calcium-induced calcium release [CICR]) imitated the effect of hyposmotic swelling. Ryanodine (15 microM), markedly inhibited the effect. These results suggest that hyposmotic swelling activates I(K(Ca)), and the activation is associated with CICR, which is triggered by extracellular calcium influx through the stretch-activated channel (SA channel).  相似文献

ObjectiveTo understand the mechanism of liver cirrhosis after the infection of hepatitis B virus.MethodsMouse fibroblast NIH3T3 cells were transfected with 3.2 kb HBV DND by exposure of the cells to calcium phosphate.The change of the levels of mRNA for tissue inhibitor of metalloproteinase 1and 2(TIMP1,2) was detected in mouse fibroblast NIH3T3 cells and the cells of transfection with HBV Genome by in situ hybridization.ResultsThe levels of mRNA for TIMP1 and TIMP2 were increased significantly.ConclusionHBV infection can induce the expression of the mRNA for TIMP1 and TIMP2.  相似文献

The incidence of invasive diseases, including meningitis caused by Haemophilus influenzae type b (Hib) was markedly decreased after routine immunization of Hib vaccine through diverse schedules in many countries. The purpose of this study was to evaluate the immunogenicity and safety of Hib conjugate vaccines in Korean children before the implementation of a national immunization program against Hib in Korea. A multicenter controlled trial was performed on two different Hib vaccines in Korean children. A total of 319 infants were enrolled: 199 infants were immunized with the Hib polysaccharide conjugated to the tetanus toxoid (PRP-T) and 120 infants with the Hib polysaccharide conjugated to the outer-membrane protein of Neisseria meningitides (PRP-OMP). Immunogenicity was evaluated by enzyme-linked immunosorbent assay (ELISA) and serum bactericidal assay. Both vaccines showed good immunologic responses after primary immunization. After 2 doses of PRP-T or PRP-OMP, 78.9% and 91.7% of infants achieved an antibody level of ≥1.0 µg/mL, respectively. Both vaccines were safe and well-tolerated. No serious adverse events were observed. Thus, Hib conjugate vaccines appear to be safe and show good immunogenicity in Korean infants. These results will be important reference data for the implementation of Hib vaccine in the national immunization program of Korea.  相似文献

我们对采用PCR和聚丙烯酰胺凝胶电泳鉴别出的37例DNA完全来自父方的遗传学完全性葡萄胎(g-CHM)进行基因组中CTTtriplex系统各基因座位(CSF1PO、TPOX和TH01)等位基因分布的分析,并初步研究了这些等位基因分布与临床预后的关系。结果显示,在37例g-CHM中,CSF1PO座位中3个等位基因(11,12和14)和TPOX座位中的1个等位基因(11)的出现率与它们在北京地区人群中的基因频率差异显著;g-CHM中CSF1PO、TPOX和TH01基因座位杂合度显著低于北京地区人群的杂合度(P值均远小于0.01);在23例良性g-CHM和10例侵袭性g-CHM中:CSF1PO座位的等位基因10、11在良性中的出现率高于在侵袭性g-CHM中(P=0.026148),等位基因12在良性中的出现率低于在侵袭性g-CHM中(P=0.023879);TPOX座位的等位基因8在良性中的出现率高于在侵袭性g-CHM中(P=0.004322),而等位基因11在良性中的出现率低于其在侵袭性g-CHM中(P=0.008671)。上述结果提示,葡萄胎基因组中存在某些等位基因分布与在人群中的分布不同,而且葡萄胎是否具有侵袭性也和某些等位基因的出现率过高或过低有相关性,这些与侵袭性有关的等位基因可能成为预测葡萄胎是否具有侵袭性的标志物。  相似文献

目的:探讨在体外葡萄糖对SD大鼠腹膜巨噬细胞诱导型一氧化氮合酶(iNOS)mRNA的表达及蛋白的合成。方法:取雄性SD大鼠腹腔巨噬细胞培养,培养的巨噬细胞分为5组:(1)正常对照组;(2)30 mmol/L甘露醇组;(3)90 mmol/L甘露醇组;(4)30 mmol/L葡萄糖组;(5)90 mmol/L葡萄糖组。每组6个培养孔,24 h后提取细胞RNA及蛋白,分别做RT-PCR及Western blotting以检测腹膜巨噬细胞iNOS mRNA的表达及蛋白的合成。结果:30 mmol/L及90 mmol/L甘露醇刺激腹膜巨噬细胞iNOS mRNA的表达及蛋白的合成均不明显, 30 mmol/L及90 mmol/L葡萄糖能明显刺激腹膜巨噬细胞iNOS mRNA的表达及蛋白的合成,90 mmol/L葡萄糖与30 mmol/L的葡萄糖相比更能刺激巨噬细胞iNOS mRNA的表达及蛋白的合成。结果:葡萄糖能明显刺激SD大鼠腹膜巨噬细胞iNOS mRNA的表达及蛋白的合成。  相似文献

目的:观察睡眠剥夺(SD)后大鼠脑组织HSP70表达的变化及对超微结构的影响。方法:44只雄性SD大鼠随机分为11组,每组4只,免疫组织化学方法检测HSP70的表达,电镜观察海马超微结构的变化。结果:睡眠剥夺后12小时即可在大脑皮质及海马观察到HSP70阳性细胞,2—3天数量达到高峰,7天时明显下降。白天睡眠剥夺12小时(SDd12h)组HSP70阳性细胞数较夜晚睡眠剥夺12小时(SDn12h)组多(P〈0.05)。RS组大脑皮质HSP70阳性细胞数较白天睡眠剥夺1天(SDd1d)组减少(P〈0.05)。白天睡眠剥夺3天(SDd3d)海马出现超微结构改变,白天睡眠剥夺7天(SDd7d)后改变更加明显。结论:睡眠剥夺可影响大鼠脑组织HSP70表达及超微结构。  相似文献

The modification of hyaluronan (HA) and gelatin using dithiobis(propanoic dihydrazide) (DTP) has provided two thiolated macromolecular components of the extracellular matrix (ECM), specifically HA-DTPH and gelatin-DTPH. Blends of these thiolated ECM components were crosslinked in air to form hydrogels that were interpenetrating disulfide-crosslinked networks. Lyophilization of the hydrogels afforded sponge-like macroporous scaffolds suitable for cell attachment and proliferation. Increasing percentages of gelatin-DTPH (0, 25, 50, and 75%) were blended with HA-DTPH, and the resulting sponges were evaluated in vitro and in vivo as scaffolds for tissue engineering by seeding with human tracheal scar (HTS) fibroblasts. While cells failed to attach and grow in HA-only sponges, the gelatin-modified HA sponges promoted cell adhesion, proliferation, and spreading in vitro. Optimal attachment and growth was observed with 50% gelatin-HA sponges. Cell attachment to the gelatin-HA sponge could be blocked by preincubation of cells with a soluble fibronectin peptide Gly-Arg-Gly-Asp (GRGD). Finally, HTS fibroblast-seeded gelatin-HA sponges were implanted into the flanks of nude mice and evaluated at 2 and 8 weeks postimplantation. The sponges were fully biocompatible and new fibrous tissue formed, gradually replacing the sponge-like scaffold. The gelatin-HA sponges act as synthetic, macroporous, covalent mimics of the ECM and constitute novel scaffolds for cell growth and tissue augmentation.  相似文献
