目的分析de novo早期结直肠癌的临床病理特征, 并评估内镜治疗的效果。方法纳入2020年6月—2022年5月在首都医科大学附属北京友谊医院行内镜切除且术后病理确诊为de novo早期结直肠癌的患者。回顾性收集患者的基本资料、内镜表现、治疗方式、术后病理结果以及转归等资料。结果共纳入33例de novo早期结直肠癌患者, 年龄(62.67±8.62)岁, 男女比例7.25∶1。病变长径(0.96±0.36)cm;病变形态以浅表型(0-Ⅱ型)为主, 占72.7%(24/33)。29例采用内镜黏膜下剥离术切除, 4例采用内镜黏膜切除术切除。术后病理显示, 11例(33.3%)为高分化管状腺癌, 其中2例侵及黏膜下浅层;20例(60.6%)为中分化管状腺癌, 其中5例侵及黏膜下浅层, 15例侵及黏膜下深层;2例(6.1%)为中-低分化管状腺癌, 均侵及黏膜下深层。病变浸润深度与病变分化程度有显著相关性(P<0.001), 中分化及中-低分化病变更容易发生黏膜下层深浸润。33例(100.0%)病变均达到整块切除, 97.0%(32/33)达到完全切除, 42.4%(14/33)达到治愈... 相似文献

Catecholaminergic polymorphic ventricular tachycardia (CPVT) is an inherited arrhythmia syndrome associated with Ca2+ leak predominantly caused by ryanodine receptor 2 (RyR2) mutations. We identified a R1760W-RyR2 mutation located between the N-terminal domain and the central domain of RyR2 in a CPVT patient by DNA sequencing. Recombinant mutant RyR2−2801mcherry plasmid generated by the overlap extension polymerase chain reaction and seamless cloning was transfected in HEK293 cells for the cell model. Single-cell luminal and cytosolic Ca2+ imaging was measured by endoplasmic reticulum (ER) luminal Ca2+-sensitive protein D1ER and Fura-2 AM on a confocal laser scanning microscope, respectively. We found that in RyR2 mutant cells, the propensity for store-overload-induced Ca2+ release (SOICR) was enhanced representing increased Ca2+ oscillations, reduced activation and termination thresholds of spontaneous Ca2+ release; and the sensitivity to cytosolic Ca2+ activation was increased manifesting reduced steady state ER Ca2+ levels. Our results indicated that R1760W-RyR2 mutation induced calcium leak, representing a gain of function. Further, antiarrhythmic drugs propafenone and flecainide significantly suppressed SOICR caused by the R1760W-RyR2 mutation at a concentration of 20 μM, which was lower than the concentration at which carvedilol suppressed SOICR. 相似文献

Ferroptosis is a newly characterized form of regulated cell death. This bibliometric analysis identified the scientific output, leading institutions and research teams, current research hotspots and trends in research on ferroptosis since the origin of the concept. We searched the Science Citation Index Expanded of Web of Science Core Collection for papers on ferroptosis up to 3 June 2022. The acquired data were analysed and visualized by Bibliometrix package and VOSviewer. The study ultimately included 3511 relevant papers, and annual production in this field has grown rapidly in recent years. Institutions and scholars from China contributed the most work, but the impact of their research was much less than that of the United States. Prof. Brent R. Stockwell's team from Columbia University in the United States has a very strong academic influence in the field. Front Cell Dev Biol published the most papers in the field of ferroptosis. As the keywords of the papers in this field changed from the most numerous ‘oxidative stress’, ‘cell-death’, ‘iron’, ‘expression’, and ‘lipid-peroxidation’, to ‘prognosis’, ‘immunotherapy’, ‘progression’, ‘tumour microenvironment’, and ‘colorectal cancer’, the hotspot of ferroptosis research is gradually shifting from basic research to clinical translational research. Information regarding the impact of the coronavirus disease 2019 (COVID-19) pandemic on cervical cancer in mainland China is lacking. We explored its impact on the hospital attendance of patients with primary cervical cancer. We included 1918 patients with primary cervical cancer who initially attended Harbin Medical University Cancer Hospital between January 23, 2019, and January 23, 2021. Attendance decreased by 31%, from 1135 in 2019 to 783 in 2020, mainly from January to June (𝜒2 = 73.362, P < .001). The percentage of patients detected by screening decreased from 12.1% in January-June 2019 to 5.8% in January-June 2020 (𝜒2 = 7.187, P = .007). Patients with stage I accounted for 28.4% in 2020 significantly lower than 36.6% in 2019 (𝜒2 = 14.085, P < .001), and patients with stage III accounted for 27.1% in 2020 significantly higher than 20.5% in 2019 (𝜒2 = 11.145, P < .001). Waiting time for treatment was extended from 8 days (median) in January-June and July-December 2019 to 16 days in January-June (𝜒2 = 74.674, P < .001) and 12 days in July-December 2020 (𝜒2 = 37.916, P < .001). Of the 179 patients who delayed treatment, 164 (91.6%) were for the reasons of the healthcare providers. Compared to 2019, the number of patients in Harbin or non-Harbin in Heilongjiang Province and outside the province decreased, and cross-regional medical treatment has been hindered. The COVID-19 pandemic has negatively impacted cervical cancer patient attendance at the initial phase. These results are solid evidence that a strategy and mechanism for the effective attendance of cervical cancer patients in response to public health emergencies is urgently needed. 相似文献
