Using K562 and HL60 cell lines, we have investigated the anti-tumoral activity of d-limonene, a monocyclic monoterpene, in human leukemia cells. Apoptosis was evaluated by Hoechst staining and by the annexin V/propidium iodide binding assay. d-Limonene induced apoptosis in a dose- and time-dependent manner in both cell lines. Our findings and data, demonstrating an increase in Bax protein expression, the release of cytochrome c from mitochondria, and an increase in caspase-9 and cleaved caspase-3, but not caspase-8, after the treatment of d-limonene, all suggest that the mitochondrial death pathway is primarily involved in the development of d-limonene-induced apoptosis. 相似文献

目的:探讨与中性粒细胞异常增殖相关的BCR/ABL-慢性白血病的临床特征.方法:报告3例不同类型与中性粒细胞异常增殖相关的BCR/ABL-慢性白血病并结合文献进行讨论.结果:3例均为老年男性,表现为以中性粒细胞为主的白细胞异常增多、BCR/ABL融合基因阴性.例1诊断为慢性中性粒细胞白血病(CNL),脾大,外周血以成熟中性粒细胞增高为特征,胞浆内中毒颗粒易见,无明显病态造血,中性粒细胞碱性磷酸酶(NAP)染色强阳性,骨髓粒系增生明显活跃.例2诊断为不典型慢性髓细胞白血病(aCML),血涂片示不成熟粒细胞比例增高伴粒系病态造血,尤其核染色质异常浓聚更为明显,骨髓象表现为髓系增生和病态造血,原始细胞轻度增多.例3诊断为慢性粒单核细胞白血病(CMML),外周血成熟单核细胞数量和比例异常偏高,粒系病态如核分叶过多、环状核和假性Pelger-Huet畸形等易见,骨髓髓系增生明显活跃伴病态造血,无原始细胞增多.结论:中性粒细胞异常增殖相关的BCR/ABL-慢性白血病较为少见,易于误诊,细胞形态学特征是诊断BCR/ABL-慢性白血病的基础.临床表现、多种实验室检查结果尤其是分子生物学等有助于正确诊断这类疾病. 相似文献

目的:观察中药在烧伤重症阶段对烧伤感染指标、抗菌药物使用和耐药菌出现的影响。方法:选取108例重症烧伤患者,使用回顾性的方法,根据患者中药使用情况,将患者分为中药+西医治疗的综合治疗组(综合组) 60例,及单纯西医治疗组(西医组) 48例,根据病例及医嘱系统的内容,提取患者的治疗资料进行分析,对比2组患者白细胞(WBC)、超敏C-反应蛋白(hs-CRP)、前降钙素(PCT)、急性胃肠功能损伤(AGI)评分、重症监护室(ICU)住院率、抗菌药物使用情况。结果:与西医组比较,综合组WBC、hs-CRP、PCT、AGI评分、ICU住院率等指标均较优,但仅WBC、AGI评分比较,差异有统计学意义(P<0.05)。与西医组比较,综合组抗菌药物使用强度(AUD)较低,非限制级抗菌药物累计用药频度(DDDs)占所在组总DDDs比率较高,而限制级、特殊使用级抗菌药物的DDDs占比较低,差异有统计学意义(P<0.05);综合组多重耐药菌总出现率较低,但差异无统计学意义(P>0.05)。火毒炽盛、热入营血、阴阳两虚为烧伤重症阶段的主要证候,中药以清热凉血、养阴益气药物为主。结论:中西医结... 相似文献

目的:探索构建消毒供应中心护理质量敏感性指标,并分析其应用效果。方法:成立消毒供应中心护理质量敏感性指标研究小组,确定器械清洗合格率、器械包装合格率、包装破损率、湿包发生率作为消毒供应中心护理质量敏感性指标,并进行护理质量控制与改进。比较观察组(实施后)、对照组(实施前)敏感指标的差异。结果:与对照组比较,观察组临床科室器械、手术器械的器械清洗合格率(98.1%VS 94.3%、98.6%VS 97.1%)、器械包装合格率(98.3%VS 93.5%、98.1%VS 94.4%)均明显升高,包装破损率(0.22%VS 0.34%、0.22%VS 0.41%)、湿包发生率(0.27%VS 0.37%、0.22%VS 0.34%)明显下降(P<0.05)。观察组医务人员对器械的满意率较对照组(97.0%VS 90.0%)显著提升(P<0.05)。结论:构建消毒供应中心护理质量敏感性指标对消毒供应中心医疗器械实施全程标准化质量控制管理,可促进护理质量的持续改进及护理服务满意度提升。 相似文献

ContextAcute ischaemic stroke (AIS) is a major cause of disability and death, which is a serious threat to human health and life. Wasp venom extracted from Vespa magnifica Smith (Vespidae) could treat major neurological disorders.ObjectiveThis study investigated the effects of wasp venom on AIS in rats.Material and methodsWe used a transient middle cerebral artery occlusion (MCAO) model in Sprague-Dawley rats (260–280 g, n = 8–15) with a sham operation group being treated as negative control. MCAO rats were treated with wasp venom (0.05, 0.2 and 0.6 mg/kg, i.p.) using intraperitoneal injection. After treatment 48 h, behavioural tests, cortical blood flow (CBF), TTC staining, H&E staining, Nissl staining, TUNEL assay, immunohistochemistry (IHC) and ELISA were employed to investigate neuroprotective effects of wasp venom.ResultsCompared with the MCAO group, wasp venom (0.6 mg/kg) improved neurological impairment, accelerated CBF recovery (205.6 ± 52.92 versus 216.7 ± 34.56), reduced infarct volume (337.1 ± 113.2 versus 140.7 ± 98.03) as well as BBB permeability as evidenced by changes in claudin-5 and AQP4. In addition, function recovery of stroke by wasp venom treatment was associated with a decrease in TNF-α, IL-1β, IL-6 and inhibition activated microglia as well as apoptosis. Simultaneously, the wasp venom regulated the angiogenesis factors VEGF and b-FGF in the brain.ConclusionsWasp venom exhibited a potential neuroprotective effect for AIS. The cell cycle inhibitor P21 has been implicated in cell senescence and plays an important role in the injury–repair process following lung injury. Pulmonary fibrosis (PF) is a fibrotic lung disorder characterized by cell senescence in lung alveolar epithelial cells. In this study, we report that P21 expression was increased in alveolar epithelial type 2 cells (AEC2s) in a time-dependent manner following multiple bleomycin-induced PF. Repeated injury of AEC2s resulted in telomere shortening and triggered P21-dependent cell senescence. AEC2s with elevated expression of P21 lost their self-renewal and differentiation abilities. In particular, elevated P21 not only induced cell cycle arrest in AEC2s but also bound to P300 and β-catenin and inhibited AEC2 differentiation by disturbing the P300–β-catenin interaction. Meanwhile, senescent AEC2s triggered myofibroblast activation by releasing profibrotic cytokines. Knockdown of P21 restored AEC2-mediated lung alveolar regeneration in mice with chronic PF. The results of our study reveal a mechanism of P21-mediated lung regeneration failure during PF development, which suggests a potential strategy for the treatment of fibrotic lung diseases. 相似文献
