本文采用负染和超薄切片技术在电镜下的观察,研究了国产头孢三嗪在高浓度下和亚最低抑菌浓度下对金葡菌、大肠杆菌和绿脓杆菌超微结构的作用。结果显示,10MIC的头孢三嗪作用3小时即可导致细菌发生不可逆性改变,出现细胞膜皱缩,胞质稀疏,细胞膜连续性丧失,甚至细胞破裂,解体死亡。经Sub-MIC的头孢三嗪作用后,金葡菌呈现菌体增大,横膈增宽;而大肠杆菌和绿脓杆菌则表现为菌体增长和菌丝体形成,胞内出现大小不一的空泡,电子密度下降.本研究证实了头孢三嗪为一杀菌类抗生素;提示本药对细菌的形态学改变,主要由于和PBP_3结合所致。 相似文献

The possibility of using commercial bayberry tannin (BT) from a Chinese source as a cross-linker and functional additive to develop soybean protein isolate (SPI)-based films was explored in this study by using the solvent casting method. In particular, the impacts of BT loading on the tensile strength, microstructure, thermal stability, water resistance and antioxidant capacity were fully investigated. The results reveal that SPI incorporated with BT yielded a phenolic–protein hybrid whose relevant films exhibited an improvement in tensile strength of around two times greater compared with native SPI as a result of the formed interactions and covalent cross-links, which could be proven using FTIR spectroscopy. The introduction of BT also led to the compact microstructure of SPI–BT films and enhanced the thermal stability, while the water vapor permeability was reduced compared with the control SPI film, especially at high loading content of tannin. Additionally, the use of BT significantly promoted the antioxidant capacity of the SPI-based films according to DPPH radical scavenging assay results. On this basis, Chinese bayberry tannin is considered a promising natural cross-linker and multifunctional additive that can be dedicated to developing protein-derived films with antioxidant activity for food packaging applications. 相似文献

目的:探讨青光眼滤过性手术浅前房的发生、发展和预防处理。方法:回顾性研究了我院过去102例(112眼),其中急性闭角型青光眼64例(67眼),慢性闭角型青光眼7例(11眼),开角型青光眼26例(29眼),继发性青光眼5例(5眼)的治疗经过。结果:112眼中术后24h有77眼前房形成,占68.8%,发生I型浅前房23眼,II型9眼,III型3眼,共占32.2%,其中5眼分别于术后2~4d变浅。荧光素试验发现结膜切口渗漏者3眼,有较明显睫状体脉络膜脱离5眼,小眼球并发晶状体膨胀2眼,恶性青光眼1眼,均经相应处理后前房恢复。结论:青光眼滤过性手术浅前房的发生率是较高的,但只要加强手术前后的治疗处理,术中仔细操作,多数浅前房是可以预防的,即使发生了浅前房只要处理及时和恰当,预后也是良好的。 相似文献

目的:通过研究脑梗死发生后脑动脉血管平滑肌中与血管收缩相关的PKC通路中关键蛋白CaP、CaD的表达变化及针刺干预效应,研究针刺对脑梗死大鼠脑血管平滑肌CaP、CaD变化的动态调节规律。方法:以线栓法复制脑梗死大鼠模型,以脑表面血管为研究对象,应用免疫印迹法检测脑血管平滑肌CaP、CaD的变化规律和电针干预效应。结果:免疫印迹结果显示:与空白组相比模型组MCAO后各时相CaD、CaP表达量下调均有显著统计学差异(P<0.01或P<0.05);模型组和针刺组组间比较显示,针刺组CaD、CaP表达量均上调,组间比较均有显著统计学差异(P<0.01,P<0.05)。结论:针刺干预可显著抑制MCAO模型大鼠脑表面血管CaD、CaP表达下调趋势,使其维持在正常值左右,这对于缓解缺血后血管平滑肌痉挛,保证缺血区血液供应具有重要作用。 相似文献

The centrosome, an organelle that functions as the major microtubule-organizing center, plays an essential role in the formation of the mitotic spindle and guiding accurate chromosome segregation. Centrosome aberrations are frequently associated with various forms of human cancers and it is thought that defects in this organelle contribute to genomic instability and malignant transformation. We recently identified and characterized a centrosome-localized protein complex that is comprised of Su48 and Nde1. Disruption of the normal function of these proteins leads to abnormal cell division. To extend our understanding of how this protein complex operates, we sought to identify Nde1-interacting molecules by the yeast two-hybrid screening method. Here, we demonstrate that both Nde1 and Su48 can associate with p78/MCRS1, a protein implicated in cancer development. We found that, whereas the majority of p78 localizes to the nucleus as reported in earlier studies, a fraction of the p78 protein can be detected in the centrosome. Moreover, we determined that a region containing the forkhead-associated domain of p78 is involved in association with Nde1 and Su48, as well as in centrosomal localization. We also provide evidence that the association between p78 and Nde1 is regulated by phosphorylation on Nde1. Furthermore, abrogation of the endogenous p78 function by small interfering RNA knockdown causes cell death and a modest delay in mitosis. These results indicate that a subset of the p78 proteins comprises a component of the centrosome and that p78 is essential for cell viability. 相似文献

In order to search for structurally novel and bioactive natural compounds from microorganisms, a halotolerant fungal strain, Penicillium citrinum B-57, which mainly produces citrinin derivatives, was isolated from sediments collected from the Jilantai salt field. From the ethyl acetate extract of P. citrinum B-57, two new citrinin dimers, pennicitrinone C ( 1) and penicitrinol B ( 2), and 11 known related compounds were isolated and identified by spectroscopic and chemical methods.
