GNAS gene mutations affecting XLαs and bone health: A long neglected relationship

The GNAS locus is an imprinted site. The α-subunit of the stimulatory G protein (Gsα) and extralarge variant (XLαs) are the two important products of the GNAS locus. The abnormal expression of Gsα is associated with pseudohypoparathyroidism (PHP) and related disorders, including Albright hereditary osteodystrophy (AHO), pseudopseudohypoparathyroidism (PPHP), and progressive osseous heteroplasia (POH). XLαs protein can mimic the catalytic intracellular synthesis of cyclic adenosine monophosphate (cAMP) by Gsα in response to parathyroid hormone (PTH) stimulation, which may be involved in the pathogenesis of PPHP and POH in patients with paternal GNAS defects. A paternally inherited nonsense variant in the first exon of XLαs in an adult patient may be associated with fractures and osteopetrosis. The relationship between the XLαs product of the GNAS locus and bone remodeling may have been overlooked. Here, we summarize the phenotypes of genetic mouse models and clinical cases of XLαs variations and suggest that the abnormal paternal expression of XLαs may be associated with the development of POH and affect osteoblast and osteoclast differentiation.     

Early warning signals for Omicron outbreaks in China: A retrospective study

The Omicron variant has become the dominant COVID-19 variant worldwide due to its rapid and cryptic spread. Therefore, successful early warning is of great importance to be able to control epidemics in their early phase, before developing into large outbreaks. COVID-19-related Baidu search index, which reflects human behavior to a certain degree, was used to retrospectively detect the warning signs for Omicron variant outbreaks in China in 2022. The characteristics and effects of warning signs were analyzed in detail. We detected the presence of early warning signs (both high and low thresholds) and found that these occurred 4–7 days earlier than traditional epidemiological surveillance and >20 days earlier than the implementation of the local “lockdown” policy. Compared with the “high threshold” warning, the early warning effect of the “low threshold” is also vital because it indicates a negligence about epidemic prevention and control. However, there is obvious heterogeneity in the optimal threshold for detecting early warning signs and their distribution in different cities. Multi-source and multi-point early warning systems should be established via combining internet-based big data in the future to conduct effective and early real-time warning. This would create precious time for the early control of COVID-19 outbreaks.     

PTF—V1在二尖瓣狭窄球囊扩张术中的临床效果分析

目的 探讨二尖瓣狭窄患者行经皮二尖瓣球囊扩张术（ＰＢＭＶ０前后心电图ＰＴＦ－Ｖ１的变化及其与血流动力学各指标的相关性。方法 分析２９例窦性心律的二尖瓣狭窄患者ＰＢＭＶ前后ＰＴＦ－Ｖ１的变化,并与二尖瓣面积（ＭＶＡ）、左房平均压（ＭＬＡＰ）和左房内径（ＬＡＤ）作相关分析。结果 ＰＢＭＶ术后ＰＴＦ－Ｖ１显著改善（Ｐ〈０．０１）,与ＭＬＡＰ、ＬＡＤ呈显著正相关,与ＭＶＡ呈显著负相关（Ｐ〈０．０５）。结论     

含CpG寡聚脱氧核苷酸对肿瘤抗原肽P815AB的免疫佐剂效应研究

目的 以肿瘤抗原Ｐ８１５ＡＢ抗原肽为模式抗原,研究ＣｐＧ寡聚脱氧核苷酸（ＣｐＧ ＯＤＮ）对该肿瘤抗原表位肽的免疫佐剂效应。方法 用［＾３Ｈ］－ＴｄＲ掺入法研究ＣｐＧ ＯＤＮ体外刺激小鼠脾细胞增殖效应,用ＥＬＩＳＰＯＴ和［＾３Ｈ］－ＴｄＲ释放法检测不同构成形式的疫苗在体诱发特异性ＣＴＬ应答的效应。结果 ＯＤＮ１８２６在体外可刺激小鼠脾细胞显著增殖,经４次免疫,在“肽＋ＯＤＮ１８２６＋ＩＦＡ”组中,４     

小鼠肥大细胞瘤P815特异抗原P1A的克隆与表达

目的 克隆小鼠肥大细胞瘤Ｐ８１５细胞株的ＰＩＡ基因以制备肿瘤ＤＮＡ疫苗,方法 用ＲＴ－ＰＣＲ方法制备ＰＩＡ基因,以哺乳细胞高效表达质粒ｐＣＩ－ｎｅｏ为载体,构建重组ＤＮＡ疫苗。重组体用克隆位点上游和下游的Ｔ７和Ｔ１３启动了序列为测序引物,用自动测序仪测定鉴定克隆的正确性。再将鉴定过的重组质粒用磷酸钙法转化２９３细胞,用ＲＴ－ＰＣＲ法鉴定转化细胞中ＰＩＡ基因的表达。结果 正确构建了ＰＩＡ／ｐＣＩ－ｎ     

高血压病及糖尿病患者肾血流的彩色多普勒速度能量图评价及其影响因素分析

充血性心力衰竭患者动态血压的变化及意义

目的 探讨慢性充血性心力衰竭（ＣＨＦ）患者２４ｈ动态血压的变化及意义。方法 将５７例ＣＨＦ患者按心功能的受损程度（ＮＹＨＡ分组）分为Ａ组（１７例,心功能Ⅱ组）、Ｂ组（１９例、心功能Ⅲ级）和Ｃ组（２１例,心功能Ⅳ级）;根据病因的不同,分为Ｄ组（１６例,扩张型心肌病）和Ｅ组（４１例,非扩张型心肌病）;全部患者行２４ｈ动态血压（２４ｈ ＡＢＰ）检查,比较Ａ,Ｂ,Ｃ三组及Ｄ、Ｃ两组之间平均收缩压、夜间平均收缩压下降率（ｎｓＢＰＲ）及２４ｈ平均收缩压变异率（２４ｈ ｓＢＰＶ）的差异。结果 Ｃ组的平均收缩压较Ａ组和Ｂ组略低,但三组之间差异无显著性（Ｐ〉０．０５）;Ｄ组的平均收缩压显著低于Ｅ组（Ｐ〈０．０５）。Ａ,Ｂ,Ｃ３组的ｎｓＢＰＲ减弱或消失,分别占５８．８％,８８．３％和９５．２％,Ａ组与Ｂ,Ｃ,组的平均收缩压显著低于
