本文报道应用高分子右旋糖酐制成软脑膜微循环障碍的动物实验模型。软脑膜微循环障碍模型的标志;软脑膜微血管血流速度减慢,血液流态呈粒状流或絮状流,血细胞聚集。 将98只家兔分成甲、乙二组。甲组38只,静脉注射平均分子量为25万的高分子右旋糖酐,乙组60只,静脉注射平均分子量为49万的高分子右旋糖酐。实验结果发现,甲组38只家兔仅28只可复制成软脑膜微循环障碍的实验模型,模型复制成功率为73.6%;乙组60只家兔,59只可形成软脑膜微循环障碍实验模型,成功率为98.3%。本实验证明复制动物模型以49万分子量的高分子有旋糖酐为宜。 本实验模型可用于脑微循环障碍和改善脑微循环药物的研究工作。 相似文献

For more than two decades, antisense oligonucleotides (ODNs) have been used to modulate gene expression for the purpose of applications in cell biology and for development of novel sophisticated medical therapeutics. Conceptually, the antisense approach represents an elegant strategy, involving the targeting to and association of an ODN sequence with a specific mRNA via base-pairing, resulting in an impairment of functional and/or harmful protein expression in normal and diseased cells/tissue, respectively. Apart from ODN stability, its efficiency very much depends on intracellular delivery and release/access to the target side, issues that are still relatively poorly understood. Since free ODNs enter cells relatively poorly, appropriate carriers, often composed of polymers and cationic lipids, have been developed. Such carriers allow efficient delivery of ODNs into cells in vitro, and the mechanisms of delivery, both in terms of biophysical requirements for the carrier and cell biological features of uptake, are gradually becoming apparent. To become effective, ODNs require delivery into the nucleus, which necessitates release of internalized ODNs from endosomal compartments, an event that seems to depend on the nature of the delivery vehicle and distinct structural shape changes. Interestingly, evidence is accumulating which suggests that by modulating the surface properties of the carrier, the kinetics of such changes can be controlled, thus providing possibilities for programmable release of the carrier contents. Here, consideration will also be given to antisense design and chemistry, and the challenge of extra- and intracellular barriers to be overcome in the delivery process. 相似文献

Poly (3-phenylthiophene) (P3PhT) films have been electrochemically synthesized by direct oxidation of 3-phenylthiophene in the electrolyte of pure boron trifluoride diethyl etherate (BFEE). The oxidation potential of the monomer was measured to be 1.29 V (vs. SCE), and about 0.15 V lower than that measured in a neutral medium of acetonitrile. Free-standing P3PhT film with tensile strength of 32–40 MPa has been obtained for the first time. The morphology and the structure of the polymer film have been studied by scanning electron microscopy, infrared and Raman spectroscopies. Raman spectral results demonstrated that the doping level of the P3PhT film increased with film thickness during electrochemical growth process. 相似文献

支气管哮喘的本质是气道慢性非特异性炎症,肾上腺糖皮质激素抗炎治疗已成为哮喘现代治疗的主要方法。但约5%-10%的患者对激素治疗不敏感而成为难治性哮喘,这种对激素治疗效果较差的哮喘被称为“激素抵抗性哮喘”(steroid-resistant asthma,SRA)。其发生机制不明,可能与糖皮质激素受体β、基因表达和热休克蛋白异常等有关,免疫和非免疫治疗可以改善哮喘症状。 相似文献

目的 探讨罗格列酮对糖尿病大鼠肾脏的保护作用机制。方法 将大鼠分为正常组、糖尿病组、小剂量罗格列酮组、大剂量罗格列酮组。药物干预4周后,检测各组大鼠的血糖(BS)、三酰甘油(TG)、胆固醇(TC)、肌酐(Scr)水平及尿白蛋白定量(24h)。随后处死大鼠,测定肾组织中丙二醛(MDA)含量、总抗氧化能力(T-AOC)及铜锌超氧化物歧化酶(Cu-ZnSOD)、谷胱甘肽过氧化酶(GSH-Px)、核转录因子-κB(NF-κB)的活性。同时留取。肾脏组织作PAS染色行病理检查。RT-PCR和免疫组化法检测。肾组织中单核细胞化学吸引蛋白质1(MCP-1)mRNA及蛋白质的表达。结果 与糖尿病组相比,大剂量罗格列酮干预组BS、TC、TG差异无统计学意义;而。肾组织MDA含量、MCP-1mRNA及蛋白质表达、NF-κB活性差异有统计学意义(P〈0.05),SOD、GSH-Px、T-AOC活性显著上升;肾小球面积及体积减小。结论大剂量的罗格列酮可显著改善糖尿病大鼠的。肾脏损害,其机制可能与其抗氧化,抑制NF-κB活性,降低MCP-1的含量有关。 相似文献

OBJECTIVE: To investigate the clinicopathlogical significance of cyclinE, cyclinD1, P21WAF1/CIP1 and P27KIP1 expression in squamous cell carcinoma of maxillary sinus. METHODS: The expression of cyclinE, cyclinD1, P21WAF1/CIP1 and P27KIP1 was examined by the method of immunohistochemistry in inflammatory mucosa of maxillary sinus(10), inverting papilloma of maxillary sinus(20) and squamous cell carcinoma of maxillary sinus (50). RESULTS: (1) In inflammatory mucosa, inverting papilloma and squamous cell carcinoma, the positive immunostaining rates of cyclinD1 protein were 10.0%, 25.0%, 48.0% respectively (P < 0.05); cyclinE protein was 20.0%, 35.0%, 58.0% respectively (P < 0.05); P21WAF1/CIP1 protein was 80.0%, 65.0%, 40.0% respectively (P < 0.05) and P27KIP1 protein was 70.0%, 75.0%, 40.0% respectively (P < 0.05). (2) There was no significance between the clinical type, T stage and clinical stage of carcinoma of maxillary sinus and the expression of cyclinD1, cyclinE, P21WAF1/CIP1 and P27KIP1 (P > 0.05). (3) P21WAF1/CIP1 positive expression in highly, moderately and poorly differtiated carcinomas were 73.2% (11/15), 58.9% (11/19), 25.0% (4/16) respectively (P < 0.05). CONCLUSION: (1) With the development of the lesion, the positive immunostaining rate of cyclinE and cyclinD1 protein increases, while the rate of P21WAF1/CIP1 and P27KIP1 protein decreases. (2) P21WAF1/CIP1 expression is associated with tumour cell differentiation. (3) The mutual mechenism of cell cycle regulators plays an important role in the squamous cell carcinoma of maxillary sinus. The quantity and the frequency of four kinds of gene expression increase with the progression of maxillary sinus carcinogenesis. 相似文献
