Acute biphenotypic leukemia followed by two color flow cytometry

In this paper is reported a case of acute biphenotypic leukemia who was treated by chemotherapy and pursued its effect by two color flow cytometry. A 33-year-old male patient was admitted due to fever and general fatigue and diagnosed as acute leukemia by hematological findings. Surface markers were investigated to find positive reaction of Leu 12 (CD19), J 5 (CD10), My 7 (CD13) and My 9 (CD33), in which Leu 12 and My 9 were simultaneously expressed on the same blast cells by two color flow cytometry. He was treated with daunorubicin, enocitabine, mercaptopurine, vincristine, and prednisolone to obtain partial remission. Then, he was administered L-asparaginase, doxorubicin, vincristine and prednisolone to reach complete remission. The effect of chemotherapy was investigated by not only bone marrow puncture, but also by two color flow cytometry. From the findings in this case, the two flow cytometry was proved to be a useful tool for not only diagnosis of acute mixed leukemia, aut also the judgement of the effect of treatment.

von Recklinghausen's disease associated with malignant lymphoma

A 59-year-old man was admitted to the Saga Medical School Hospital in November 1986 because of unconsciousness and lymph node swelling. He had café au leit spots and multiple neurofibromas since his thirties. The leukocyte count was increased. The level of serum LDH and Ca were extremely elevated. His bone marrow aspiration showed the invasion of lymphoma cells. Lymph node biopsy from his left neck and the study of surface markers were performed. And then, his illness was diagnosed as non-Hodgkin's lymphoma (diffuse, medium sized cell type, B cell type, stage IV B.) following von Recklinghausen's disease. After the admission, his general condition improved by chemotherapy, but he had a relapse 6 months later. And he died in November 1987. The case of von Recklinghausen's disease associated with malignant lymphoma in rarely reported. We reviewed the literature and discussed von Recklinghausen's disease associated with malignant lymphoma including this case.     

Accurate multiplex polymerase chain reaction assay for gender determination from a single cell

For gender determination of preimplantation embryos or circulating fetal cells in maternal blood, we developed a multiplex polymerase chain reaction assay from a single cell. This assay which co-amplifies X (DXZ1)- and Y (DYZ1)-specific repeat sequences, yields a 308-bp band in females and two bands of 154 and 308 bp in males. In a randomized, blinded assay of 100 isolated single amniocytes, 99 (99%) were amplified successfully. All 50 of the XY cells were correctly diagnosed as male (100%), whereas 49 of the 50 XX cells were diagnosed as female (98%). This accurate and efficient assay may be applicable in these clinical settings.     

The Effects of Age and Abnormal Sperm Count on the Nondisjunction of Spermatozoa

Purpose: The effect of paternal age on the nondisjunctionof sex chromosomes is controversial. Also, the prevalenceof chromosomal anomalies in infertile patients iscontroversial, it has been reported that the sex chromosomalaneuploidy rate following treatment with intracytoplasmic sperminjection (ICSI) is higher than in naturally conceivedpregnancies. We investigated the influence of paternal age andoligozoospermia on the nondisjunction of spermatozoa. Methods: We determined the rate of aneuploidy forgonosomes and autosomes, using two-color fluorescence in situhybridization (FISH) of the X and Y chromosomes andchromosomes 12 and 18 in 10 donors under 25 years of agewho had a normal sperm count (20 × 10⁶/ml), 10 donorsover the age of 39 years with idiopathic infertility andnormozoospermia (20 × 10⁶/ml), and 5 oligozoospermicdonors (<20 × 10⁶/ml). Results: There was no obvious relationship betweenincreasing age and autosomal disomy (disomy 12 and disomy 18).Neither autosomal disomy nor diploidy was increased inany group. The frequency of X-, Y-, XX-, and YY-bearingsperm did not differ significantly among groups, but thefrequency of XY-bearing sperm was significantly higher inthe older infertile group than in the control donors. Conclusions: The incidence of nondisjunction of paternalsex chromosome in meiosis I was higher in older men withidiopathic infertility. The present results suggest that therisk of producing XXY fetuses is higher among men >39years of age with idiopathic infertility.     

Symmetric Replication of the Bacillus subtilis Chromosome

Synchronously replicating chromosomes in germinating spores of Bacillus subtilis were labeled near the origin with [(3)H]bromouracil. The label appeared in heavy-light DNA. When cell growth and chromosome replication were continued in unlabeled bromouracil, nearly all the tritium label was transferred to heavy-heavy DNA while the terminus remained unreplicated. This implies that both origins of the replicating chromosome can undergo reinitiation. Therefore, during multifork replication the chromosome takes on the symmetric "dichotomous" form rather than the asymmetric configuration predicted by the rolling circle model.
