Suppression of fibrous adhesion by proteoglycan decorin.   总被引:6,自引:0,他引:6  
Small proteoglycan decorin is known to suppress the bioactivity of TGF-beta through a competitive binding with the cell surface receptors for the cytokine. Based on this knowledge, we hypothesized that decorin could reduce the formation of fibrous adhesion, because our previous study showed the neutralizing antibody to TGF-beta1 has that effect. An intra-articular adhesion model in the rabbit knee joint was employed in this study, and decorin was administered into the joint cavity continuously during the 4 weeks of the experiment. The results of the dose-response study demonstrated that decorin suppresses formation of fibrous adhesion in a dose-dependent manner. When the administration of decorin was limited to shorter periods, this effect was considerably impaired and the necessity of long-term administration was demonstrated. On the other hand, when administered together with TGF-beta1, decorin still suppressed adhesion but to a lesser extent, and it was suggested that this proteoglycan could have other significant mechanism(s) to suppress adhesion besides the neutralization of TGF-beta. Thus, the present study showed that decorin could inhibit adhesion formation by both TGF-beta dependent and independent mechanisms. Considering that decorin exists ubiquitously in the body, its administration might be a promising approach to suppress adhesion.  相似文献

Our aim in this study was to evaluate the effect of geometry for measuring section thickness in tomosynthesis by using a metal bead device (bead method). Tomosynthesis images were obtained from two types of tomosynthesis equipment, Safire17 (ST, Shimadzu, Kyoto, Japan) and XR650 (GT, GE Healthcare, Milwaukee, WI). After tomosynthesis radiography with each device, the bead tomosynthesis images were obtained by image reconstruction. The digital profile was obtained from the digital value of the bead central coordinate in the perpendicular direction, and we acquired the slice sensitivity profile (SSP). The section thickness was defined with the full width at half maximum obtained from the SSP. We investigated the change in section thickness under different evaluation conditions: the angular range, the height of the bead position, the source–image receptor distance (SID), and image processing. The section thickness decreased when the angular range and height of the bead position increased. Also, the section thickness varied with a change in the SID. The section thickness differed according to the geometry for measuring the section thickness. Thus, the effect of the geometry used for measurement should be considered when the section thickness in tomosynthesis is measured by the bead method.  相似文献

OBJECTIVE: While diffusion-weighted magnetic resonance imaging (MRI) has been used to study malignant brain tumours, this modality has not been used to study MRI abnormalities surrounding meningiomas. METHODS: We examined intensity and apparent diffusion coefficient (ADC) on diffusion weighted imaging (DWI) for predicting postoperative persistence of MRI abnormalities surrounding meningiomas as well as characterizing the tumours. RESULTS: Of 36 meningiomas who underwent gross total resection, 27 (75%) showed hyperintensity on DWI at b=1100s/mm(2). No atypical meningiomas were hypointense on DWI. Of the 26 supratentorial meningiomas, 18 (69.0%) had associated MRI abnormality. No significant correlation was seen between tumour intensity on DWI and existence of surrounding MRI abnormality. Meningothelial meningiomas showed a relatively low prevalence of MRI abnormalities surrounding tumour (30%). Of 11 patients who underwent sequential MRI, all MRI abnormalities surrounding tumour showing isointensity and high ADC on preoperative DWI disappeared after surgery (from 3 weeks to 10 months). All MRI abnormalities surrounding tumour showing hyperintensity and low ADC on preoperative DWI persisted on final follow-up MRI (from 6 months to 20 months). CONCLUSION: The postoperative course of MRI abnormality surrounding tumour might be predictable from the intensity and ADC on preoperative DWI. Since MRI abnormalities associated with meningiomas can cause preoperative neurologic deficits. We hypothesise that abnormalities with restricted diffusion will be more likely to be associated with a preoperative deficit, and more likely to remain after removal of the causative meningioma.  相似文献

Vascular endothelial growth factor (VEGF) is known to be a mediator of angiogenesis and vascular permeability. A cystic component and haemorrhage are often found in pituitary adenomas. We assessed the VEGF expression based on immunohistochemical examinations in 48 pituitary adenomas. All the adenomas showed some VEGF immunoreactivity mainly in the cytoplasm of tumour cells. Of the 48 adenoma-cases, 16 cases had a strong VEGF immunoreactivity, 26 cases had a moderate one, and 6 cases had a weak one. On the MR images, a cystic component was found in 16 cases (33.3%), and a haemorrhage was found in 18 cases (37.5%). The VEGF immunoreactivity had a significant relationship with the cystic component but not the haemorrhage, size, recurrence, or HE classification. These findings suggest that VEGF plays any potential role in the pathogenesis of cystic formation in pituitary adenomas.  相似文献

[Purpose] This study aimed to determine whether muscle atrophy induced by ischemic reperfusion injury in rats can be prevented by the administration of antioxidants and exercise. [Subjects] Rats were randomly divided into five groups: non-treated, ischemic, exercise, ascorbic acid and exercise, and tocopherol and exercise. [Methods] The relative weight ratio of the soleus muscle and the length of the soleus muscle fiber cross-section minor axis were used for the evaluation of muscle atrophy. Pain was assessed as the weight-bearing ratio of the ischemic side. A multiple comparison test and the paired t-test were used for the statistical analyses. [Results] Compared with the non-treated group, the relative weight ratios of the soleus muscle and the lengths of the soleus muscle fiber cross-section minor axis significantly decreased in the other groups. Excluding the non-treated group, the relative weight ratios of the soleus muscle were heaviest in the tocopherol and exercise group. Excluding the non-treated group, the lengths of the soleus muscle fiber cross-section minor axis were longest in the tocopherol and exercise group, followed by the ischemic, exercise, and ascorbic acid and exercise groups. The amount of antioxidant substances did not decrease on the weight-bearing ratio of the ischemic side. [Conclusion] In this study, using an experimental rat model, we confirmed that antioxidants and exercise effect muscle atrophy induced by ischemic reperfusion. The results show that muscle regeneration was facilitated by phagocytosis in the tocopherol and exercise group.Key words: Ischemic reperfusion, Exercise, Antioxidant  相似文献

[Purpose] The purpose of the present study was to investigate if lactate thresholds in rats could be accurately determined during downhill running in rats. [Subjects] Twenty-four female Wistar rats were used in this study. Rats were randomly divided into two groups: the level running group and downhill running group. [Methods] Catheters were inserted into the right external jugular veins of the rats. Two days after the operation, the rats performed incremental exercise tests. The level running group ran on a treadmill at a 0° incline; the downhill running group ran on a treadmill at a −16° incline. The lactate threshold was determined using the 2-line combination that minimizes the total sum of squares for fitting all data. [Results] Maximal running speeds and running speeds at the lactate threshold for the downhill running group were significantly higher than those for the level running group. [Conclusion] In this study, it was possible to accurately determine the lactate threshold during downhill running. These findings suggest that when the treadmill speed is the same, downhill running can be done at lower metabolic intensity than level running.Key words: Lactate threshold, Downhill running, Eccentric contraction  相似文献

A perivascular epithelioid tumor (PEComa) is a rare tumor probably arising from the perivascular epithelioid cells. Only three cases of pancreatic PEComa have been reported in the English-language literature. The present report describes an extremely rare case of pancreatic PEComa. A 47-year-old Japanese woman complained of lower abdominal pain and a well-demarcated solid tumor was found in the pancreatic head. There was no history of tuberous sclerosis complexes. Pylorus-preserving pancreaticoduodenectomy was thus performed. There was a well-demarcated, solid tumor measuring 17 mm in the pancreatic head. The tumor was composed of a diffuse proliferation of epithelioid tumor cells with many blood vessels but no adipose tissue. The tumor cells expressed HMB45 and α-smooth muscle actin. Ultrastructurally, the tumor cells possessed many membrane-bound granules that were positive for HMB45 on immunoelectron microscopy. The results of immunoelectron microscopy show that some PEComas possess not only typical melanosomes or premelanosomes but also aberrant melanosomes.  相似文献

In this paper, we discuss subspace-based support vector machines (SS–SVMs), in which an input vector is classified into the class with the maximum similarity. Namely, for each class we define the weighted similarity measure using the vectors called dictionaries that represent the class, and optimize the weights so that the margin between classes is maximized. Because the similarity measure is defined for each class, for a data sample the similarity measure to which the data sample belongs needs to be the largest among all the similarity measures. Introducing slack variables, we define these constraints either by equality constraints or inequality constraints. As a result we obtain subspace-based least squares SVMs (SSLS–SVMs) and subspace-based linear programming SVMs (SSLP–SVMs). To speedup training of SSLS–SVMs, which are similar to LS–SVMs by all-at-once formulation, we also propose SSLS–SVMs by one-against-all formulation, which optimize each similarity measure separately. Using two-class problems, we clarify the difference of SSLS–SVMs and SSLP–SVMs and evaluate the effectiveness of the proposed methods over the conventional methods with equal weights and with weights equal to eigenvalues.  相似文献
