The dynamics of echo-morphometric parameters of crural skin was studied in patients with congenital shortening of lower extremity in the process of its operative elongation. It is demonstrated that dosed stretch results in increased skin thickness. After the completion of distraction, the skin thickness exceed preoperation levels by 48.7% (in patients aged 5-8 years), 20.9% (in patients aged 10-14 years) and 26.7% (in adults). Skin acoustic density after distraction termination reached the preoperation level in children and adolescents, while it was increased in adults.  相似文献

Monoclonal antibody (mAb) 38C2 belongs to a group of catalytic antibodies that were generated by reactive immunization and contains a reactive lysine. 38C2 catalyzes aldol and retro-aldol reactions, using an enamine mechanism, and mechanistically mimics natural aldolase enzymes. In addition, mAb 38C2 can be redirected to target integrins alpha(v)beta(3) and alpha(v)beta(5) through the formation of a covalent bond between a beta-diketone derivative of an arginine-glycine-aspartic acid (RGD) peptidomimetic and the reactive lysine residue in the antibody combining site to provide the chemically programmed mAb cp38C2. In this study, we investigated the potential of enhancing the activity of receptor-binding small molecule drug (SCS-873) through antibody conjugation. Using a M21 human melanoma xenograft model in nude mice, cp38C2 inhibited the growth of the tumor by 81%. The chemically programmed antibody was shown to be highly active at a low concentration while SCS-873 alone was ineffective even at dosages 1,000-fold higher than those used for the chemically programmed antibody. In vitro programming of the catalytic antibody was shown to be as effective as in vivo programming. In an experimental metastasis assay, treatment with mAb cp38C2 significantly prolonged overall survival of tumor-bearing severe combined immuno-deficient (SCID) mice when compared to treatment with unprogrammed mAb 38C2, SCS-873 alone or the integrin-specific monoclonal antibody LM609. In vitro, cp38C2 inhibited human and mouse endothelial and human melanoma cell adhesion, migration and invasion. Additionally, cp38C2 inhibited human and mouse endothelial cell proliferation and was active in complement-dependent cytotoxicity assays. These studies establish the potential of chemically programmed monoclonal antibodies as a novel and effective class of immunotherapeutics that combine the merits of traditional small molecule drug design with immunotherapy.  相似文献

The results of long-term studies made by the Department of General Chemistry, I. M. Sechenov Moscow Medical Academy, that deals with unification of methods for determining some ions of metals (lead, zinc, sodium, and potassium) in the assessment of the quality of drinking water and transfusion fluids are summarized. A procedure was developed to determine the trace impurities of zinc, lead, and silver by atomic absorption spectrometry (AAS) by using sorption concentration. C-80-2-aminothiazole, a new sorbent synthesized at the Research Institute of Polymers, was used to detect these ions in the drinking water. With regards to the chosen optimal conditions, drinking water samples were analyzed via their direct spraying in the air-acetylene flame. The prior sorption concentration determined drinking water zinc and lead ions in the concentrations equal to or less than their permissible dose concentrations. The studies indicated that the used methods to determine the trace quantities of metals in the drinking water and aqueous solutions show a high sensitivity, rapidity, simplicity of sample preparation.  相似文献

We studied morphological characteristics of the regenerative process in gingival tissues during therapy of chronic periodontitis. Energostim stimulated macrophageal reaction and promoted neoangiogenesis in the inflammatory infiltrate. It was not observed after traditional drug therapy. Energostim promoted vascularization in regenerating tissues, normalized the structure at a greater area of the lamina propria of the gingiva, and prevented fibrous and sclerotic changes. The volume of histiolymphocytic infiltrates in regenerating gingival tissues decreased after application of intradental splints.  相似文献

We studied the effect of local anesthetic richlocaine on proliferation and intracellular calcium content in cultured osteoblasts from rat parietal bone. In a concentration of 1 mg/ml this drug produced a cytotoxic effect on osteoblasts. In concentrations of 0.01 and 0.001 mg/ml richlocaine in the absence and presence of subtoxic dose of sodium cyanide (0.2 mM) increased the number of osteoblasts by 15.4 and 36.6 or 13.8 and 38.6%, respectively. In a concentration of 1 mg/ml, richlocaine increased the content of cytosolic calcium in osteoblasts by 105%. These effects of richlocaine in low concentrations (0.01 and 0.001 mg/ml) can be related to stimulation of metabolic processes in osteoblasts.  相似文献

Treatment with richlocaine alone and, especially, in combination with antihypoxant energostim decreased the total content of hydroxyproline in the ischemic skin flap on day 3 after excision. Combination therapy with richlocaine and energostim normalized the redox potential in the energy supply system, improved antioxidant protection, and promoted the recovery of a balance between various components in the antioxidant system. These changes were not accompanied enhanced production of malonic dialdehyde. Our results suggest that combination therapy with richlocaine and energostim maintains the adaptive reserves of detoxifying systems in keratinocytes and prevents endotoxemia. Richlocaine primarily stimulates glycolytic synthesis of ATP, activates nonmitochondrial antioxidant enzymes, and increases RNase activity in lysosomes.  相似文献
