Background: The prevalence of liver disorders and metabolic syndrome has increased among youth. Glyphosate, the most widely used herbicide worldwide, could contribute to the development of these conditions.Objective: We aimed to assess whether lifetime exposure to glyphosate and its degradation product, aminomethylphosphonic acid (AMPA), is associated with elevated liver transaminases and metabolic syndrome among young adults.Methods: We conducted a prospective cohort study (n=480 mother–child dyads) and a nested case–control study (n=60 cases with elevated liver transaminases and 91 controls) using data from the Center for the Health Assessment of Mothers and Children of Salinas (CHAMACOS). We measured glyphosate and AMPA concentrations in urine samples collected during pregnancy and at child ages 5, 14, and 18 y from cases and controls. We calculated glyphosate residue concentrations: [glyphosate +(1.5×AMPA)]. We estimated the amount of agricultural-use glyphosate applied within a 1-km radius of every residence from pregnancy to age 5 y for the full cohort using California Pesticide Use Reporting data. We assessed liver transaminases and metabolic syndrome at 18 y of age.Results: Urinary AMPA at age 5 y was associated with elevated transaminases [relative risk (RR) per 2-fold increase=1.27, 95% confidence interval (CI): 1.06, 1.53] and metabolic syndrome (RR=2.07, 95% CI: 1.38, 3.11). Urinary AMPA and glyphosate residues at age 14 y were associated with metabolic syndrome [RR=1.80 (95% CI: 1.10, 2.93) and RR=1.88 (95% CI: 1.03, 3.42), respectively]. Overall, a 2-fold increase in urinary AMPA during childhood was associated with a 14% and a 55% increased risk of elevated liver transaminases and metabolic syndrome, respectively. Living near agricultural glyphosate applications during early childhood (birth to 5 y of age) was also associated with metabolic syndrome at age 18 y in the case–control group (RR=1.53, 95% CI: 1.16, 2.02).Discussion: Childhood exposure to glyphosate and AMPA may increase risk of liver and cardiometabolic disorders in early adulthood, which could lead to more serious diseases later in life. https://doi.org/10.1289/EHP11721  相似文献

The treatment of patients with early stage breast cancer has changed in recent years due to the introduction of pembrolizumab, olaparib, and abemaciclib. These and other drugs with the same class of active ingredient are currently in trial for various indications. This review article summarizes the latest results that have either been presented at major conferences such as the ESMO 2022 or published recently in international journals. This includes reports on newly discovered breast cancer genes, atezolizumab in neoadjuvant therapy in HER2-positive patients, long-term data from the APHINITY study, and on how preoperative peritumoral application of local anesthetics can influence the prognosis. 为探讨下丘脑室旁核对哮喘大鼠的神经调控途径,取健康雄性SD大鼠制备哮喘模型并诱发哮喘发作,运用WGA-HRP逆行追踪法与免疫组织化学染色法(ABC法)相结合的双重标记方法,在光镜下观察向迷走神经复合体(DVC)发出投射的下丘脑室旁核(PVN)神经元内Fos蛋白的表达情况。结果显示:PVN内有三种阳性细胞,即HRP逆行标记神经元、Fos样免疫阳性神经元和HRP/Fos双标神经元。这些神经元主要见于PVN小细胞部的内侧亚核和外侧亚核,散在分布于背侧亚核,在前小细胞亚核内未见阳性反应;Fos样免疫阳性细胞呈双侧分布,且HRP逆标神经元,HRP/Fos双标神经元也为双侧分布,但以注射区同侧占优势。HRP/Fos双标神经元占HRP标记细胞的44.22%。本研究结果提示,哮喘大鼠发作时中枢内包括下丘脑PVN、延髓DVC的多个脑区内神经元兴奋,且两者之间通过直接投射联系参与哮喘的调控。  相似文献

Background  To determine the accuracy of contrast-enhanced multislice computed tomography (CT) in the assessment of treatment success immediately after CT-guided radiofrequency (RF) ablation. Methods  26 patients with 38 Colorectal liver metastasis (CRM) were treated by CT-guided RF ablation. Pre-contrast and portal phase CT features before and immediately after ablation were retrospectively evaluated quantitatively and qualitatively: Influence of attenuation characteristics, safety margin, congruency between tumor and coagulation, and morphological criteria (shape, margin distinction, margin configuration, and margin continuity) were investigated. Findings were statistically analyzed with regard to local tumor progression. Results  Mean observation period for follow-up scans was 6.4 months (range: 3–40 months). Attenuation characteristics, safety margin, and congruency had no significant effect on the probability of local tumor progression. Coagulations whose margin was categorized as “discontinuous” were significantly more often associated with local recurrence (p = 0.038). No significant influence on local recurrence could be detected regarding coagulation shape, margin distinction, and configuration. Conclusion  Computed tomography imaging immediately after RF ablation allows for morphological characterization of the coagulation and provides a valid baseline status for follow-up imaging. However, in CRM, morphological image criteria and attenuation characteristics have limited predictive value for immediate detection of persistent tumor.  相似文献

The Kunjin strain of West Nile virus (WNVKUN) is a mosquito-transmitted flavivirus that can infect farmed saltwater crocodiles in Australia and cause skin lesions that devalue the hides of harvested animals. We implemented a surveillance system using honey-baited nucleic acid preservation cards to monitor WNVKUN and another endemic flavivirus pathogen, Murray Valley encephalitis virus (MVEV), on crocodile farms in northern Australia. The traps were set between February 2018 and July 2020 on three crocodile farms in Darwin (Northern Territory) and one in Cairns (North Queensland) at fortnightly intervals with reduced trapping during the winter months. WNVKUN RNA was detected on all three crocodile farms near Darwin, predominantly between March and May of each year. Two of the NT crocodile farms also yielded the detection of MVE viral RNA sporadically spread between April and November in 2018 and 2020. In contrast, no viral RNA was detected on crocodile farms in Cairns during the entire trapping period. The detection of WNVKUN and MVEV transmission by FTATM cards on farms in the Northern Territory generally correlated with the detection of their transmission to sentinel chicken flocks in nearby localities around Darwin as part of a separate public health surveillance program. While no isolates of WNVKUN or MVEV were obtained from mosquitoes collected on Darwin crocodile farms immediately following the FTATM card detections, we did isolate another flavivirus, Kokobera virus (KOKV), from Culex annulirostris mosquitoes. Our studies support the use of the FTATM card system as a sensitive and accurate method to monitor the transmission of WNVKUN and other arboviruses on crocodile farms to enable the timely implementation of mosquito control measures. Our detection of MVEV transmission and isolation of KOKV from mosquitoes also warrants further investigation of their potential role in causing diseases in crocodiles and highlights a “One Health” issue concerning arbovirus transmission to crocodile farm workers. In this context, the introduction of FTATM cards onto crocodile farms appears to provide an additional surveillance tool to detect arbovirus transmission in the Darwin region, allowing for a more timely intervention of vector control by relevant authorities.  相似文献
