We sequenced a 705-bp fragment of the recA gene from 113 Vibrio cholerae strains and closely related species. One hundred eighty-seven nucleotides were phylogenetically informative, 55 were phylogenetically uninformative, and 463 were invariant. Not unexpectedly, Vibrio parahaemolyticus and Vibrio vulnificus strains formed out-groups; we also identified isolates which resembled V. cholerae biochemically but which did not cluster with V. cholerae. In many instances, V. cholerae serogroup designations did not correlate with phylogeny, as reflected by recA sequence divergence. This observation is consistent with the idea that there is horizontal transfer of O-antigen biosynthesis genes among V. cholerae strains. 相似文献

Extrahepatic bile duct (EBD) carcinoma is an aggressive cancer with a poor prognosis. Fascin is an actin-bundling protein with roles in forming cell protrusions and mesenchymal and neuronal cell motility. Many human neoplasms up-regulate fascin. High fascin expression is thought to be a poor prognostic factor in some cancers; however, few data are available for the role of fascin in EBD carcinoma. We investigated fascin immunoreactivity in EBD carcinoma and tested it for correlations between fascin expression and clinicopathological parameters.Conventional tissue sections of 114 cases of EBD carcinomas were immunohistochemically analyzed for fascin expression.Fascin expression was tested by cytoplasmic staining. Negative, weak positive, and strong positive fascin staining was observed in 49 (43.0%), 32 (28.1%), and 33 cases (28.9%), respectively. Fascin expression in EBD carcinomas was significantly correlated with histological grade (P<0.0001), primary tumor (T) (P=0.002), TNM stage (P=0.036), lymphatic invasion (P=0.048), venous invasion (P=0.024), and adjacent organ invasion (P<0.0001). High fascin expression was a significant poor prognostic factor (P=0.0001) in EBD carcinoma. High fascin expression (P=0.004) and TNM stage (P=0.001) in EBD carcinoma were independent adverse prognostic factors.High fascin expression is significantly correlated with aggressive tumor phenotype in EBD carcinoma and is an independent poor prognostic factor in EBD carcinoma. 相似文献

In adult male rats, 3beta-hydroxysteroid dehydrogenase/delta5-delta4-isomerase (3beta-HSD) expressing cells were identified in the spinal cord from the cervical to the sacral segments. An in situ hybridization study, using an oligonucleotide common to the four known isoforms of rat 3beta-HSD, revealed its mRNA in gray matter. Measurements of optical densities in autoradiograms showed the following regional distribution: dorsal horn (layers I-III) > central canal (layer X) > or = ventral horn (layers VIII-IX) > ventral funiculus = lateral funiculus. At the cellular level, the number of grains was higher on the large motoneurons than on small neurons of the dorsal horn, but the grain density per cell was similar. Further evidence for the expression of 3beta-HSD in the spinal cord was obtained by western blot analysis, which revealed an immunoreactive protein of approximately 45 kDa in the dorsal and ventral parts of the spinal cord. Castration and adrenalectomy did not influence the expression of 3beta-HSD mRNA and protein. Gas chromatography/mass spectrometry measurements showed higher levels of pregnenolone and progesterone in the spinal cord than in the plasma. After castration and adrenalectomy, their levels remained elevated in the spinal cord, suggesting that these neurosteroids may be synthesized locally. The wide distribution of 3beta-HSD, and the high levels of pregnenolone and progesterone in the spinal cord even after castration and adrenalectomy, strongly suggest a potential endogenous production of progesterone and an important signalling function of this steroid in the spinal cord. 相似文献

采用静电纺丝技术制备胶原/壳聚糖复合纳米纤维膜,研究其作为引导骨再生生物膜的细胞生物相容性及诱导成骨性。以乙酸为溶剂,聚环氧乙烯(PEO)为增塑剂,采用静电纺丝技术制备胶原纳米纤维膜及不同比例的胶原/壳聚糖复合纳米纤维膜(胶原、壳聚糖、PEO质量比5∶1∶4,5∶2∶3,5∶4∶1),电子显微镜观察4种纳米纤维膜的表面形态;将骨髓间充质干细胞种植于胶原纳米纤维膜及表面形态较好的胶原/壳聚糖纳米纤维膜上,通过MTT法、碱性磷酸酶检测、细胞内胶原检测、免疫荧光染色及茜素红染色法观察,研究其细胞生物相容性及诱导成骨性。扫描电子显微镜观察胶原纳米纤维膜及质量比为5∶1∶4的胶原/壳聚糖复合纳米纤维膜的纤维光滑,直径均一。MTT法检测显示,胶原纳米纤维膜和胶原/壳聚糖复合纳米纤维膜均可促进骨髓间充质干细胞的粘附和增殖。细胞培养14 d后,胶原/壳聚糖复合纳米纤维膜上细胞内胶原含量检测为2.02 mg/gport,高于胶原纳米纤维膜组的1.63 mg/gport胶原含量(P<0.05),且胶原/壳聚糖复合纳米纤维膜上细胞内碱性磷酸酶、骨钙素及钙化结节的形成均高于胶原纳米纤维膜组。胶原/壳聚糖复合纳米纤维膜可促进骨髓间充质干细胞的增殖和分化,有望应用于骨再生的研究。 相似文献

The hypoxic environment imposes severe selective pressure on species living at high altitude. To understand the genetic bases of adaptation to high altitude in dogs, we performed whole-genome sequencing of 60 dogs including five breeds living at continuous altitudes along the Tibetan Plateau from 800 to 5100 m as well as one European breed. More than 150× sequencing coverage for each breed provides us with a comprehensive assessment of the genetic polymorphisms of the dogs, including Tibetan Mastiffs. Comparison of the breeds from different altitudes reveals strong signals of population differentiation at the locus of hypoxia-related genes including endothelial Per-Arnt-Sim (PAS) domain protein 1 (EPAS1) and beta hemoglobin cluster. Notably, four novel nonsynonymous mutations specific to high-altitude dogs are identified at EPAS1, one of which occurred at a quite conserved site in the PAS domain. The association testing between EPAS1 genotypes and blood-related phenotypes on additional high-altitude dogs reveals that the homozygous mutation is associated with decreased blood flow resistance, which may help to improve hemorheologic fitness. Interestingly, EPAS1 was also identified as a selective target in Tibetan highlanders, though no amino acid changes were found. Thus, our results not only indicate parallel evolution of humans and dogs in adaptation to high-altitude hypoxia, but also provide a new opportunity to study the role of EPAS1 in the adaptive processes.The mechanisms of organismal adaptation to high-altitude hypoxia are of great interest during recent years. Highland wild animals have a long life history at high altitude, and the whole genomes of yak (descendants of wild yak) (Qiu et al. 2012), Tibetan antelope (Ge et al. 2013), snow leopard (Cho et al. 2013), and wild boar (Li et al. 2013) have been sequenced. In contrast, the human settlement history on highland is rather short, which dates from ∼25,000 yr ago (Zhao et al. 2009). Whole-genome genotyping and re-sequencing have been performed for three typical highland populations including Tibetans (Beall et al. 2010; Bigham et al. 2010; Simonson et al. 2010; Yi et al. 2010; Peng et al. 2011; Xu et al. 2011), Andeans (Bigham et al. 2009, 2010), and Ethiopians (Alkorta-Aranburu et al. 2012; Scheinfeldt et al. 2012).The increased oxygen uptake and delivery are physiological hallmarks of high-altitude adaptation. On one hand, the capacity of oxygen uptake is determined by hemoglobin concentration and oxygen affinity. For example, the Andean highlanders display a high level of hemoglobin concentration (Beall et al. 2002; Beall 2007). The high oxygen affinity of hemoglobin is found in many highland animals such as yak (Weber et al. 1988), alpaca (Piccinini et al. 1990), deer mice (Storz et al. 2007; Storz et al. 2009), bar-headed goose (Zhang et al. 1996; Liang et al. 2001), and Andean goose (Jessen et al. 1991). On the other hand, the rate of oxygen delivery is determined by blood flow. For example, although Tibetans maintain a nearly normal level of hemoglobin concentration and a low level of oxygen saturation, they display a high level of blood flow, resulting in the increase of oxygen delivery (Beall et al. 2001; Erzurum et al. 2007).Whole-genome scans revealed that positive selection for human high-altitude adaptation occurred in the hypoxia-inducible factor (HIF) pathway (Bigham et al. 2009; Beall et al. 2010; Bigham et al. 2010; Simonson et al. 2010; Yi et al. 2010; Peng et al. 2011; Xu et al. 2011; Alkorta-Aranburu et al. 2012; Scheinfeldt et al. 2012), which regulates genes associated with blood physiology. In addition, metabolic pathways may also be involved in the adaptive process of yak (Qiu et al. 2012) and Tibetan antelope (Ge et al. 2013).Although a lot of studies focused on wildlife and human highlanders, no research was performed on domesticated animals that migrated to the plateau with humans, which represent an adaptation pattern on a short evolutionary time scale of thousands of years. For example, the Tibetan Mastiff is a native dog living in the Tibetan Plateau with an altitude of 3000–6000 m. It is also an ancient dog in the world (Li and Zhang 2012). However, the genetic and physiological mechanisms of its adaptation to high-altitude environments remain elusive.In this study, we sampled five dog breeds including the Tibetan Mastiff from continuous altitudes along the Ancient Tea Horse Road in southwestern China as well as one European breed. We performed whole-genome sequencing for the dogs and identified candidate genes for high-altitude adaptation using selective sweep mapping. 背景:前期研究成功制备了纳米羟基磷灰石-聚羟基丁酸戊酯/聚乙二醇-5-氟尿嘧啶材料。
目的:观察纳米羟基磷灰石-聚羟基丁酸戊酯/聚乙二醇-5-氟尿嘧啶的填充骨缺损和抗肿瘤作用。
方法:取72只新西兰兔建立右膝关节VX2 骨肿瘤模型,切除部分胫骨形成骨缺损,且使骨缺损与肿瘤相邻,造模后随机分为实验组与对照组,分别于骨缺损处植入纳米羟基磷灰石-聚羟基丁酸戊酯/聚乙二醇-5-氟尿嘧啶与纳米羟基磷灰石-聚羟基丁酸戊酯/聚乙二醇人工骨。
结果与结论:①无机质含量:两组均随时间的延长逐渐降低,尤以实验组明显。②X射线检查:实验组术后24周材料体积较植入初期变小,肿瘤开始变小。对照植入材料体积始终无明显变化,肿瘤体积进行性增大。③生物力学检测:术后24周,实验组下肢标本最大扭转强度明显高于对照组(P < 0.01)。④骨密度检测:术后2-24周,两组均呈先减低后增加趋势,以实验组明显。⑤肿瘤体积变化:术后24周,实验组减小,对照组增加。表明纳米羟基磷灰石-聚羟基丁酸戊酯/聚乙二醇-5-氟尿嘧啶复合材料能很好填充骨缺损并有明显抑制肿瘤生长的作用。 相似文献
