目的:探讨复方中药金水清合剂对IgA肾病大鼠TGF-β1、CTGF、VEGF蛋白及mRNA表达的影响。方法:4周龄SPF级SD大鼠40只,随机分为正常组、模型组、治疗1组、治疗2组、治疗3组,复制大鼠IgA肾病模型,观察组在模型组的基础上,分别于第5周、第7周、第9周,给予金水清合剂灌胃治疗,1次/d,至第12周末,HE及PAS染色观察肾脏病理学改变,Western-blot检测肾组织TGF-β1、CTGF、VEGF蛋白的表达,RT-PCR检测TGF-β1、VEGF及CTGF mRNA的表达。结果:正常组大鼠肾组织结构完整清晰,模型组大鼠可见肾小球系膜区增宽,系膜基质、系膜细胞中到重度增生,观察组病变程度比模型组轻,治疗1组改善最为明显。与正常组比较,模型组TGF-β1、CTGF、VEGF蛋白的表达升高,且差异有统计学意义(P0.05);与模型组比较,观察组表达降低,且差异有统计学意义(P0.05);观察组间比较差异有统计学意义(P0.05);与正常组比较,模型组TGF-β1、CTGF、VEGF mRNA的表达升高,且差异有统计学意义(P0.05);与模型组比较,观察组表达降低,且差异有统计学意义(P0.05);观察组间比较,差异有统计学意义(P0.05)。结论:金水清合剂能显著改善IgA肾病大鼠肾脏损害,早期治疗改善最为明显,其作用机制可能为下调TGF-β1、CTGF、VEGF蛋白及mRNA的表达。  相似文献

Objective: To study the antipyretic and anti-inflammatory constituents from the active fraction of Reduning(RDN) Injection.Methods: In this study, the active fraction of RDN Injection was screened by the LPS-induced mouse endotoxin shock model. The chemical constituents were isolated by chromatography on HP-20 macroporous adsorptive resins, silica gel, ODS columns and reverse phase MPLC and HPLC repeatedly, and their structures were elucidated based on spectroscopic analysis(HR-ESI-MS, NMR, ECD) and chemical methods.Meanwhile, we evaluated the anti-inflammatory activities of the isolates by measuring their inhibitory effects on TNF-α production in LPS stimulated RAW 264.7 macrophages.Results: The 95% ethanol eluate of RDN Injection by the macroporous adsorption resin column was proved to be the antipyretic and anti-inflammatory active fraction of this injection. A novel iridoid, named jasminoide A(1), and a new guaiane sesquiterpenoid, named(1 R,7 R,8 S,10 R)-7,8,11-trihydroxy-4-guaien-3-one(2), were isolated from Reduning injection, and compound 2 can inhibit TNF-α production with IC50 values of 72.24 μmol/L.Conclusion: In this study, two new terpenoids were isolated from Reduning Injection, and compound 2 showed inhibitory activity against LPS-activated TNF-α production in RAW 264.7 cells in vitro.  相似文献

Background:Plantar fasciitis (PF) is the most common cause of heel pain in adult. There are a variety of ways to treat PF, but these treatments have varied result in their effectiveness, and exist different degrees of limitations. At present, clinical studies focus on the effect of glucocorticoid (GC) and platelet rich plasma (PRP) in the treatment of PF, but there is a lack of systematic evaluation PRP and GC''s clinical effect towards PF. This study aims to evaluate the long-term efficacy of GCs and PRP in the treatment of PF by means of meta-analysis.Methods:The literature of a randomized controlled clinical trial of PRP in the treatment of plantantifasciitis was searched on the Internet. Retrieve 7 databases. EndNote X9 software was used for document management. The Jadad scale was used to score the literature. Risk assessment of the literature was conducted according to Cochrane''s systematic evaluation manual 5.0. RevMan5.3 software was used for literature risk bias analysis. Stata12.0 software is used for sensitivity analysis.Results:This study will provide effective evidence-based evidence for the long-term efficacy of PRP and GC in treating PF.Conclusion:A systematic review and meta-analysis were conducted for the comparison of the long-term effect of PRP and GC on plantar fascia in the treatment of PF.  相似文献

Jingyin granules, a marketed antiviral herbal medicine, have been recommended for treating H1N1 influenza A virus infection and Coronavirus disease 2019 (COVID-19) in China. To fight viral diseases in a more efficient way, Jingyin granules are frequently co-administered in clinical settings with a variety of therapeutic agents, including antiviral drugs, anti-inflammatory drugs, and other Western medicines. However, it is unclear whether Jingyin granules modulate the pharmacokinetics of Western drugs or trigger clinically significant herb–drug interactions. This study aims to assess the inhibitory potency of the herbal extract of Jingyin granules (HEJG) against human drug-metabolizing enzymes and to clarify whether HEJG can modulate the pharmacokinetic profiles of Western drug(s) in vivo. The results clearly demonstrated that HEJG dose-dependently inhibited human CES1A, CES2A, CYPs1A, 2A6, 2C8, 2C9, 2D6, and 2E1; this herbal medicine also time- and NADPH-dependently inhibited human CYP2C19 and CYP3A. In vivo tests showed that HEJG significantly increased the plasma exposure of lopinavir (a CYP3A-substrate drug) by 2.43-fold and strongly prolonged its half-life by 1.91-fold when HEJG (3 g/kg) was co-administered with lopinavir to rats. Further investigation revealed licochalcone A, licochalcone B, licochalcone C and echinatin in Radix Glycyrrhizae, as well as quercetin and kaempferol in Folium Llicis Purpureae, to be time-dependent CYP3A inhibitors. Collectively, our findings reveal that HEJG modulates the pharmacokinetics of CYP substrate-drug(s) by inactivating CYP3A, providing key information for both clinicians and patients to use herb–drug combinations for antiviral therapy in a scientific and reasonable way.  相似文献

Specific and accurate detection of single nucleotide variants (SNVs) plays significant roles in pathogenic gene research and clinical applications. However, the sensitive but ultra-specific detection of rare variants in biological samples still remains challenging. Herein, we report a novel, robust and practical SNV assay by integrating the outstanding features of high selectivity of an artificial mismatched probe, and the powerful loop-mediated isothermal amplification. In this strategy, we rationally introduce artificial mismatched bases into the 3′-terminal regions of the probe located in the ligation region to reduce the risk of nonspecific ligation, which can dramatically improve the specificity for the SNV assay. The proposed method can discern as little as 0.01% mutant DNA in the high background of wild-type DNA with high sensitivity (10 aM). In virtue of its outstanding performance, the artificial mismatched probe may also be employed and expanded in various DNA and RNA genetic analyses with ligase-assisted approaches, showing great potential in biomedical research, clinical diagnostics, and bioanalysis.

An artificial mismatched base introduced in a ligation probe can effectively reduce nonspecific ligation and improve the specificity for SNV assay.  相似文献

Flexible electric-double-layer (EDL) thin film transistors (TFTs) based on a vertical InGaZnO4 (IGZO) channel are fabricated at room temperature. Such TFTs show a low operation voltage of 1.0 V due to the large specific gate capacitance of 3.8 μF cm−2 related to electric-double-layer formation. The threshold voltage, drain current on/off ratio and subthreshold swing are estimated to be −0.1 V, 1.2 × 106 and 80 mV per decade, respectively. The combination of low voltage, high current on-to-off ratio and room temperature processing make the flexible vertical-IGZO-channel TFTs very promising for low-power portable flexible electronics applications.

In this study, we report a simple method to improve the thermal conductivity of epoxy resin by using new magnetic composites as fillers. The rGO@Fe3O4 nanocomposite has been prepared by a solvothermal method, and its morphology and chemical structure were characterized and analyzed by various characterization methods. Afterwards, the rGO@Fe3O4/EP composite material was obtained in an external magnetic field, in which the rGO@Fe3O4 is uniformly dispersed in the epoxy resin matrix, arranged along the direction of the magnetic field. In addition, the orientation of rGO@Fe3O4 increases with the magnetic field intensity. After doping 30% (wt) rGO@Fe3O4 into epoxy resin and curing under a 500 Gs magnetic field, the rGO@Fe3O4/EP composite material is anisotropic and has a higher thermal conductivity (increased by 196.60%) parallel to the direction of the magnetic field compared to a pure ring oxygen resin.

A rGO@Fe3O4 nanocomposite was prepared by a solvothermal method. Then the rGO@Fe3O4/epoxy resin composite was cured in a magnetic field. This shows that rGO@Fe3O4 can enhance the heat-conductivity of epoxy resins.  相似文献
