Background and aims  

It has been reported that a dorsal cloacal plate defect is associated with anorectal malformations (ARMs); however, there has been very little information reported about the developmental mechanisms involved with cloacal plate formation. Danforth’s short tail (Sd) mutant mice show ARMs. In our previous study, the co-presence of Skt Gt mutation, in which Skt gene is disrupted by the gene-trap vector (p-U8), increased the incidence of ARMs in Sd mutant to 100%. Our aims in this study are determining the Skt expression around the cloaca during the anorectal development and demonstrating the role of Skt gene in ARMs.  相似文献

Background: The cardiac progenitor cells for the outflow tract (OFT) reside in the visceral mesoderm and mesodermal core of the pharyngeal region, which are defined as the secondary and anterior heart fields (SHF and AHF), respectively. Results: Using chick embryos, we injected fluorescent‐dye into the SHF or AHF at stage 14, and the destinations of the labeled cells were examined at stage 31. Labeled cells from the right SHF were found in the myocardium on the left dorsal side of the OFT, and cells from the left SHF were detected on the right ventral side of the OFT. Labeled cells from the right and left AHF migrated to regions of the ventral wall of the OFT close to the aortic and pulmonary valves, respectively. Conclusion: These observations indicate that myocardial progenitors from the SHF and AHF contribute to distinct conotruncal regions and that cells from the SHF migrate rotationally while cells from the AHF migrate in a non‐rotational manner. The purpose of this study was to evaluate whether a Gallium Nitride (GaN) -based violet laser diode (VLM500) could be used as a light source for light-cured dental resins. Three experimental unfilled resins containing different photoinitiators (camphorquinone, CQ; phenyl propanedione, PPD; or mono acylphosphineoxide, MAPO) were evaluated. These resins were light-cured with a VLM500 laser diode, and their ultimate micro-tensile strengths (μTS) were compared to those cured with three different LED light sources (Curenos, G-Light Prima-normal mode and G-Light Prima-PL mode). The VLM500 produced high μTS values in all three resins, and we concluded that this violet laser diode can be used as a light source for light-cured dental resin materials.  相似文献

Many patients visit oral malodor clinics because of malodors which are brought to their attention by friends and family, or because they note the behavior of people around them, they suspect a problem and develop a fear of having an oral malodor. However, only around 30% of such patients actually have levels of malodor high enough to bother other people. Many patients exhibit halitophobia symptoms, which present as self-perception of malodor, and thus have a strong obsession about their smell which results in distress. Here, we carried out a study on 300 outpatients who visited the Tokyo Dental College Chiba Hospital Odor Clinic. We used the Tokyo University Egogram (TEG) to elucidate character traits of affected outpatients and compared the occurrence of TEG types in these patients with those of normal individuals. We discovered that 10.4% of patients were A-dominant type, which was 10.6% lower than the 21.0% of normal individuals. On the other hand, 18.4% of patients were N-type (NP high, FC low), which was 9.9% higher than the 8.5% of normal individuals. Results revealed that very few of the malodor outpatients exhibited the trait that shows intelligence, calm judgment, and self-affirmation, and as a result enjoy their life. Instead, many of these patients tended to show high levels of kindness and appeared to be holding themselves back and exercising patience.  相似文献

Human polymeric immunoglobulin receptor (pIgR) was expressed in baby hamster kidney (BHK) cells using a recombinant vaccinia virus transfection system. Cleavage of pIgR on the cell surface was partially inhibited by the proteinase inhibitor, leupeptin. We addressed the question whether some particular regions of pIgR could affect the efficient cleavage of this molecule, with the following results: (1) a mutant lacking the entire cytoplasmic region resulted in release of secretory component (SC) into the culture supernatant much faster than wild-type; (2) a pIgR mutant lacking the entire extracellular domain 6, the region containing the susceptible cleavage sites, could be cleaved and released as a mutant SC. The transport kinetics of this mutant between endoplasmic reticulum (ER) and Golgi or Golgi and the cell surface was equivalent to wild-type pIgR. Our results indicate that although the main cleavage site is in domain 6, at least one other cleavage site may exist.  相似文献

The purpose of this study was to evaluate whether violet-laser diode (VLD) can be used as light-curing source. The ultimate (micro-)tensile strength (μTS) of three adhesives was determined when cured by VLD in comparison with curing by two different types of commercial LED light-curing units. One VLD (VLM 500) and two LED units (Curenos and G-Light Prima) were used to cure the adhesive resin of the two-step self-etch adhesives Clearfil SE Bond, Tokuso Mac Bond II, and FL-Bond II. A 0.6-mm thick acrylic mould was filled with adhesive resin and cured for 60 s. After 24-h water storage, specimens were trimmed into an hourglass shape with a width of 1.2 mm at the narrowest part, after which the μTS was determined (n=10). In addition, the light transmittance of each adhesive was characterized using a UV-vis-NIR spectrometer. No significant difference in curing efficiency between VLD and LED were observed for both Tokuso Mac Bond II and FL-Bond II (p>0.05). For Clearfil SE Bond, the μTS of VLD-cured specimens was higher than that of the specimens cured by the LED Curenos unit (p<0.05). Spectrometry revealed that this marked difference must be attributed to a different light transmittance of Clearfil SE Bond for visible blue light versus for the lower area of UV and visible violet light. In conclusion, A GaN-based violet laser diode can be used as light-curing source to initiate polymerization of dental resins.  相似文献

Long-term clinical aspects after kidney transplantation using cyclosporine (CsA) were studied in 33 patients who received kidney grafts from one haplotype identical living related donor and who were followed for at least one year. Both actual graft and patient survival rates were 97 per cent at one year. Incidence and severity of acute rejection were reduced to a greater extent in patients treated with CsA than in patients treated with azathioprine (AZ). The incidence of infections was low, and no serious bacterial infection occurred in these 33 patients. In 17 of 33 patients with a deteriorative graft function caused by intractable nephrotoxicity, CsA was converted to AZ. The mean serum creatinine level of converted patients was significantly higher than that of patients maintained with CsA at each time when a dose of CsA was stepwise reduced from 14 mg/kg/day to 6 mg/kg/day. Conversion to AZ improved graft function dramatically, although it resulted in reversible acute rejections in 4 patients. CsA induced hepatotoxicity occurred in 10 patients, but in all normal liver function was restored with decrease in the dose. The potent immunosuppressive effect of CsA compensates for its side effects. However, CsA should be converted to AZ when the chronic nephrotoxicity persists at a late stage of post-transplantation.  相似文献
