BACKGROUND: Previous studies have reported that the current DSM-IV eating disorder (ED) criteria do not adequately describe ED symptomatology. The objective of the current study was to examine the clustering of ED symptoms in a general population sample using latent class analysis (LCA). METHOD: ED symptoms from 3723 female young adult twins (mean age 22) were analyzed using LCA, and resulting classes were compared on external validators reflecting ED and other co-morbid psychiatric diagnoses, substance use disorders (SUDs), and suicidality. RESULTS: The optimal solution consisted of five latent classes characterized as: (1) Unaffected; (2) Low Weight Gain; (3) Weight Concerned; (4) Dieters; and (5) ED. Members of the ED class had significantly higher prevalence of co-morbid psychiatric disorders, SUDs, and suicidality than the Unaffected and Low Weight Gain classes, and elevated rates of suicidality and major depression compared to the Weight Concerned and Dieter classes, which differed from each other primarily in terms of current body mass index (BMI). Dieter class members were more likely to be overweight and obese and less likely to be underweight than Weight Concerned class members. The majority of women with an ED diagnosis were assigned to the ED class, and few differences were found between ED class members with and without an ED diagnosis. CONCLUSIONS: The results add to the evidence that many women with significant ED psychopathology are not being identified by the DSM-IV ED categories. 相似文献

We describe a hierarchical multiscale computational approach based on molecular dynamics simulations, free energy-based molecular
docking simulations, deterministic network-based kinetic modeling, and hybrid discrete/continuum stochastic dynamics protocols
to study the dimer-mediated receptor activation characteristics of the Erb family receptors, specifically the epidermal growth
factor receptor (EGFR). Through these modeling approaches, we are able to extend the prior modeling of EGF-mediated signal
transduction by considering specific EGFR tyrosine kinase (EGFRTK) docking interactions mediated by differential binding and
phosphorylation of different C-terminal peptide tyrosines on the RTK tail. By modeling signal flows through branching pathways
of the EGFRTK resolved on a molecular basis, we are able to transcribe the effects of molecular alterations in the receptor
(e.g., mutant forms of the receptor) to differing kinetic behavior and downstream signaling response. Our molecular dynamics
simulations show that the drug sensitizing mutation (L834R) of EGFR stabilizes the active conformation to make the system
constitutively active. Docking simulations show preferential characteristics (for wildtype vs. mutant receptors) in inhibitor
binding as well as preferential enhancement of phosphorylation of particular substrate tyrosines over others. We find that
in comparison to the wildtype system, the L834R mutant RTK preferentially binds the inhibitor erlotinib, as well as preferentially
phosphorylates the substrate tyrosine Y1068 but not Y1173. We predict that these molecular level changes result in preferential
activation of the Akt signaling pathway in comparison to the Erk signaling pathway for cells with normal EGFR expression.
For cells with EGFR over expression, the mutant over activates both Erk and Akt pathways, in comparison to wildtype. These
results are consistent with qualitative experimental measurements reported in the literature. We discuss these consequences
in light of how the network topology and signaling characteristics of altered (mutant) cell lines are shaped differently in
relationship to native cell lines.

We report a case of multiple, unifocal, subconjunctival cysticercosis in an 8-year-old boy. Two cysts, each with a scolex, were in the subconjunctival space. They were removed surgically, and histopathologically proven to be Cysticercus cellulosae. This is only the second report of twin cysticercus in the literature, to the best of our knowledge. 相似文献

Subdural empyema is a surgical emergency; if not recognized and managed early, it may prove fatal. In most of the cases, condition is preceded by paranasal sinusitis, otitis media or trauma. The authors report a previously undescribed case of spontaneous subdural empyema associated with Gaucher disease that had a good outcome following burr hole evacuation of subdural empyema and parenteral antibiotics. Patients with Gaucher disease may be vulnerable to severe bacterial infections, due to defective function of phagocytic cells and hypersplenism. 相似文献

Human immunodeficiency virus-1 (HIV-1) is the most frequent cause of dementia in adults under 40. We sought to use gene delivery to protect from HIV-1-related neuron loss. Because HIV-1 envelope (Env) gp120 elicits oxidant stress and apoptosis in cultured neurons, we established reproducible parameters of Env-mediated neurotoxicity in vivo, then tested neuroprotection using gene delivery of antioxidant enzymes. We injected 100-500 ng mul(-1)gp120 stereotaxically into rat caudate-putamens (CP) and assayed brains for apoptosis by terminal deoxynucleotidyl transferase-mediated biotinylated UTP nick end labeling (TUNEL) 6-h to 14-day post-injection. Peak apoptosis occurred 1 day after injection of 250 and 500 ng microl(-1)gp120. TUNEL-positive cells mostly expressed neuronal markers (NeuroTrace), although some expressed CD68 and so were most likely microglial cells. Finally, we compared neuroprotection from gp120-induced apoptosis provided by localized and generalized intra-central nervous system (CNS) gene delivery. Recombinant SV40 vectors carrying Cu/Zn superoxide dismutase (SOD1) or glutathione peroxidase (GPx1) were injected into the CP, where gp120 was administered 4-24 weeks later. Alternatively, we inoculated the vector into the lateral ventricle (LV), with or without prior intraperitoneal (i.p.) administration of mannitol. Intracerebral injection of SV(SOD1) or SV(GPx1) significantly protected neurons from gp120-induced apoptosis throughout the 24-week study. Intraventricular vector administration protected from gp120 neurotoxicity comparably, particularly if preceded by mannitol i.p. Thus, HIV-1 gp120 is neurotoxic in vivo, and intracerebral or intra-ventricular administration of rSV40 vectors carrying antioxidant enzymes is neuroprotective. These findings suggest the potential utility of both localized and widespread gene delivery in treating neuroAIDS and other CNS diseases characterized by excessive oxidative stress. 相似文献

The deadly disease swine flu is, without a speck of doubt, causing a massive havoc among the common people of India and has created fear across the various strata of the society. The objective was to find out the awareness, perception, and myths of school going children of class 9 th to 12 th toward swine flu. The present cross-sectional study was carried out in two randomly selected (using random number table) senior secondary schools of Bareilly among 400 students of class 9 th to 12 th . A total of 200 students were selected from each school. Chi-square test was applied for statistical analysis. Almost all the students (97.75%) have heard about of swine flu and are aware of it as a disease entity. Fever was found to be the main symptom while coughing and sneezing were main way of spread of swine flu known to them. About 97% of the students mention use of mask as most effective way to prevent them from swine flu. Knowledge of availability of medicine was present in less than half of the students. TV was found to be the main source from which they get knowledge (79%), and they are trying to get knowledge (53.2%) of swine flu.
