Six male Lewis rats were tested for the effect of 9-tetrahydrocannabinol (9-THC) on feeding evoked by electrical stimulation of the lateral hypothalamus. Treatment with 9-THC (0.4 mg/kg IP) decreased frequency threshold for feeding by 20.5% (±4.3), causing a leftward shift in the function relating stimulation frequency to the latency to begin eating 45-mg food pellets upon stimulation onset; there was no change in the asymptotic performance that was approached with sufficiently high stimulation frequencies. Naloxone (1 and 2 mg/kg) reduced the facilitory effect of 9-THC, but did so at doses that can inhibit feeding in the no-drug condition. These data are consistent with evidence implicating endogenous opioids in feeding, and suggest (but do not confirm) that the facilitation of feeding by 9-THC may be mediated by endogenous opioids. The facilitation of stimulation-induced feeding by doses of 9-THC that have been found to facilitate brain stimulation reward is consistent with evidence suggesting common elements in the brain mechanisms of these two behavioral effects of medial forebrain bundle stimulation. 相似文献

BackgroundAntidepressant drugs, like fluoxetine, a selective serotonin reuptake inhibitor, desipramine, a nonselective noradrenaline reuptake inhibitor, and mirtazapine, an antagonist of noradrenaline α2 auto- and heteroreceptors, are widely used for the treatment of depressive symptoms in cancer patients. Since these antidepressants have different activities targeting the immune system, they might also modulate tumor growth in cancer patients.MethodsIn the present study, we investigated the effects of administration of antidepressant drugs: fluoxetine, desipramine and mirtazapine on B16F10 melanoma tumor growth. These drugs were administered intraperitoneally (ip) for 17 days after subcutaneous injection of B16F10 melanoma cells to male C57BL/6J mice.ResultsFluoxetine significantly inhibited melanoma solid tumor growth and desipramine tended to decrease this parameter whereas mirtazapine had no effect.ConclusionThe inhibitory effect of fluoxetine on melanoma growth was associated with an increased mitogen-induced T cell proliferation which may at least partly participate in the mechanism of the antitumor effect of this antidepressant. It was found previously that in the rat, unilateral electrolytic lesion of the ventral tegmental area (VTA) facilitated feeding induced by electrical stimulation of the homologous VTA tissue in the contralateral hemisphere. In the present work, VTA stimulation-induced locomotor response was tested in male Wistar rats using a latency to move/stimulation frequency curve shift paradigm in order to check for functional generality of the "contralateral facilitation effect" and also with the aim of elaborating an easy and reliable behavioral model to study this phenomenon. In a further step, the hypothesis was tested that enhancement of function of the intact VTA results from elimination of tonic GABAergic influence derived normally from the lesioned VTA. GABA(A) (bicuculline, doses 0, 0.5 and 5.0 ng) and GABA(B) (phaclofen, doses 0, 500 and 1000 ng) receptors antagonists, and for comparison, a GABA(A) receptor agonist (muscimol, doses 0, 12.5, 25. 0 and 50.0 ng), were injected unilaterally to VTA and their effect on locomotor response elicited by electrical stimulation of the contralateral VTA was tested in a latency/frequency paradigm. It was found that similar to feeding, locomotor response evoked by unilateral electrical stimulation of the VTA was facilitated after contralateral VTA lesion which manifested as a decrease of the locomotion threshold and a leftward shift of the function relating latency to move to stimulation frequency. The effect was immediate, long-lasting and specific to the VTA destruction; lesions outside the VTA area caused gradual impairment of the locomotor response to stimulation. The facilitatory effect of the electrolytic lesion could be replicated by bicuculline, which significantly facilitated stimulation-induced behavior. Phaclofen exerted slight facilitating influence only at a low dose. No effect of muscimol on the locomotion threshold was found. We conclude that "the contralateral facilitation effect" at the level of VTA reflects the interhemispheric regulation of activity of the dopaminergic (DA) cells in which GABA(A)-mediated interhemispheric communication plays a significant role. 相似文献

The growing scientific interest in one-dimensional (1D) nanostructures based on metal-oxide semiconductors (MOS) resulted in the analysis of their structure, properties and fabrication methods being the subject of many research projects and publications all over the world, including in Poland. The application of the method of electrospinning with subsequent calcination for the production of these materials is currently very popular, which results from its simplicity and the possibility to control the properties of the obtained materials. The growing trend of industrial application of electrospun 1D MOS and the progress in modern technologies of nanomaterials properties investigations indicate the necessity to maintain the high level of research and development activities related to the structure and properties analysis of low-dimensional nanomaterials. Background: Overnutrition is one of the risk factors of chronic kidney disease (CKD). The factors related to both obesity and CKD are adiponectin and ghrelin. The aim of the study was to assess if there is a link of nutritional status and selected nutrients intake with adiponectin and ghrelin in patients with diabetic nephropathy (DN). Methods: The study involved 55 patients diagnosed with DN in the pre-dialysis period (two groups: GFR < 30 and >30 mL/min/1.73 m2). In all participants standard blood tests, total ghrelin and total adiponectin plasma concentrations and anthropometric measurements (BMI, WHR- waist–hip ratio, body composition analysis) were performed. The evaluation of energy and nutrient intakes was made using the three-day food record method. Results: Excessive body weight was found in 92.80% patients. The average daily energy intake was 1979.67 kcal/day (14.45% protein energy, 28.86% fat, and carbohydrates 56.89%). In the group with eGFR < 30 mL/min/1.73 m2 the analysis showed a negative relationship between ghrelin and WHR value, and the creatine and albumin concentrations. There was a positive correlation between ghrelin concentration and the consumption of carbohydrates and sucrose. In the group of patients with eGFR > 30 mL/min/1.73 m2, a positive correlation was found between the concentration of ghrelin and the consumption of vegetable protein, carbohydrates, and glucose. Conclusions: The study confirms the high prevalence of obesity in patients with DN-Excessive supply of protein was found in the patients’ diets, which may contribute to the deterioration of the course of the disease and its prognosis. In patients with eGFR < 30 there was a negative correlation between ghrelin concentration and nutritional status, and in patents with eGFR > 30 between ghrelin concentration and some nutrients intake. 相似文献

The paper presents the optimisation of a safe diode laser irradiation process applied to the surface of titanium implants in order to reduce microbial numbers in the treatment of inflammation classified as periimplantitis. The study comprised isolation and identification of microorganisms inhabiting surfaces of dental implants, crowns, teeth and saliva from patients with fully symptomatic periimplantitis. Microorganisms were detected by a culture-dependent method and identified with the use of MALDI-TOF mass spectrometry. The isolated microorganisms were inoculated on the surface of a new implant and then irradiated by a diode laser (wavelength of 810 ± 10 nm) in one, two or three repetitions and biocidal efficacy was assessed. To evaluate impact of laser irradiation on roughness, morphology and structure of the implant surface, optical profilometry, scanning electron microscopy and optical microscopy were used. Examination of the tested surfaces and saliva revealed the presence of Gram-positive and Gram-negative bacteria and one fungal species. In all patients, cultures from the endosseous part of the implant revealed the presence of the pathogenic and pyogenic bacterium Streptococcus constellatus. In 13 out of 20 samples laser-irradiated in duplicate and triplicate, all microorganisms were eliminated. The irradiation used did not cause any changes in the properties of the implant surface. 相似文献

The materials based on concrete with an addition of rubber are well-known. The interaction between concrete components and rubber particles is in the majority cases insufficient. For this reason, different substances are introduced into concrete-rubber systems. The aim of this paper is to establish the influence of five different polymer additives, i.e., 1. an aqueous dispersion of a styrene-acrylic ester copolymer (silanised) (ASS), 2. water dispersion of styrene-acrylic copolymer (AS), 3. anionic copolymer of acrylic acid ester and styrene in the form of powder (AS.RDP), 4. water polymer dispersion produced from the vinyl acetate and ethylene monomers (EVA), 5. copolymer powder of vinyl acetate and ethylene (EVA.RDP)) on the properties of the self-leveling rubberised concrete. Scanning electron microscopy has allowed to establish the interaction between the cement paste and rubber aggregates. Moreover, the compressive strength and flexural strength of the studied materials were evaluated. The results indicate that the mechanical properties depend extensively on the type as well as the amount of the polymer additive introduced into the system. 相似文献

Non-alcoholic fatty liver disease (NAFLD) is an increasingly common condition associated with type 2 diabetes (T2DM) and cardiovascular disease (CVD). Since systemic metabolic dysfunction underlies NAFLD, the current nomenclature has been revised, and the term metabolic-associated fatty liver disease (MAFLD) has been proposed. The new definition emphasizes the bidirectional relationships and increases awareness in looking for fatty liver disease among patients with T2DM and CVD or its risk factors, as well as looking for these diseases among patients with NAFLD. The most recommended treatment method of NAFLD is lifestyle changes, including dietary fructose limitation, although other treatment methods of NAFLD have recently emerged and are being studied. Given the focus on the liver–gut axis targeting, bacteria may also be a future aim of NAFLD treatment given the microbiome signatures discriminating healthy individuals from those with NAFLD. In this review article, we will provide an overview of the associations of fructose consumption, gut microbiota, diabetes, and CVD in patients with NAFLD. 相似文献
