目的 探究风湿性心脏病病人干扰素调节因子4(IRF4)去泛素化与去泛素化酶USP4互相作用的分子机制,为风湿性心脏病提供新的治疗策略。方法 来自安徽医科大学第一附属医院2015年7月至2016年1月20例风湿性心脏病病人,均行体外循环下瓣膜置换手术,均顺利出院。10例健康对照来自于健康志愿者。分别抽取以上参与实验人员5 mL的外周血,经过细胞培养与转染;通过免疫共沉淀实验观察IRF4的DNA结合结构域以及和配体结合结构域是否可以和USP4互相结合;通过NI-NTA镍螯合树脂纯化实验观察IRF4蛋白针对48位和63位赖氨酸相关的去泛素化是否可以被USP4介导;应用荧光素酶检测技术观察白细胞介素(IL)-4是否可以在IRF4、活化T细胞核因子(NFATC2)和USP4共同促进下表达;利用基因沉默方法在人源Th2细胞中下调USP4。结果 下调USP4后Th2细胞相关细胞因子的转录水平也明显减少;利用流式细胞术检测经USP4抑制剂处理后的Th2细胞表达的IRF4和IL-4均减少;以流式细胞技术测得风湿性心脏病中IL-4表达增高,IRF4表达也增高。结论 风湿性心脏病病人血中IRF4的蛋白稳定表达可以通过USP4去泛素化的作用而实现;IL-4在IRF4表达增加协同NFATC2的情况下可增加表达。 相似文献

目的 探讨基于单元的综合安全项目在复发性卵巢癌化疗患者安全用药中的应用效果。方法 选取安徽省某三级甲等医院2个妇科病区,实施基于单元的综合安全项目指导下的复发性卵巢癌化疗药物安全用药管理,实施前后使用医院安全文化调查问卷、安全用药知信行水平调查问卷、复发性卵巢癌化疗药物理论知识考核和复发性卵巢癌化疗患者药物不良反应发生率进行效果评价。结果 实施后医院安全文化调查问卷中待改进区域的积极反应率较干预前有所改善(P<0.05);实施后医护人员的安全用药知识、态度和行为水平以及理论知识考核得分均高于实施前(P<0.05);实施后复发性卵巢癌化疗患者药物不良反应的发生率较实施前明显下降(P<0.05)。结论 基于单元的综合安全项目指导下的复发性卵巢癌化疗患者安全用药管理,有利于建立病区安全文化,提高医护人员安全用药水平,减少化疗药物不良反应的发生率。 相似文献

The aims of this study were to evaluate the feasibility of integrating archival datasets from depression projects involving pregnant women recruited from obstetric clinics and then assess the representativeness of the integrated dataset. Datasets from six studies were standardized and integrated. Chi-square, t-, and Wilcoxon rank-sum tests were used to compare characteristics between women who completed a depression screening questionnaire (DSQ) and were (1) eligible and ineligible for research participation and (2) eligible women who accepted and declined participation. The integrated dataset comprises 9,112 pregnant women, of whom 71.0 % (n?=?6,472) were ineligible for participation because their DSQ scores indicated no-to-minimal depressive symptoms (NDS). Among the 23.9 % (2,176) of women identified as eligible, in part, because their DSQ scores indicated elevated levels of depressive symptoms (EDS), 29.6 % (644) of women participated (P-EDS) and 47.6 % (1,036) of women did not participate (D-EDS). While the NDS and EDS groups were significantly different on almost all variables, the P-EDS and D-EDS groups were significantly different on only a few variables. Compared to the D-EDS group, the P-EDS group was earlier in pregnancy and, on the Edinburgh Postnatal Depression Screen, was more likely to endorse impaired “ability to laugh” and “enjoy oneself”, and endorse at greater severity “ability to laugh.” It is a reasonable and feasible strategy to integrate thematically similar datasets to increase statistical power. Additionally, typical recruitment strategies for minimal risk perinatal depression research at obstetric clinics, during routine prenatal care visits, appear to produce an externally valid study cohort. 相似文献

Hybrid SERS configurations, which combine manufactured metallic chips with nanoparticles, have emerged as powerful and promising SERS substrates because they not only provide cost-effective and high-yield manufacture, but also demonstrate excellent sensitivity and outstanding reproducibility. Herein, a plasmonic hybrid structure, a particle on an Au film over nanoparticles (particle-AuFON) configuration, was studied for SERS application. In a previous study, we constructed a hybrid substrate by grafting Au@Ag core–shell NPs onto the AuFON structure. In this study, the hybrid substrate is designed and simulated to optimize electromagnetic enhancement while also affording exceptional uniformity, repeatability and stability, which are essential factors in SERS applications. This hybrid substrate provides good SERS performance with a detection limit of 1 × 10−10 M, which is 100-fold improvement compared to AuFON substrate or Au@Ag NPs. The excellent signal enhancement originates from the hotspot improvement and densification, as visualized by the FDTD calculations. Additional hotspots were created at the gaps between the Au@Ag NPs and the AuFON, thus improving the density of hotspots. Moreover, the intensity of the hotspots was improved due to EM coupling between the original hotspots and additional hotspots. To validate the feasibility of this hybrid substrate in SERS-based detection, melamine was detected as an example. The detection limit was 10 nM, which was much lower than the maximum limit of melamine in infant formula (1 ppm) legislated by the governments of both the United States and China. A calibration curve was plotted between the SERS intensity and melamine concentration with a correlation coefficient of 0.98. This hybrid SERS substrate shows great potential in SERS-based sensing and imaging, as it provides high sensitivity and outstanding reproducibility with a simple fabrication procedure, facilitating the cost-effective and high-yield manufacture of SERS substrates.A plasmonic hybrid structure of particles on a Au film over nanoparticles (particle-AuFON) configuration was studied for application in SERS. To determine whether Hyperbaric oxygen preconditioning (HBO-PC) promotes neovascularization by increasing Stromal cell derived factor-1 (SDF-1) and CXC chemokine receptor 4 (CXCR4) in transplanted skin flaps of rats. The epigastric pedicle skin flap was established in a rat model. Rats were randomly assigned to the following five groups: 1) sham-operated group (SH); 2) ischemia followed by reperfusion 3 days postoperatively group (IR3d); 3) ischemia followed by reperfusion 5 days postoperatively group (IR5d); 4) hyperbaric oxygen preconditioning and ischemia followed by reperfusion 3 days postoperatively group (HBO-PC3d); and 5) hyperbaric oxygen preconditioning and ischemia followed by reperfusion 5 days postoperatively group(HBO-PC5d). For the groups receiving HBO-PC, animals underwent 1 hour of HBO at 2.0 ATA in 100% O2 twice per day for 3 days consecutively prior to surgery. After perfusion, Laser Doppler perfusion imaging (LDPI) was performed, and skin flap tissue samples were harvested for histological evaluation and western blot analysis. Perfusion was significantly improved in the HBO-PC groups compared with the IR groups on postoperative 3 and 5. Microvessel density (MVD) was significantly increased by HBO-PC compared with IR groups postoperatively. Western blot analysis revealed that SDF-1 and CXCR4 expression in the HBO-PC groups was significantly increased compared with IR groups. HBO-PC promoted neovascularization via increasing expression levels of SDF-1 and CXCR4 in transplanted skin flaps of rats. 相似文献

Conductive polymers, such as polyaniline (PANI), polypyrrole (PPy), polythiophene (PTh) and poly 3,4-ethylenedioxythiophene (PEDOT), play an important role in the application of pseudocapacitors. It is necessary to explore the effects of different conductive polymers in electrode composites. Herein, we prepare zinc sulfide/reduced graphene oxide (ZnS/RGO) by the hydrothermal method, and conductive polymers (PANI, PPy, PTh and PEDOT) doped with the same mass ratio (polymer to 70 wt%) via in situ polymerization on the surface of ZnS/RGO composite. For the supercapacitor application, the ZnS/RGO/PANI ternary electrode composite possesses the best capacitance performance and cycle stability out of all of the polymer-coated ZnS/RGO composites. In the three-electrode system, the discharge specific capacitance and cycle stability of ZnS/RGO/PANI are 1045.3 F g−1 and 160% at 1 A g−1 after 1000 loops. In a two-electrode symmetric system, the discharge specific capacitance and cycle stability of ZnS/RGO/PANI are 722.0 F g−1 and 76.1% at 1 A g−1 after 1000 loops, and the greatest energy and power density of the ZnS/RGO/PANI electrode are 349.7 W h kg−1 and 18.0 kW kg−1. In addition, conductive polymers can effectively improve the voltage range of the electrode composites in 6 M KOH electrolyte for the two-electrode system.
