Bax is an oncogene that has proapoptotic properties but not all cells that express Bax undergo apoptosis. Bax may have a function unrelated to apoptosis. To elucidate the role of Bax in cell signaling, an epithelial cell line called SMG-C6 was transfected with the human bax gene. Stable transfectants were studied for their response to carbachol, a muscarinic receptor agonist, by measuring the increase in intracellular free Ca(2+) and Ca(2+) influx. Carbachol-mediated release of Ca(2+) from intracellular stores was significantly higher in Bax transfectants compared to control transfectants (empty vector). Ca(2+) influx was also increased in Bax transfectants. Bax had no affect on the storage operated channels. However, the concentration of Ca(2+) in the intracellular stores (i.e., mitochondria and granules) was 40-50% lower in the Bax transfectants. There was no significant difference in thapsigargin-mediated apoptosis in Bax transfectants compared to wild-type and control transfectants. Measurement of glutathione was reduced in the Bax transfectant. Restoration of glutathione levels with glutathione monoethyl ester partially normalized Ca(2+) mobilization and storage capacity in the mitochondria to control levels. This study shows that sub-apoptotic levels of Bax can reduce Ca(2+) content in intracellular stores and Ca(2+) homeostasis. Bax may mediate these effects by reducing the levels of antioxidants resulting in mild oxidative stress.  相似文献

The goal for external ear reconstruction is to restore the function of the helical rim in order to provide support for eyeglasses and to provide a cosmetically acceptable auricle. Reconstruction necessitates both flap coverage and flexible framework formation. This article reviews 22 patients who underwent subtotal reconstruction of the auricles after burns. The study was conducted from 1996 to 2003. The study group consisted of 14 male and 8 female patients aged 6-43 years (mean 21.9 years). Different modalities have been used: the temporoparietal fascial flap (eight patients), the subcutaneous pocket technique (11 patients), the pre-auricular skin flap (four patients), and the post-auricular skin flap (three patients). The flaps were used for coverage of fabricated autogenous cartilage in all patients. Both aesthetic and functional outcome was satisfactory.  相似文献

Several new platinum(II) complexes of the general formulae cis-[PtCl(2)(DMSO)L], where L is a Schiff base or hydrazone derived from tetrazolo[1,5-a]quinoline-4-carboxaldehyde as carrier ligands, have been synthesized and characterized physicochemically and spectroscopically. These platinum complexes which are structurally analogues to what so called cisplatin [cis-[PtCl2(NH3)2]; the first generation anticancer agent] were evaluated for their cytotoxicity on HL-60 (human promyelocytic leukemia) cells. Two of the platinum complexes were almost similar in their activity to cisplatin, while the remaining three complexes have demonstrated higher efficacy than that of cisplatin. Based on our findings, these novel platinum complexes appear to be valuable leading compounds with high efficacy.  相似文献
