BACKGROUND: The effect of glucose on local anesthetic-induced neural damage has not been fully studied. We examined the effect of glucose on hemolysis induced by local anesthetics. METHODS: The mean EC50 values (the local anesthetic level that causes destruction of half of the red blood cells in vitro) of lidocaine HCl, tetracaine HCl and dibucaine HCl were determined with 0% and 7.5% glucose contained in Krebs solution at pH 6.4. RESULTS: The mean EC50 values of lidocaine HCl, tetracaine HCl, and dibucaine HCl in 0%-glucose Krebs solution were 6.51%, 0.45%, 0.17%, respectively, which increased significantly to 7.05%, 0.64% and 0.23%, in 7.5% glucose Krebs solution at pH 6.4. CONCLUSIONS: Glucose may have a protective role in local anesthetic-induced neural damage.  相似文献

BACKGROUND: Only a few studies on the mortality of Japanese immigrants have been conducted in Brazil despite a large population of Japanese immigrants and their different environment and lifestyle from Japanese living in Japan. METHODS: To compare cancer mortality between Japanese in Japan and Japanese immigrants or Brazilians in the state of S?o Paulo, Brazil, we obtained official death certificates registered during 1999-2001. The standardized mortality ratio (SMR) or the standardized proportional mortality ratio (SPMR) of major cancer sites was calculated for the first generation of Japanese immigrants to Brazil (Japan-born), their Brazil-born Japanese descendants, and native Brazilians using mortality data of Japanese in Japan as a standard. RESULTS: The SMRs of stomach and colorectal cancer did not differ between the Japan-born residents of Brazil and the native Japanese, but significantly low SMRs were found among the native Brazilians. Compared with the native Japanese, we observed significantly lower SMRs for liver, gallbladder and lung cancer and significantly higher SMRs for prostate, cervical, and brain and nervous system cancer among both the Japan-born residents of Brazil and the Brazilians. Generally, the SPMR results were similar to those of the SMRs. Significantly high SPMRs for breast and uterine cancer were found for both the Japan- and Brazil-born residents of Brazil, although the Japan-born residents had increased SMRs, but not significantly so. CONCLUSIONS: We confirmed the different cancer mortality pattern in the Japanese immigrants from that in Japanese in Japan, thus demonstrating the relative importance of the environment in the development of cancer.  相似文献

Although long-chain n-3 polyunsaturated fatty acids (Ln-3 PUFA), which are abundant in fish, have shown protective effects on colorectal cancer in laboratory studies, epidemiological studies to date have not been consistent. We evaluated the relationship of consumption of fish and Ln-3 PUFA to the colon and rectal cancer risk in the two cohorts of the Japan Public Health Center-based prospective study of 42,525 men and 46,133 women. Dietary and other exposure data were obtained between 1990 and 1994. Through December 1999, 705 cases of colon and rectal cancer were documented. When data from the two cohorts were pooled, multivariable relative risks (RRs) for the highest quartile compared with the lowest quartile of fish consumption were 1.07 (95% confidence interval, CI = 0.77-1.48) for colon cancer and 0.95 (95% CI = 0.63-1.43) for rectal cancer with no dose-risk trend. RRs for the highest quartile compared with the lowest quartile of eicosapentaenoic acid consumption were 1.05 (95% CI = 0.76-1.46) for colon cancer and 0.91 (95% CI = 0.60-1.38) for rectal cancer with no dose-risk trend. This study does not support the role of fish and Ln-3 PUFA in the etiology of colon and rectal cancer in this population whose fish consumption was high and the variation in Ln-3 PUFA consumption was large.  相似文献

Adenovirus-mediated wild-type p53 gene transfer induces apoptosis in a variety of human cancer cells. Although clinical trials have demonstrated that a replication-deficient recombinant adenovirus expressing the wild-type p53 gene (Ad-p53) is effective in suppressing growth of non-small cell lung cancer (NSCLC), we often experienced late resistance to this treatment. To elucidate the mechanism of late resistance to Ad-p53 in human lung cancer cells, we generated 5 different resistant variants from p53-susceptible H1299 NSCLC cells by repeated infections with Ad-p53. We first examined the transduction efficiency of adenoviral vector by Ad-LacZ transduction followed by X-gal staining in parental and 5 resistant H1299 cell lines. Their sensitivity to viral infection decreased in correlation with the magnitude of resistance, and Ad-p53-mediated tumor suppression could be restored by dose escalation of Ad-p53 in the resistant variants. The expression of Coxsackie and adenovirus receptor (CAR) and alphaV integrins, which are cellular receptors for attachment and internalization of the virus, respectively, was next investigated in these cell lines. Flow cytometry revealed that alphaVbeta3 and alphaVbeta5 integrin expression was consistent, while p53-resistant cell lines showed that diminished CAR expression correlated with the magnitude of the resistance. Our results demonstrated that decreased CAR expression could be one of the mechanisms of late resistance to Ad-p53, which may have a significant impact on the outcome of adenovirus-based cancer gene therapy.  相似文献

BACKGROUND: The associations between supplement use and certain demographics, lifestyles, health characteristics, and dietary intakes have not been studied in a large population in non-Western societies. The objective of our study was to investigate the association between supplement use and demographics, lifestyles, health characteristics, and dietary intake in a population-based cohort study in Japan. METHODS: Subjects were the 78 531 participants (45-74 years) who completed a self-administered questionnaire in 1995 or 1998 in a 5-year follow-up survey by the Japan Public Health Center-based prospective Study on cancer and cardiovascular disease. The questionnaire included enquiries about supplement use, occupation, height, weight, smoking, alcohol, physical activity, dietary behaviours, working hours, subjective stress, as well as intakes for 138 foods. RESULTS: The supplement users were likely to have formerly smoked or never smoked. Female supplement users were likely to consume alcohol moderately. The prevalence of users was higher in the elderly, the self-employed, those with lower body mass index, greater physical activity, lower frequency of eating prepared food, higher frequency of eating out, and higher stress level in both sexes after mutual adjustment. Mean intakes of energy and nutrients were lower for users than for non-users. CONCLUSION: The demographics, lifestyles, health characteristics, and dietary intakes may need to be adjusted when evaluating the effect of dietary supplements on disease because they can become potential confounding factors.  相似文献

Residual 152Eu activities induced by neutrons from the Nagasaki atomic bomb were measured for nine mineral samples located up to 1,061 m in the slant range and one control sample at 2,850 m from the hypocenter. A chemical separation to prepare europium-enriched samples was performed for all samples, and gamma ray measurements were carried out with a low background well-type germanium detector. In this paper, the measured specific activities of 152Eu are compared with activation calculations based on the DS86 neutron fluence and the 93Rev one. The calculated-to-measured ratios are also compared with those of 60Co and 36Cl. The present results indicate that the measurements agree to the calculation within a factor of three as observed in the nuclear tests at Nevada. The activation level of environmental neutrons and the detection limit for 152Eu are also discussed.  相似文献

OBJECTIVE: This study was aimed at evaluating the effect of long-term vitamin C supplementation on serum and dietary vitamin C and identifying the factors associated with change in serum concentration. METHODS: A total of 439 subjects with atrophic gastritis initially participated in a randomized clinical trial using vitamin C and beta-carotene to prevent gastric cancer. We originally randomized the participants into four treatment groups using a 2x2 factorial design, whereby 0 or 15 mg/day beta-carotene and 50 or 500 mg/day vitamin C were administered in a double-blind manner. The beta-carotene component was terminated early after a mean treatment duration of four months. Before and upon early termination of beta-carotene supplementation, 134 subjects dropped out this trial, while 120 and 124 subjects took the vitamin C supplement at either 50 mg or 500 mg daily for five years. RESULTS: Changes in serum vitamin C were significantly higher in the high-dose group (38.5% increase, 95% CI = 27.0-49.9) than in the low-dose group (13.0% increase, 5.1-20.9) or in the dropout group (3.3% increase, -2.1-8.6) after five-year supplementation. The serum vitamin C at baseline was negatively associated with changes in serum vitamin C (p < 0.0001), while high-dose (p < 0.0001) and low-dose (p < 0.05) supplementation and female gender (p < 0.001) were positively associated. Dietary intake of vitamin C in the supplementation group was almost identical before and after five-year supplementation of vitamin C (2.31 mg/day decrease, 95% CI = -15.3-10.7), while a 17.7 mg/day decrease (95% CI = -44.2-8.86) was observed in the drop-out group. CONCLUSION: Five-year vitamin C supplementation induces a remarkable increase in serum vitamin C concentration, and our intervention program appears to have no effect on dietary vitamin C intake.  相似文献

We assessed the possible association of gastrointestinal cancers with cruciferous vegetables and mushrooms in a multicenter, hospital-based case-control study in an agricultural area of Japan. One hundred forty-nine cases and 287 controls for stomach cancer and 115 cases and 230 controls for colorectal cancer were matched by age, sex, and residential area. In stomach cancer, the protective effect of vegetables (consumption of total vegetable) was obscure, but it became clearer when we examined specific kinds of vegetables. Marginal associations were observed in the group of the highest consumption of Chinese cabbage (odds ratio [OR] = 0.61; 95% confidence interval [CI] = 0.35-1.07), broccoli (OR = 0.60; 95% CI = 0.34-1.08), Hypsizigus marmoreus (Bunashimeji) (OR = 0.57; 95% CI = 0.31-1.04) and Pholita nameko (Nameko) (OR = 0.56; 95% CI = 0.30-1.06). In colorectal cancer, we observed decreased risks from the highest tertile of total vegetables (OR = 0.22; 95% CI = 0.08-0.66) and low-carotene-containing vegetables (OR = 0.28; 95% CI = 0.08-0.77), and inverse associations were observed in the group of the highest consumption of broccoli (OR = 0.18; 95% CI = 0.06-0.58). Although the sample size was limited, subgroup analyses showed that the associations differed with the histopathological subtype. These findings suggest that cruciferous vegetables decrease the risk of both stomach and colorectal cancer, and that mushrooms are associated with a decreased risk of stomach cancer.  相似文献

BACKGROUND: Although epidemiologic studies including the Japan Public Health Center-based Prospective Study on Cancer and Cardiovascular Disease (JPHC study) have frequently used the basic health examination participants as study subjects, their background characteristics have rarely been investigated. The aim of this study is to clarify the background characteristics of participants and to discuss their impact on epidemiologic studies. METHODS: Subjects were 43,140 (Cohort I) and 34,892 (Cohort II) respondents aged 40-59 years who completed a self-administered questionnaire in 1990 or 1993-94 by the JPHC Study. Respondents whose data of the basic health examination were also available were defined as participants. We compared their sociodemographic factors, personal medical history, and lifestyle-related factors with those of non-participants. RESULTS: Participants tended to be older and less educated. They were more likely to engage in agriculture, forestry and fisheries or to be self-employed persons, or homemakers. Male participants smoked less and were more likely to drink alcohol beverage moderately. Female participants smoked and drank less but tended to participate more in sports and physical exercise in their leisure time. Both male and female participants tended to eat fruits and green vegetables more often than non-participants. In short, participants had a different socioeconomic status from non-participants and a favorable lifestyle profile, especially among women. These findings were principally consistent between the two cohorts. CONCLUSION: These differences between participants and non-participants in the basic health examination might cause a selection bias that limits the application of the results to only participants in the basic health examination.  相似文献
